Lactic acid is a representative biomarker reflecting the body's fatigue. Lactic acid is present in human sweat and blood, and if it accumulates in the body and is not properly removed, it can lead to various pathological symptoms ranging from circulatory disorders to heart attacks, trauma, and stress. Developing a convenient biosensor for the prevention and timely intervention of such conditions is crucial. In this study, we propose a method for detecting lactic acid by creating a paper-based sensor using Tween 80-coated polyaniline (TPAni) nanoparticles, inducing a color change in response to lactate present in human sweat. For the synthesis of TPAni nanoparticles, the Solvent Shift Method was employed, coating the surface of Polyaniline nanoparticles with Tween 80 as a surfactant. The synthesized TPAni nanoparicles were adsorbed onto the surface of filter paper using the Dip Coating Method. Subsequently, to facilitate electron transfer, ferrocene, and lactate oxidase enzymes were additionally adsorbed, resulting in the fabrication of a final paper sensor for lactic acid detection. The TPAni-based paper sensor created in this manner exhibited a color change from blue to green within 30 seconds in response to lactate and chemical reactions, which was observable to the naked eyes.To assess the detection performance of the colorimetric paper sensor, lactic acid concentration assays were conducted. The TPAni nanoparticle-based sensor platform demonstrated a detection limit (LOD) of 1 mM for lactic acid, linear behavior with a coefficient of 0.9684. In conclusion, through this study, we confirmed that the TPAni nanoparticle-based paper sensor platform can detect clinically significant lactic acid concentrations. The technology and approach developed here could serve as a foundation for the future development of a self-diagnostic biosensor kit for conveniently measuring individual physical fatigue.
