Parkinson's disease (PD) is a neurodegenerative disease caused by the gradual destruction of dopaminergic neurons in the part of the brain called the substantia nigra. Although the prevalence of Parkinson's disease is increasing all around the world, the current Parkinson's disease treatment system is only focusing on slowing the progress of the disease or improving symptoms, so there is a necessity and demand for a system that can solve the underlying cause of Parkinson's disease. Recently, many researches have shown that intestinal microbiome is related to the neurodegenerative disease including Parkinson's disease through the gut-brain axis and short-chain fatty acids (SCFAs). In this study, the neuroprotective effect of probiotics was investigated using a soil nematode model, Caenorhabditis elegans, regarding the connection of intestine-nervous system. Analysis of short-chain fatty acid production of entire 11 probiotics strains, and body movement measurement, basal slowing response assay, dopaminergic neurons fluorescence analysis, α-synuclein aggregation observation, gene expression in metabolic pathway of dopamine in probiotics-treated C. elegans were observed. diet with the selected three LAB strains efficiently prevented Parkinson's disease by improving neurodegenerative locomotive behaviors and dopaminergic gene expression in PD-induced C. elegans model. These results prove an anti-Parkinsonism effect of the probiotic strains through the SCFAs-related gut-brain connected mechanism and the potential to be used in the area of functional food and pharmaceutical industries.
