SIMPER 분석 결과 하구는 해역별, 하구유형별 군집의 차이를 보였으며, 한반도 하구의 대표어종은 숭어, 붕어, 민물검정망둑으로 확인되었다. 서해역은 1차담수어인 살치와 붕어, 남해역은 기수성어종인 숭어와 가숭어, 동해역은 회유성어종인 황어와 기수성어종인 숭어로 확인되었으며, 열린하구에서는 숭어, 민물검정망둑, 닫힌하구에서는 붕어, 숭어로 확인되었다. 물리·화학적 특성에서 동해역은 열린하구의 특성을 보였으며, 서해역은 닫힌하구의 특성을 보였다. 닫힌하구로 대표되는 서해역은 탁도, COD, TN, TP 등 오염항목으로 대표되는 항목이 열린하구와 통계적 차이를 보였으며, 어류상에 큰 영향을 끼치는 것으로 나타났다. 반대로 열린하구로 대표되는 동해역은 DO, 전기전도도, 염도가 높으며, 서해역과 반대되는 특성을 보였다. CCA 결과에서도 해역별, 하구유형별 차이는 뚜렷하게 나타났으며, 어류군집 또한 SIMPER 결과와 합치되는 결과가 도출되었다. 한반도 하구의 KEFAI 평균은 50.52(C등급)으로 나타났으며, 해역별로 서해역 41.84점(C등급), 남해역 52.71점(C등급), 동해역 55.48점(C등급)으로, 서해역에서 동해역으로 갈수록 KEFAI가 높게 나타났다. 서해역에서 동해역으로 갈수록 열린하구의 비율이 높아지며, 하구의 연결성이 확보되어 다양한 종의 출현에 의한 결과로 확인되었다. KEFAI는 해역별, 하구유형별로 차이를 보였으며, 특성 또한 다르게 나타났다. 하구의 메트릭은 8개 항목으로, 해역별로 M8 항목을 제외한 모든 항목에서 통계적 차이(P < 0.05)를 보였고, 열린하구와 닫힌하구는 M1, M2 항목을 제외한 6개 메트릭 항목에서 통계적 차이를 보였다(P < 0.05). PCA 결과에서 서해역은 닫힌하구의 빈도가 높아, 닫힌하구의 특성을 공유하며, 높은 다양도 및 종수, 낮은 기수성어종수, 높은 내성종개체수 비율이 확인되었다. 남해역은 열린하구의 빈도가 높고, 열린하구의 특성을 공유하며, 낮은 다양도, 적은 종수, 높은 저서성어종의 개체수 비율이 확인되었다. 동해역은 열린하구의 빈도가 가장 높고, 열린하구의 특성을 띠며, 높은 다양도, 많은 종수, 낮은 내성종개체수 비율 및 낮은 저서종어종의 개체수 비율이 확인되었다. 국내에서 참조하구와 훼손하구에 관한 연구는 진행되지 않았으나, 참조하천과 훼손하천에 준하는 기준을 적용한 결과, 8개의 메트릭에서 모두 통계적 차이가 발생하였다(P < 0.05). 참조하구는 남해역, 열린하구의 특성을 띠며, 훼손하구는 서해역, 닫힌하구의 특성을 갖는다. 메트릭 항목은 2008년부터 현재까지 지속적으로 개정됐으나, 최초전수조사 결과를 반영하지 못하고 있다. 본 연구 결과에서 각각의 메트릭 항목을 분석한 결과 M1, M2, M3, M6, M7 항목은 일부 조정이 필요한 것으로 판단된다. 개선된 메트릭지수를 이용해 KEFAI를 산출한 결과, 건강성이 상승하는 것으로 나타났으며, 현재의 하구 건강성 평가체계는 KEFAI가 저평가된 것으로 판단된다. 본 연구는 2016년부터 3년 단위로 조사한 최초전수조사 결과를 바탕으로 분석하였다. 2008년 전후로 건강성 평가의 개념 정립 및 조사가 수행되었으며, 다년간의 연구로 많은 데이터가 누적되었다. 하지만 하구 수생태계 조사 및 건강성 평가 사업의 특성상 모니터링을 중점으로 하고 있다. 현재까지 다수의 논문과 분석 방법이 적용되었으나 일부 하구를 중심으로 연구가 진행되었으며 어류를 중심의 총괄적인 분석은 이루어지지 않았다. 따라서 본 연구에서 대한민국의 하구에 서식하는 어류의 군집구조, 하구의 물리·화학적 특성, 하구의 형태 등을 분석하였다. 또한, 기존의 멀티 메트릭을 검증하여 개선 방향을 제시하였다. 하구 수생태계 조사 및 건강성 평가는 다양한 분류군을 종합적으로 평가하지만, 본 연구는 어류를 중심으로 분석하였기 때문에 추후 다른 분류군과의 총체적인 분석이 필요하다. 이를 통해 하구수생태의 특성을 명확하게 파악하여 하구의 관리, 정책, 연구, 활용 등의 기준 정보로 제시할 수 있을 것으로 판단된다. 환경유전자 연구 결과에서 문헌연구 결과 66종, 환경유전자 결과 59종의 어류가 확인되었으며, 공통 출현종은 29종으로 나타났다. 기존의 전통적인 어류 연구방법을 통해서는 충분한 어류상을 확인할 수 없음을 확인하였다. 우점종은 문헌조사에서 민물검정망둑, 블루길, 피라미가 하구, 호소, 하천습지에서 우점하는 것으로 나타났으나, 환경유전자 연구에서는 모두 기벨리오붕어가 우점하는 것으로 나타났다. 본 연구에서 사용한 Primer는 국내 어류를 대상으로 개발된 Primer가 아니기 때문에 국내종을 정확히 판단하기에는 어려울 것으로 판단된다. 기벨리오붕어와 유사종인 붕어, 떡붕어 등은 완벽하게 구분할 수 없을 것으로 판단되며, 기벨리오붕어의 높은 OTUs는 기존의 붕어와 떡붕어의 유전자도 중복적으로 산출된 결과로 판단된다. 기벨리오붕어 외에도, 붕어교잡종(Carassius auratus x Megalobrama amblycephala pentaploid hybrid), 미꾸리교잡종(Misgurnus anguillicaudatus x Misgurnus bipartitus) 등 기존에 확인되지 않던 종들이 출현하여, 추가적인 연구가 필요할 것으로 판단된다. 알파종다양성과 베타종다양성은 일부 항목에서 차이를 보였다(P < 0.05). 각각 8개의 메트릭으로 구성된 FAI, KEFAI 메트릭에서 각각 3개, 4개 항목은 종수에 영향을 받는다. 따라서 명확한 평가를 위해 환경유전자 조사 결과를 보조도구로 사용하면 정확한 지표 및 결과를 생산할 수 있을 것으로 판단된다. 현재 국내의 환경유전자 연구는 초기단계이며, 국내 어종을 정확히 구분할 수 있는 universal primer의 부재 등 다양한 문제점을 내포하고 있다. 본 연구에서도 붕어와 그 유사종의 구분에 대한 문제점을 확인하였다. 향후 어류조사에서 보편적인 방법으로 활용하기 위해서는 다양한 환경에서의 비교조사를 통한 검증이 필요하다. 따라서 국내 어종에 맞는 Primer의 개발, 습지별 보편적인 연구방법의 정착, OTUs의 개체수준에서의 활용 연구 및 검증이 필요하며, 이를 통해 보편적인 조사 방법으로 활용하고, 신뢰 있는 결과를 도출할 수 있을 것으로 판단된다. 1. 서 설(General Introduction)
한반도의 하구 연구는 2000년대 초반까지 국가적인 관심이 크게 나타나는 대형하구와 일부 소형하구 등 단일 하구를 중심으로 이루어졌다(Lim et al., 2020). 과거의 국내에서 이루어진 하구 연구는 대부분 수질 관련 현황 조사가 주로 실시되었으며, 이외에 유동, 퇴적모델에 관한 연구가 일부 수행되었다. 대형하구와 달리 중·소규모의 하구 연구는 적었으며, 국내 하구에 대한 이해가 부족했다. 세계적으로 생태 및 환경에 관한 관심이 증대되면서, 국내에서도 이에 맞춰 목표를 설정하여, 집중적인 관리를 수행하였다. 환경부에서는 물환경관리 목표를 “이화학적 수질”에서 “수생태계 건강성”으로 개념을 확대하고 2008년부터 하천 및 하구의 수생태계 현황 조사 및 건강성 평가를 하고 있다(Won et al., 2022). 2008년 전후로 부착돌말류, 저서성대형무척추동물, 어류를 이용한 건강성 평가 방법이 국내 상황에 맞춰 개발되었으며, 지속적인 개정을 통해 완성도를 높였다. 하구 수생태계 현황 조사 및 건강성 평가는 2017년부터 “물환경보전법” 제9조의 3(수생태계 현황 조사 및 건강성 평가), 동법 시행규칙 제 24조의 2(수생태계 현황 조사), 제24조의 3(수생태계 건강성 평가)에 따라 진행되고 있다. 하구 수생태계 현황 조사 및 건강성 평가는 자연환경보전법 제 30조의 규정에 따른 자연환경조사 및 습지보전법 제4조의 규정에 따른 습지조사와 함께 법정조사에 속한다. 하구 수생태계 현황 조사 및 건강성 평가는 기존의 법정조사보다 늦게 시작되었으나, 과학적인 평가 방법을 도입하여 조사 대상지를 등급화하여 평가하고 있다. 표준화된 조사지침을 개발하여, 모든 연구원들이 동일한 방법으로, 안정적이고 신뢰성 높은 조사가 가능하게 되었으며, 최근 정도관리의 중요성이 높아지면서 더욱더 높은 신뢰도의 데이터를 양산하고 있다. 하구는 하천의 끝단에 위치하며, 담수와 해수가 섞이고, 반폐쇄해의 특성을 갖는 지역이다(Pritchard, 1967). 하구는 담수와 해수가 혼합되며, 조석의 영향으로 인해 다양한 환경조건을 갖춰 생물의 구성이 다양하다(van Westen and Scheele, 1996; Goudie, 2004). 하구는 육상기원의 퇴적물 및 오염물질이 분해되고, 다양한 생물의 주요 서식지 및 산란지로서의 가치가 매우 높다(Costanza et al., 1997). 어류에 있어 하구는 다양한 어종이 공존하는 서식지이며, 하천과 바다를 잇는 회유성어종의 이동 통로이다(Potter et al., 1986). 하구의 다양한 수심과 높은 탁도는 어류의 생존율을 높이며, 다양한 산란장 및 서식처를 제공한다(McLusky and McCrory 1989). 한반도는 삼면이 바다와 접하며, 3개의 해역이 각각 다른 특성을 띠어 다양성을 더욱 증대시킨다. 3개의 해역은 각기 다른 지형적 특성으로 인해 하구의 어류군집이 다르게 형성된다. 또한 고생물지리학 측면에서 민물고기류의 지리적 구분은 서한아지역(West Korea subdistrict), 남한아지역(South Korea subdistrict), 동북한아지역(Eastnorth Korea subdistrict)으로 구분되며(Kim and Park, 2002) 해역별 하구의 어류군집에서 차이를 발생시킨다. 횡구조물 유무(Park et al., 2022), 물리·화학적 특성(Eick and Thiel 2014; Pease 1999; Han and An 2013; Harrison and Whitfield 2006; Meador and Goldstein 2003; Nicolas et al., 2010; Park et al., 2022), 하상기질(Morrow and Fischenich 2000; Pihl et al., 2002; Pusey and Arthington 2003; Hossain et al., 2012) 등은 모두 어류군집에 영향을 끼치며 개체군의 생존, 성장, 번식, 분포에 영향을 끼치기 때문에 모니터링을 기반으로 지속적으로 관리해야 한다. 미국 및 유럽 등 선진국에서는 수자원의 총체성을 반영하는 수생태계 건강성 회복을 물관리의 최상 목표로 설정하고 1990년대 초반부터 수질에 대한 생물학적 평가체계를 도입하여 활용하고 있다. “하구 수생태계 현황 조사 및 건강성 평가(MOE/NIER, 2008~2022)”는 국내 전체 하구(대규모하구, 도서 지역, 하구호 제외)를 대상으로 동시에 조사를 수행하는 대규모 국가 프로젝트이다. 이를 통해 도출된 결과는 한반도 하구의 어류군집의 현황과 특성을 이해하고, 하구의 보전 및 복원을 위한 자료로 사용된다. 하구별 3년 주기로 연 2회 평가를 통해 KEFAI를 산출하여, 하구의 건강성을 파악한다. KEFAI는 현재까지 8회에 걸쳐서 정교화를 추진하여, 2016년부터 현재 형태의 메트릭 평가 방법을 사용하고 있다. 하지만 현재 사용되는 멀티 메트릭은 3년 주기로 연 2회씩 조사된 최초전수조사 결과를 반영하지 못하고 있다. 또한, 현재의 연구체계에서는 표본조사(sample survey)시 발생하는 문제점을 내포하고 있다. 일반적인 생태계 조사에서 표본조사는 과거부터 현재까지 널리 사용한 방법이다. 하지만 표본조사는 표본크기, 채집위치, 채집방법, 측정방법에 따라 결과값이 다르며, 이외에 조사강도, 비용, 인원 등의 문제가 발생한다(Kim et al., 2020a; Portt et al., 2006). 따라서 환경유전자 조사 방법은 이와 같은 표본조사의 문제점을 보완하는 방법으로 사용할 수 있다. 환경유전자(environmental DNA, eDNA)는 다양한 환경 시료에서 추출된 DNA를 총칭하며, 모든 유기물로부터 데이터를 수집한다(Taberlet et al., 2012, Thomsen and Willerslev, 2015). 환경유전자는 미생물의 다양성에 관한 연구를 중심으로 연구되었으며, 2000년대부터 다른 분야에서 활용하기 시작하였다. 최근 차세대 염기서열분석(next-generation sequencers, NGS)의 발달로 생물다양성 연구 및 희귀종 탐색의 방법론으로 사용하고 있다(Kwak et al., 2021a). 특히 수환경에 서식하는 생물에 대한 유전정보를 비교적 간편하게 획득하고 분석할 수 있어, 수생물을 대상으로 다양한 연구들이 수행되고 있다(Sassoubre et al., 2016; Shaw et al., 2016; Evans et al., 2017). 환경유전자를 활용한 수생생물 조사는 수체 내 잔존 유전자를 분석하기 때문에 포획없이 생물의 존재 여부를 확인할 수 있다(Kim et al., 2020a). 이와 같은 특성으로 인해 기존의 현장조사 방법의 단점을 보완할 수 있다. 전통적인 모니터링 방법은 직접적인 포획방식으로 정량적인 결과를 산출할 수 있고, 서식 어종을 직접 확인할 수 있으며, 질병 및 기형 등의 유무를 파악할 수 있다. 반면, 어구 특성으로 인해 채집종의 편향성(gear selectivity)이 발생하며, 다양성 및 특정종의 개체군의 크기가 저평가될 수 있다(Bonar et al., 2009; Kim et al., 2020b). 물리적으로 채집이 어려운 환경에 주로 서식하는 종이거나, 개체수가 매우 적은 멸종위기종의 경우 직접적인 채집을 위해서는 많은 시간과 인력이 필요하며, 채집되지 않는 경우 해당 지역의 생물상이 저평가되는 결과를 발생시킨다. 국내에서 환경유전자 연구는 법정조사에 속하지 않기 때문에 그 지원과 연구는 현재 매우 부족한 상황이다. 따라서, 본 연구는 가장 최근에 이루어진 2016~2018년의 한반도 하구의 어류조사 결과를 활용하여 한반도 하구의 어류군집 현황과 특성에 대한 종합적인 분석 자료와 함께, 해역별, 하구유형별, 수계별, 하구규모별 어류군집의 특성 및 주요종의 서식 환경 및 분포에 대한 정보를 제공하였다. 어류군집을 물리·화학적 특성을 비교하고, 어류군집과의 상관관계를 도출하였으며, 정준상관분석(CCA)을 통해 물리·화학 항목과 어류군집의 상관성을 도출하였다. 또한 KEFAI 평가를 통해 한반도 하구의 현황을 제시하였으며, 메트릭 분석을 통해 최초전수조사 결과를 반영한 신규 메트릭 평가안을 제시하였다. 환경유전자 연구를 통해 경남권역에서 하천, 습지, 하구 등 다양한 환경에 적용하여, 어종의 다양성을 확인하고, 국내 내수면에서의 환경유전자 연구방법의 적용 가능성을 파악하였다. 또한, 하구 수생태계 현황 조사 및 건강성 평가 방법에 적용되는 멀티메트릭을 환경유전자 조사 방법을 통해 보정 및 적용 가능성에 대해 검토하였다.

PCA 결과에서 서해역은 닫힌하구의 빈도가 높아, 닫힌하구의 특성을 공유하며, 높은 다양도 및 종수, 낮은 기수성어종수, 높은 내성종개체수 비율이 확인되었다. 남해역은 열린하구의 빈도가 높고, 열린하구의 특성을 공유하며, 낮은 다양도, 적은 종수, 높은 저서성어종의 개체수 비율이 확인되었다. 동해역은 열린하구의 빈도가 가장 높고, 열린하구의 특성을 띠며, 높은 다양도, 많은 종수, 낮은 내성종개체수 비율 및 낮은 저서종어종의 개체수 비율이 확인되었다. 국내에서 참조하구와 훼손하구에 관한 연구는 진행되지 않았으나, 참조하천과 훼손하천에 준하는 기준을 적용한 결과, 8개의 메트릭에서 모두 통계적 차이가 발생하였다(P < 0.05). 참조하구는 남해역, 열린하구의 특성을 띠며, 훼손하구는 서해역, 닫힌하구의 특성을 갖는다. 메트릭 항목은 2008년부터 현재까지 지속적으로 개정됐으나, 최초전수조사 결과를 반영하지 못하고 있다. 본 연구 결과에서 각각의 메트릭 항목을 분석한 결과 M1, M2, M3, M6, M7 항목은 일부 조정이 필요한 것으로 판단된다. 개선된 메트릭지수를 이용해 KEFAI를 산출한 결과, 건강성이 상승하는 것으로 나타났으며, 현재의 하구 건강성 평가체계는 KEFAI가 저평가된 것으로 판단된다. 본 연구는 2016년부터 3년 단위로 조사한 최초전수조사 결과를 바탕으로 분석하였다. 2008년 전후로 건강성 평가의 개념 정립 및 조사가 수행되었으며, 다년간의 연구로 많은 데이터가 누적되었다. 하지만 하구 수생태계 조사 및 건강성 평가 사업의 특성상 모니터링을 중점으로 하고 있다. 현재까지 다수의 논문과 분석 방법이 적용되었으나 일부 하구를 중심으로 연구가 진행되었으며 어류를 중심의 총괄적인 분석은 이루어지지 않았다. 따라서 본 연구에서 대한민국의 하구에 서식하는 어류의 군집구조, 하구의 물리·화학적 특성, 하구의 형태 등을 분석하였다. 또한, 기존의 멀티 메트릭을 검증하여 개선 방향을 제시하였다. 하구 수생태계 조사 및 건강성 평가는 다양한 분류군을 종합적으로 평가하지만, 본 연구는 어류를 중심으로 분석하였기 때문에 추후 다른 분류군과의 총체적인 분석이 필요하다. 이를 통해 하구수생태의 특성을 명확하게 파악하여 하구의 관리, 정책, 연구, 활용 등의 기준 정보로 제시할 수 있을 것으로 판단된다. 환경유전자 연구 결과에서 문헌연구 결과 66종, 환경유전자 결과 59종의 어류가 확인되었으며, 공통 출현종은 29종으로 나타났다. 기존의 전통적인 어류 연구방법을 통해서는 충분한 어류상을 확인할 수 없음을 확인하였다. 우점종은 문헌조사에서 민물검정망둑, 블루길, 피라미가 하구, 호소, 하천습지에서 우점하는 것으로 나타났으나, 환경유전자 연구에서는 모두 기벨리오붕어가 우점하는 것으로 나타났다. 본 연구에서 사용한 Primer는 국내 어류를 대상으로 개발된 Primer가 아니기 때문에 국내종을 정확히 판단하기에는 어려울 것으로 판단된다. 기벨리오붕어와 유사종인 붕어, 떡붕어 등은 완벽하게 구분할 수 없을 것으로 판단되며, 기벨리오붕어의 높은 OTUs는 기존의 붕어와 떡붕어의 유전자도 중복적으로 산출된 결과로 판단된다. 기벨리오붕어 외에도, 붕어교잡종(Carassius auratus x Megalobrama amblycephala pentaploid hybrid), 미꾸리교잡종(Misgurnus anguillicaudatus x Misgurnus bipartitus) 등 기존에 확인되지 않던 종들이 출현하여, 추가적인 연구가 필요할 것으로 판단된다. 알파종다양성과 베타종다양성은 일부 항목에서 차이를 보였다(P < 0.05). 각각 8개의 메트릭으로 구성된 FAI, KEFAI 메트릭에서 각각 3개, 4개 항목은 종수에 영향을 받는다. 따라서 명확한 평가를 위해 환경유전자 조사 결과를 보조도구로 사용하면 정확한 지표 및 결과를 생산할 수 있을 것으로 판단된다. 현재 국내의 환경유전자 연구는 초기단계이며, 국내 어종을 정확히 구분할 수 있는 universal primer의 부재 등 다양한 문제점을 내포하고 있다. 본 연구에서도 붕어와 그 유사종의 구분에 대한 문제점을 확인하였다. 향후 어류조사에서 보편적인 방법으로 활용하기 위해서는 다양한 환경에서의 비교조사를 통한 검증이 필요하다. 따라서 국내 어종에 맞는 Primer의 개발, 습지별 보편적인 연구방법의 정착, OTUs의 개체수준에서의 활용 연구 및 검증이 필요하며, 이를 통해 보편적인 조사 방법으로 활용하고, 신뢰 있는 결과를 도출할 수 있을 것으로 판단된다. 1. 서 설(General Introduction)
한반도의 하구 연구는 2000년대 초반까지 국가적인 관심이 크게 나타나는 대형하구와 일부 소형하구 등 단일 하구를 중심으로 이루어졌다(Lim et al., 2020). 과거의 국내에서 이루어진 하구 연구는 대부분 수질 관련 현황 조사가 주로 실시되었으며, 이외에 유동, 퇴적모델에 관한 연구가 일부 수행되었다. 대형하구와 달리 중·소규모의 하구 연구는 적었으며, 국내 하구에 대한 이해가 부족했다. 세계적으로 생태 및 환경에 관한 관심이 증대되면서, 국내에서도 이에 맞춰 목표를 설정하여, 집중적인 관리를 수행하였다. 환경부에서는 물환경관리 목표를 “이화학적 수질”에서 “수생태계 건강성”으로 개념을 확대하고 2008년부터 하천 및 하구의 수생태계 현황 조사 및 건강성 평가를 하고 있다(Won et al., 2022). 2008년 전후로 부착돌말류, 저서성대형무척추동물, 어류를 이용한 건강성 평가 방법이 국내 상황에 맞춰 개발되었으며, 지속적인 개정을 통해 완성도를 높였다. 하구 수생태계 현황 조사 및 건강성 평가는 2017년부터 “물환경보전법” 제9조의 3(수생태계 현황 조사 및 건강성 평가), 동법 시행규칙 제 24조의 2(수생태계 현황 조사), 제24조의 3(수생태계 건강성 평가)에 따라 진행되고 있다. 하구 수생태계 현황 조사 및 건강성 평가는 자연환경보전법 제 30조의 규정에 따른 자연환경조사 및 습지보전법 제4조의 규정에 따른 습지조사와 함께 법정조사에 속한다. 하구 수생태계 현황 조사 및 건강성 평가는 기존의 법정조사보다 늦게 시작되었으나, 과학적인 평가 방법을 도입하여 조사 대상지를 등급화하여 평가하고 있다. 표준화된 조사지침을 개발하여, 모든 연구원들이 동일한 방법으로, 안정적이고 신뢰성 높은 조사가 가능하게 되었으며, 최근 정도관리의 중요성이 높아지면서 더욱더 높은 신뢰도의 데이터를 양산하고 있다. 하구는 하천의 끝단에 위치하며, 담수와 해수가 섞이고, 반폐쇄해의 특성을 갖는 지역이다(Pritchard, 1967). 하구는 담수와 해수가 혼합되며, 조석의 영향으로 인해 다양한 환경조건을 갖춰 생물의 구성이 다양하다(van Westen and Scheele, 1996; Goudie, 2004). 하구는 육상기원의 퇴적물 및 오염물질이 분해되고, 다양한 생물의 주요 서식지 및 산란지로서의 가치가 매우 높다(Costanza et al., 1997). 어류에 있어 하구는 다양한 어종이 공존하는 서식지이며, 하천과 바다를 잇는 회유성어종의 이동 통로이다(Potter et al., 1986). 하구의 다양한 수심과 높은 탁도는 어류의 생존율을 높이며, 다양한 산란장 및 서식처를 제공한다(McLusky and McCrory 1989). 한반도는 삼면이 바다와 접하며, 3개의 해역이 각각 다른 특성을 띠어 다양성을 더욱 증대시킨다. 3개의 해역은 각기 다른 지형적 특성으로 인해 하구의 어류군집이 다르게 형성된다. 또한 고생물지리학 측면에서 민물고기류의 지리적 구분은 서한아지역(West Korea subdistrict), 남한아지역(South Korea subdistrict), 동북한아지역(Eastnorth Korea subdistrict)으로 구분되며(Kim and Park, 2002) 해역별 하구의 어류군집에서 차이를 발생시킨다. 횡구조물 유무(Park et al., 2022), 물리·화학적 특성(Eick and Thiel 2014; Pease 1999; Han and An 2013; Harrison and Whitfield 2006; Meador and Goldstein 2003; Nicolas et al., 2010; Park et al., 2022), 하상기질(Morrow and Fischenich 2000; Pihl et al., 2002; Pusey and Arthington 2003; Hossain et al., 2012) 등은 모두 어류군집에 영향을 끼치며 개체군의 생존, 성장, 번식, 분포에 영향을 끼치기 때문에 모니터링을 기반으로 지속적으로 관리해야 한다. 미국 및 유럽 등 선진국에서는 수자원의 총체성을 반영하는 수생태계 건강성 회복을 물관리의 최상 목표로 설정하고 1990년대 초반부터 수질에 대한 생물학적 평가체계를 도입하여 활용하고 있다. “하구 수생태계 현황 조사 및 건강성 평가(MOE/NIER, 2008~2022)”는 국내 전체 하구(대규모하구, 도서 지역, 하구호 제외)를 대상으로 동시에 조사를 수행하는 대규모 국가 프로젝트이다. 이를 통해 도출된 결과는 한반도 하구의 어류군집의 현황과 특성을 이해하고, 하구의 보전 및 복원을 위한 자료로 사용된다. 하구별 3년 주기로 연 2회 평가를 통해 KEFAI를 산출하여, 하구의 건강성을 파악한다. KEFAI는 현재까지 8회에 걸쳐서 정교화를 추진하여, 2016년부터 현재 형태의 메트릭 평가 방법을 사용하고 있다. 하지만 현재 사용되는 멀티 메트릭은 3년 주기로 연 2회씩 조사된 최초전수조사 결과를 반영하지 못하고 있다. 또한, 현재의 연구체계에서는 표본조사(sample survey)시 발생하는 문제점을 내포하고 있다. 일반적인 생태계 조사에서 표본조사는 과거부터 현재까지 널리 사용한 방법이다. 하지만 표본조사는 표본크기, 채집위치, 채집방법, 측정방법에 따라 결과값이 다르며, 이외에 조사강도, 비용, 인원 등의 문제가 발생한다(Kim et al., 2020a; Portt et al., 2006). 따라서 환경유전자 조사 방법은 이와 같은 표본조사의 문제점을 보완하는 방법으로 사용할 수 있다. 환경유전자(environmental DNA, eDNA)는 다양한 환경 시료에서 추출된 DNA를 총칭하며, 모든 유기물로부터 데이터를 수집한다(Taberlet et al., 2012, Thomsen and Willerslev, 2015). 환경유전자는 미생물의 다양성에 관한 연구를 중심으로 연구되었으며, 2000년대부터 다른 분야에서 활용하기 시작하였다. 최근 차세대 염기서열분석(next-generation sequencers, NGS)의 발달로 생물다양성 연구 및 희귀종 탐색의 방법론으로 사용하고 있다(Kwak et al., 2021a). 특히 수환경에 서식하는 생물에 대한 유전정보를 비교적 간편하게 획득하고 분석할 수 있어, 수생물을 대상으로 다양한 연구들이 수행되고 있다(Sassoubre et al., 2016; Shaw et al., 2016; Evans et al., 2017). 환경유전자를 활용한 수생생물 조사는 수체 내 잔존 유전자를 분석하기 때문에 포획없이 생물의 존재 여부를 확인할 수 있다(Kim et al., 2020a). 이와 같은 특성으로 인해 기존의 현장조사 방법의 단점을 보완할 수 있다. 전통적인 모니터링 방법은 직접적인 포획방식으로 정량적인 결과를 산출할 수 있고, 서식 어종을 직접 확인할 수 있으며, 질병 및 기형 등의 유무를 파악할 수 있다. 반면, 어구 특성으로 인해 채집종의 편향성(gear selectivity)이 발생하며, 다양성 및 특정종의 개체군의 크기가 저평가될 수 있다(Bonar et al., 2009; Kim et al., 2020b). 물리적으로 채집이 어려운 환경에 주로 서식하는 종이거나, 개체수가 매우 적은 멸종위기종의 경우 직접적인 채집을 위해서는 많은 시간과 인력이 필요하며, 채집되지 않는 경우 해당 지역의 생물상이 저평가되는 결과를 발생시킨다. 국내에서 환경유전자 연구는 법정조사에 속하지 않기 때문에 그 지원과 연구는 현재 매우 부족한 상황이다. 따라서, 본 연구는 가장 최근에 이루어진 2016~2018년의 한반도 하구의 어류조사 결과를 활용하여 한반도 하구의 어류군집 현황과 특성에 대한 종합적인 분석 자료와 함께, 해역별, 하구유형별, 수계별, 하구규모별 어류군집의 특성 및 주요종의 서식 환경 및 분포에 대한 정보를 제공하였다. 어류군집을 물리·화학적 특성을 비교하고, 어류군집과의 상관관계를 도출하였으며, 정준상관분석(CCA)을 통해 물리·화학 항목과 어류군집의 상관성을 도출하였다. 또한 KEFAI 평가를 통해 한반도 하구의 현황을 제시하였으며, 메트릭 분석을 통해 최초전수조사 결과를 반영한 신규 메트릭 평가안을 제시하였다. 환경유전자 연구를 통해 경남권역에서 하천, 습지, 하구 등 다양한 환경에 적용하여, 어종의 다양성을 확인하고, 국내 내수면에서의 환경유전자 연구방법의 적용 가능성을 파악하였다. 또한, 하구 수생태계 현황 조사 및 건강성 평가 방법에 적용되는 멀티메트릭을 환경유전자 조사 방법을 통해 보정 및 적용 가능성에 대해 검토하였다.

This study investigated the fish assemblages and physical and chemical characteristics in 328 estuaries sites from 2016 to 2018. The study utilized KEFAI's multi-metric index, providing an improved metric system based on the initial comprehensive survey results. In 2022, a comparison was made between literature research and environmental genetic results in 21 wetland areas in Gyeongsangnam-do, to assess the characteristics of each survey method and explore the applicability of supplementary tools in KEFAI's multi-metric index for current estuaries ecosystem status surveys and health assessments. Over the course of three years, the Korean Peninsula's estuaries revealed a higher diversity of fish species, with 52 families and 150 species identified. The dominant family was the Cyprinidae, accounting for 42.8% of the species, with the T. hakonensis(RA, 12.5%) as the dominant species and the M. cephalus(RA, 9.5%) as the subdominant species. Two endangered species, the P. sinensis and C. hangiongensis, were found, both in waters closer to the rivers in the East Sea. There were 21 endemic species, 5 exotic species, 8 anadromours species, 41 brackish species, 39 marine fish species, and 62 primary freshwater fish species recorded. In particular, the anadromours species consisted of valuable resources. The management of A. japonica should be prioritized in the West Sea, P. altivelis and A. japonica should be focused on in the South Sea, and T. hakonensis, O. keta, P. altivelis, and H. nipponensis should be managed intensively in the East Sea. It is necessary to address the decline and management of anadromours species through active water management of transverse structures that block estuaries connectivity. The SIMPER analysis revealed differences in cluster composition among estuaries based on the sea area and estuary type. The representative species for estuaries on the Korean Peninsula were M. cephalus, C. auratus, and T. brevispinis. In the West Sea region, the representative species were H. leucisculus and C. auratus, while in the South Sea region, they were M. cephalus and C. haematocheilus. In the East Sea region, the representative species T. hakonensis and M. cephalus. Open estuaries showed a prevalence of M. cephalus and T. brevispinis, while closed estuaries showed a prevalence of T. hakonensis and M. cephalus. In terms of physical and chemical characteristics, the East Sea exhibited the characteristics of an open estuary, while the West Sea displayed the characteristics of a closed estuary. The West Sea, representing a closed estuary, showed statistical differences from the East Sea in terms of pollution indicators such as turbidity, chemical oxygen demand (COD), total nitrogen (TN), and total phosphorus (TP). These indicators were found to have a significant impact on fish populations. In contrast, the East Sea, representing an open estuary, exhibited high levels of dissolved oxygen (DO), electrical conductivity, and salinity, showing characteristics opposite to those of the West Sea. The Canonical Correspondence Analysis (CCA) results also clearly demonstrated differences between the sea areas and estuary types, and the fish communities corresponded to the results obtained from the SIMPER analysis. The average KEFAI score for estuaries on the Korean Peninsula was 50.52(grade C), with variations observed among marine regions. The West Sea region scored 41.84(grade C), the South Sea region scored 52.71(grade C), and the East Sea region scored 55.48(grade C), indicating higher KEFAI scores as we move from the West Sea to the East Sea. This trend can be attributed to the higher proportion of open estuaries and improved connectivity, which results in the presence of a greater variety of species. KEFAI showed differences and distinct characteristics among marine regions and estuary types. The metric system for estuaries consisted of eight components, and statistically significant differences(P < 0.05) were observed for all components except M8 across sea area. Open and closed estuaries showed statistically significant differences (P < 0.05) in six metric components, excluding M1 and M2. PCA results indicated that the West Sea region exhibited characteristics of closed estuaries, with high diversity and species richness, low benthic fish species richness, and a high ratio of tolerant species. The South Sea region exhibited characteristics of open estuaries, with lower diversity, fewer species, and a higher ratio of benthic fish species. The East Sea region showed high diversity, a larger number of species, a lower ratio of tolerant species, and a lower ratio of benthic species. While there have been no specific studies on reference and impaired estuaries in Korea, applying criteria similar to reference and impaired rivers revealed statistically significant differences in all eight metrics(P < 0.05). Reference estuaries showed characteristics similar to the South Sea region and open estuaries, while impaired estuaries showed characteristics similar to the West Sea region and closed estuaries. Although the metric system has been continuously revised since 2008, it does not currently reflect the initial comprehensive survey results. The analysis of the initial comprehensive survey data in this study suggests that adjustments are needed for metrics M1, M2, M3, M6, and M7. Using the improved metric index to calculate KEFAI scores resulted in an increase in estuary health, indicating that the current estuary health assessment system underestimates the health status based on KEFAI. This study analyzed the results of the initial comprehensive survey conducted every three years since 2016. The concept and investigation were established around 2008, and extensive data has been accumulated over the years. However, due to the “survey and assessment estuary ecosystem health” evaluation projects, the focus has primarily been on monitoring. Numerous papers and analysis methods have been applied to date, but research has been conducted primarily on specific estuaries, and a comprehensive analysis of fish species has not been carried out. Therefore, this study aimed to analyze the overall community structure of fish species inhabiting estuaries in South Korea, as well as the physical and chemical characteristics and morphology of estuaries. Additionally, the study validated the existing multi-metric approach and suggested areas for improvement. This research focused on fish species and aimed to comprehensively evaluate the estuaries ecosystem and health assessment, considering various taxonomic groups. Therefore, future studies should conduct holistic analyses involving other taxonomic groups to gain a clear understanding of estuaries characteristics. This will provide valuable information for estuary management, policies, research, and utilization. According to the literature review, a total of 66 fish species were identified, while the environmental DNA analysis revealed 59 species. Among them, 29 species were found to be common to both methods. It was confirmed that traditional fish research methods alone were insufficient to adequately assess the fish assemblage. The dominant species identified through literature research were T. brevispinis, L. macrochirus, and Z. platypus in the estuaries, lakes, and river wetlands. However, the environmental DNA analysis indicated that all these habitats were dominated by C. gibelio. The primers used in this study were not specifically developed for domestic species, making it difficult to accurately identify local species. It is believed that it would be challenging to distinguish C. gibelio from similar species such as C. auratus and C. cuvieri, as their genetic material may have been detected redundantly within the high OTUs of C. gibelio. In addition to C. gibelio, previously unreported species such as the hybrid of C. auratus and M. amblycephala and the hybrid of M. anguillicaudatus and M. bipartitus were also found, indicating the need for further research. Alpha and beta diversity showed differences in some metrics (P < 0.05). Three out of the eight metrics in the FAI and four out of the eight metrics in the KEFAI were found to be influenced by species richness. Therefore, it is suggested that the results of environmental DNA analysis can be used as a supplementary tool to produce accurate indicators and results. Currently, environmental DNA research in South Korea is in its early stages and faces various issues, including the absence of universal primers for accurately distinguishing domestic species. In order to utilize it as a universal method in future fish surveys, it is necessary to validate it through comparative studies in various environments. Therefore, the development of primers specific to domestic fish species, the establishment of universally applicable research methods for different wetland types, research and validation of OTUs at the individual level are needed. Through these efforts, it is anticipated that the method can be used as a universal approach, leading to reliable results.

The metric system for estuaries consisted of eight components, and statistically significant differences(P < 0.05) were observed for all components except M8 across sea area. Open and closed estuaries showed statistically significant differences (P < 0.05) in six metric components, excluding M1 and M2. PCA results indicated that the West Sea region exhibited characteristics of closed estuaries, with high diversity and species richness, low benthic fish species richness, and a high ratio of tolerant species. The South Sea region exhibited characteristics of open estuaries, with lower diversity, fewer species, and a higher ratio of benthic fish species. The East Sea region showed high diversity, a larger number of species, a lower ratio of tolerant species, and a lower ratio of benthic species. While there have been no specific studies on reference and impaired estuaries in Korea, applying criteria similar to reference and impaired rivers revealed statistically significant differences in all eight metrics(P < 0.05). Reference estuaries showed characteristics similar to the South Sea region and open estuaries, while impaired estuaries showed characteristics similar to the West Sea region and closed estuaries. Although the metric system has been continuously revised since 2008, it does not currently reflect the initial comprehensive survey results. The analysis of the initial comprehensive survey data in this study suggests that adjustments are needed for metrics M1, M2, M3, M6, and M7. Using the improved metric index to calculate KEFAI scores resulted in an increase in estuary health, indicating that the current estuary health assessment system underestimates the health status based on KEFAI. This study analyzed the results of the initial comprehensive survey conducted every three years since 2016. The concept and investigation were established around 2008, and extensive data has been accumulated over the years. However, due to the “survey and assessment estuary ecosystem health” evaluation projects, the focus has primarily been on monitoring. Numerous papers and analysis methods have been applied to date, but research has been conducted primarily on specific estuaries, and a comprehensive analysis of fish species has not been carried out. Therefore, this study aimed to analyze the overall community structure of fish species inhabiting estuaries in South Korea, as well as the physical and chemical characteristics and morphology of estuaries. Additionally, the study validated the existing multi-metric approach and suggested areas for improvement. This research focused on fish species and aimed to comprehensively evaluate the estuaries ecosystem and health assessment, considering various taxonomic groups. Therefore, future studies should conduct holistic analyses involving other taxonomic groups to gain a clear understanding of estuaries characteristics. This will provide valuable information for estuary management, policies, research, and utilization. According to the literature review, a total of 66 fish species were identified, while the environmental DNA analysis revealed 59 species. Among them, 29 species were found to be common to both methods. It was confirmed that traditional fish research methods alone were insufficient to adequately assess the fish assemblage. The dominant species identified through literature research were T. brevispinis, L. macrochirus, and Z. platypus in the estuaries, lakes, and river wetlands. However, the environmental DNA analysis indicated that all these habitats were dominated by C. gibelio. The primers used in this study were not specifically developed for domestic species, making it difficult to accurately identify local species. It is believed that it would be challenging to distinguish C. gibelio from similar species such as C. auratus and C. cuvieri, as their genetic material may have been detected redundantly within the high OTUs of C. gibelio. In addition to C. gibelio, previously unreported species such as the hybrid of C. auratus and M. amblycephala and the hybrid of M. anguillicaudatus and M. bipartitus were also found, indicating the need for further research. Alpha and beta diversity showed differences in some metrics (P < 0.05). Three out of the eight metrics in the FAI and four out of the eight metrics in the KEFAI were found to be influenced by species richness. Therefore, it is suggested that the results of environmental DNA analysis can be used as a supplementary tool to produce accurate indicators and results. Currently, environmental DNA research in South Korea is in its early stages and faces various issues, including the absence of universal primers for accurately distinguishing domestic species. In order to utilize it as a universal method in future fish surveys, it is necessary to validate it through comparative studies in various environments. Therefore, the development of primers specific to domestic fish species, the establishment of universally applicable research methods for different wetland types, research and validation of OTUs at the individual level are needed. Through these efforts, it is anticipated that the method can be used as a universal approach, leading to reliable results.
