Peptidomics is a rapidly growing field that systematically identifies and characterizes bioactive peptides (BPs). This study aimed to produce novel BPs from the domestic soybean species "Aritaunkong" using microbial hydrolysis and enzyme reaction. In silico digestion showed that microbial proteases yield different BPs than those from human digestive enzymes. We compared the sequence variations between wild and reference soybeans (Glycine_max_v4.0, RefSeq GCF_000004515.6), revealing that “Aritaunkong” has 25,126 SNPs and 4800 INDELs. We used Chryseobacterium, Lactiplantibacillus, and Alcalase 2.4 as biocatalysts, growing them in defined media supplemented with Aritaunkong for 72 h and performing 6-hour digestion with Alcalase. Consequently, we obtained soybean hydrolysates and fractionated them through ultrafiltration to yield 0~1 kDa and 1~10 kDa fractions, and analyzed the peptide distributions by size exclusion chromatography. HR-LC-Orbitrap-mass spectrometry (MS) identified 1049 novel soybean peptide sequences, suggesting that microbial hydrolysis and enzyme reaction can provide differential BPs in strain- and enzyme-specfic manners. Moreover, GPCR β-arrestin result demonstrated that these soybean peptides can bind to several specific GPCRs in humans, indicating potential health benefits. These findings suggest that peptidomics-based identification of soybean-derived BPs may have implications for the development of functional foods and nutraceuticals with health-promoting properties.
