In this study, the transforming growth factor-β receptor I (TGFβRI) kinase inhibitor SD-208 and a toll-like receptor (TLR)-7/8 agonist resiquimod (R848) were incorporated into serum-derived exosomes (EXOs). SD-208-encapsulated EXOs (SD-208/EXOs) and R848-encapsulated EXOs (R848/EXOs) were successfully prepared by sonication and electroporation, respectively. Also, SD-208/EXO and R848/EXO are stable at neutral pH condition with size below 150 nm. However, these particles were destabilized at acidic pH, similar to intracellular endosomal environment. In consideration of enhanced permeability and retention effect, SD-208/EXO and R848/EXO could be used for cancer immunotherapy. A novel polyhistidine (pHis)-incorporated lipid nanoparticle (pHis/LNP) was developed for the delivery of therapeutic siRNAs using a microfluidic device. I found incorporated siRNA into pHis/LNP, having spherical morphology and homogeneous size. LNP successfully protected siRNAs from degradation in serum condition. After treatment with fluorescence-labeled LNPs, dotted fluorescent signals were observed in cells with LNP, whereas strong and diffused fluorescence intensity was observed in cells with pHis/LNPs. The extent of gene silencing by siRNA pHis/LNP was greatly improved compared to that by siRNA/LNP, probably due to the high rate of endosome escape by pHis. Taken together, these particles can be utilized for the delivery of a wide range of therapeutic siRNAs including cancer therapy.
