The aim of the present study was to assess the CH4 mitigation potential and impacts on the rumen fermentation status of red seaweed species and establish the ideal feed characteristics for a superior species by determining the optimal drying method and supplementation dosage. Furthermore, the current study elucidated the effects of the final feed form of red seaweed on microbial communities, enzymes, and metabolic pathways to suggest its mechanisms for alteration in rumen fermentation. The first part of the study (Chapter 2) investigated the CH4-reducing abilities of 38 species of red seaweeds and their effects on rumen fermentation characteristics. All samples were sun-dried, and each was added at 10% OM in semi-closed in vitro batch cultures. Among all the species, 11 species showed a reduction in gas production, with 6 species notably mitigating CH¬4 production. Hypnea charoides and Gracilaria chorda which reduced CH4 production by 35.68 and 30.47%, respectively, were effective in abating both CH4 production and proportion compared to the control. These candidates also did not induce significant changes in rumen fermentation parameters including total or individual volatile fatty acids and ammonia. This suggests that H. charoides and G. chorda effectively mitigated CH4 production without adversely affecting rumen fermentation status. The objective of the second study (Study 3.1 of Chapter 3) was to determine the optimal drying process for G. chorda based on CH4 production, rumen fermentation characteristics, and the contents of halogenated methane analogues (HMAs). The raw G. chorda was dried using various methods: oven-dried at 40℃ (OV40), at 60℃ (OV60), at 105℃ (OV105), or freeze-dried at -80℃ (FD). Each sample was added at 10% OM in semi-closed in vitro batch cultures. The 4 different methods did not notably affect gas production, but all treatments showed significant mitigations in both CH¬4 production and proportion. The OV105 was identified as the most effective form in abating CH4 production, exhibiting mitigation rates of 46.08, 29.78, and 17.63% at 12, 24, and 48 h of incubation times, respectively. The other processes (OV60, OV40, and FD) also reduced CH4 production at all sampling times, with FD showing the least CH4-reducing capability. All treatments resulted in an increase in pH, and only OV methods (OV105, OV60, and OV40) led to an increase in ammonia. Total volatile fatty acids were unaffected by the different drying methods, while acetate as well as acetate to propionate ratio were increased. A significant decrease in butyrate was observed, with no effects on propionate or branched-chain fatty acids, including iso-butyrate and iso-valerate. Some halogenated methane analogues, including bromoform and dibromochloromethane, were not detected in any G. chorda samples dried by different methods except for Asparagopsis taxiformis, which served as a positive control. These findings suggest that G. chorda dried using the OV105 method is the most viable form for use as a natural CH4-reducing additive. The purpose of the third study (Study 3.2 of Chapter 3) was to select the ideal supplementation dosage of G. chorda based on CH4 production and rumen fermentation parameters. The hot-air-dried G. chorda was added at 7 concentrations of 0, 0.625, 1.25, 2.5, 5, 10, and 20% OM in semi-closed in vitro batch cultures. Only treatment with the highest dosage mitigated gas production at all sampling times, whereas addition levels of 5, 10, and 20% OM significantly mitigated CH4 production throughout, exhibiting CH4-reducing rates of 18.87 to 31.36%, 25.68 to 52.47%, and 45.25 to 72.89%, respectively. Concurrently, CH4 proportion was also decreased by the same dosages at the end of the incubation. Rumen pH and acetate were increased with 10 and 20% OM addition of G. chorda, while no differences were observed in total volatile fatty acids and propionate. The acetate to propionate ratio was increased in the 20% OM treatment, and a notable reduction in butyrate was started from 5% OM inclusion. The highest level of G. chorda led to a decrease in branched-chain fatty acids (iso-butyrate and iso-valerate) and valerate. These results indicate that dietary supplementation of G. chorda with more than 5% OM might hold the potential as a natural CH4-reducing feed additive without detrimental impacts on rumen fermentation. The final study (Chapter 4) elucidated the effects of G. chorda on microbial communities, enzymes, and metabolic pathways through metagenomic analysis. Rumen fluid supplemented with 5% OM of hot-air-dried G. chorda was used for 16S rRNA sequencing. The treatment resulted in a reduction in the relative abundances of the genus Methanobrevibacter and CH4 metabolism. Notably, there were significant downregulations in several enzymes, such as cobU/cobT, cobS/cobV, cobP/cobU, cbiB/cobD, cobQ/cbiP, cobH/cbiC, cbiD, cbiG, cobM/cbiF, and cobJ/cbiH, all associated with porphyrin, especially cobalamin (vitamin B12), metabolism with a strong positive correlation with CH4 production. The relative abundances of propionate metabolism and gene K15024 were increased, along with upregulations of the genes acyP and K15024 involved in the acetate synthesis pathway. In the treatment group, the relative abundances of the family Lachnospiraceae and genus Butyrivibrio, as well as genes fadN, paaH/hbd/fadB/mmgB, and gcdH related to butyrate synthesis were decreased. This implies that secondary metabolites within G. chorda biomass might impede synthesis from cobalt to cobalamin, thereby reducing the activity of the cobalamin-dependent coenzyme M methyltransferase step in rumen methanogenesis. In summary, G. chorda was one of the promising red seaweed species, and hot-air-drying with a supplementation dosage of 5% OM was the most optimal feed form with the least effects on rumen fermentation while maintaining the CH4-reducing effectiveness. In addition, the metagenomic analysis alluded that the anti-methanogenic ability of G. chorda was due to the decrease in the genus Methanobrevibacter, and this might be attributed to the downregulation of cobalamin synthesis, which is an essential cofactor for coenzyme M methyltransferase in archaeal methanogenesis. Overall, G. chorda has the potential to be used as a natural CH4 reducer without negative impacts on rumen fermentation characteristics.

Concurrently, CH4 proportion was also decreased by the same dosages at the end of the incubation. Rumen pH and acetate were increased with 10 and 20% OM addition of G. chorda, while no differences were observed in total volatile fatty acids and propionate. The acetate to propionate ratio was increased in the 20% OM treatment, and a notable reduction in butyrate was started from 5% OM inclusion. The highest level of G. chorda led to a decrease in branched-chain fatty acids (iso-butyrate and iso-valerate) and valerate. In the treatment group, the relative abundances of the family Lachnospiraceae and genus Butyrivibrio, as well as genes fadN, paaH/hbd/fadB/mmgB, and gcdH related to butyrate synthesis were decreased. This implies that secondary metabolites within G. chorda biomass might impede synthesis from cobalt to cobalamin, thereby reducing the activity of the cobalamin-dependent coenzyme M methyltransferase step in rumen methanogenesis. In summary, G. chorda was one of the promising red seaweed species, and hot-air-drying with a supplementation dosage of 5% OM was the most optimal feed form with the least effects on rumen fermentation while maintaining the CH4-reducing effectiveness. In addition, the metagenomic analysis alluded that the anti-methanogenic ability of G. chorda was due to the decrease in the genus Methanobrevibacter, and this might be attributed to the downregulation of cobalamin synthesis, which is an essential cofactor for coenzyme M methyltransferase in archaeal methanogenesis. Overall, G. chorda has the potential to be used as a natural CH4 reducer without negative impacts on rumen fermentation characteristics.
