In this study, dopamine and genipin were employed as a colorant to dye gray hairs. Whether the dopamine and genipin dye can reduce dyeing time, improve durability, and elevate biocompatibility was investigated. Dopamine is a neurotransmitter found in the central nervous system. Polydopamine polymerized from dopamine has the potential to readily stain solid surfaces dark-brown color. However, staining of solid surfaces using polydopamine requires a long time and has difficulties to apply as a hair dye. To overcome these drawbacks, genipin was employed in the dopamine-based hair dyeing system because it can efficiently induce crosslinking between polydopamine and hair.
Hair dyeing was conducted by using the aqueous solution containing dopamine hydrochloride and genipin. Briefly, dopamine hydrochloride (40 mg) was dissolved in 5 mL of 50 mM Tris-HCl buffer (pH 5.5, 6.5, 7.5, 8.5, and 9.5) and then genipin (20 mg) with the desired concentration was added into the dopamine solution. Then, 0.2 g of gray hair (5 cm) was soaked in the dopamine-genipin solution, and then dyeing processes were performed at room temperature. After a desired time, the dyed hair samples were washed with ultrapure water from three times and dried in air.
The gray hairs dyed with dopamine/genipin-based hair dye (DGHD) as a novel hair colorant was characterized by usingcolorimeter, scanning electron microscope (SEM), ultraviolet-visible (UV-Vis) spectrophotometer, and attenuated total reflection (ATR)-Fourier transform infrared (FTIR) spectrometer. Furthermore, the MTT assay was employed to assess in vitro cell cytotoxicity of DGHD against human epithelial keratinocyte cell line (HaCaT).
According to the increase of pH from 5.5 to 9.5, the colors of hairs by DGHD were changed from green to black. Furthermore, an L* value in CIEL*a*b* color coordinates, which means brightness, tended to gradually decrease with change of pH from 5.5 to 8.0. The lowering L* value of the dyed hair reached an equilibrium at pH 8.5. The L* value of the dyed hair at pH 8.5 was 15.54±0.82. When DGHD was used for dyeing of gray hairs, L* value showed 2.52 or 2.39 time higher than dopamine only or genipin only was used for the dyeing. According to the increase of genipin content in DGHD, the colors of gray hairs were changed from light green to dark green or black. Furthermore, the colors of gray hairs were changed from blue green to black when dopamine content increased in DGHD. When 8 mg/mL of dopamine and 4 mg/mL of genipin was added in the solution at pH 8.5 (Tris-HCl buffer) to dye gray hairs, the color of the hairs rapidly changed to black color than those cases of other pH conditions. The hair surface was covered with the pigment of DGHD. The CIE L*a*b* values of the gray hairs dyed with DGHD were significantly lowered compared to the use of dopamine only or genipin only. The a * value of the gray hairs dyed with DGHD increased according to increase of the amounts of dopamine. Furthermore, the gray hairs dyed with DGHD showed negative b * values. The color of the gray hairs dyed with DGHD was almost unchanged despite washing more than 30 times with a commercial shampoo. DGHD did not show cytotoxic in HaCaT cells up to 100 μg/mL of concentration. In conclusion, it can be suggested that DGHD is could be as a new hair dye that can replace existing products because it can dye gray hair under mild conditions without the aid of other chemical dyes, metallic mordants, or strong alkaline agents.
