As the number of small molecule compounds is large, various combinations can be applied and the experimental method is simple. Nevertheless, it is reproducible only in the same research team that developed the chemical reprogramming technique for the first time, so active research is not being conducted. In this dissertation, an experiment was conducted with the goal of establishing a more detailed experimental method to solve these problems and applying it to human cells to lay the foundation for making chemically induced pluripotent stem cells (CiPSCs). Chemical reprogramming and the stage of extraembryonic endoderm (XEN), which is only passed through in chemical reprogramming, were described in Chapter 1 along with a variety of reprogramming strategies and their limitations. Chapter 2 describes how to generate CiPSCs in mouse cells. In the process of inducing XEN cells in mouse embryonic fibroblasts using a small molecule compounds cocktail, it was confirmed that other types of cells were also induced along with XEN cells. It was impossible to induce pluripotency in a heterogeneous condition of different cell types, including XEN cells. Therefore, it was revealed that the homogeneity of XEN cells is the key to improving the efficiency of CiPSC induction and chemical reprogramming. I attempted to generate a human XEN cell line in Chapter 3 under the assumption that human cells would require the XEN stage to induce CiPSCs, as with mouse cells. I tried to induce XEN cells by approaching them in various ways, including chemical methods, and human XEN cells were induced in the OCT4-EGFP/NANOG-tdTomato reporting system iPSCs that can screen small molecular compounds required for induction to CiPSCs or increase their efficiency. The method for increasing the homogeneity of XEN cells, which is an intermediate stage for chemical reprogramming, is expected to be a breakthrough for researchers who have not been able to reproduce chemical reprogramming.
