L-ascorbic acid (AA)의 2번 위치의 수산기에 부위 특이적 활성을 갖는 Paenibacillus sp. JB-13 유래의 cyclodextrin glucanotransferase (CGTase, EC 2.4.1.19) 유전자(cgt gene)를 pEXP7 발현 vector에 클로닝하여 재조합 균주를 구축하였다. 재조합 균주의 CGTase생산 최적 조건을 검토하여 본 결과 LB 배지에 0.5% glucose, 3.0% polypeptone, 0.3% $K_2HPO_4$, 0.5% NaCl이 되도록 추가하고, 초기 pH 7.0, 접종량 2%, 통기량 0.1 vvm, 배양 온도 $37^{\circ}C$, 배양 시간 14시간의 조건에서 최대 활성을 나타내었다. 재조합 균주와 기존 균주의 CGTase 활성을 비교한 결과 재조합 균주는 배양 14시간째 640 units/ml의 활성을 가져 기존 균주에 비해 70%의 활성을 나타내지만 배양 시간을 1/4로 단축시킬 수 있다는 이점이 있음을 확인하였다. 재조합 균주가 생산한 CGTase를AA-2G합성에 적용하여 AA-2G를 합성하고 HPLC로 분석한 결과 단일 peak를 확인할 수 있었고 ${\alpha}$-glucosidase를 처리하여 확인한 결과 AA와 glucose로 분리됨을 확인할 수 있었다.

The purpose of this study is to clone cgt gene from Paenibacillus sp. JB-13 and to overexpress the protein in E. coli. For this purpose, the cgt gene was amplified from Paenibacillus sp. JB-13 genomic DNA by PCR using degenerate oligonucleotide primers. The sequence analysis results showed that the cgt gene from Paenibacillus sp. JB-13 has 98% homology with the cgt gene of Bacillus sp. To overexpress the protein, the cgt gene was cloned into pEXP7 expression vector and transformed into E. coli. The production of CGTase by recombinant E. coli was optimized under following conditions: 0.5% glucose, 3.0% polypeptone, 0.3% $K_2HPO_4$, 0.5% NaCl, and 7.0 of initial pH, 2.0% of inoculum, $37^{\circ}C$ of culture temperature for 14 hr. And the optimal agitation was found at 0.1 vvm. The synthesis of 2-O-${\alpha}$-D-Glucopyranosyl L-Ascorbic acid (AA-2G) using the CGTase expressed in E. coli was identified as AA-2G by HPLC and HPLC confirmed that treating AA-2G made by cloned CGTase with ${\alpha}$-glucosidase substantially produced AA and glucose.

PstI, Sall 제한효소를 사용하여 cgt gene을 발현 vector인 pEXP7 vector (13)에 클로닝하고 E. coli DH5a (Promega, US A) 에 형질전환하여 (8) CGTase를 생산하는 재조합 E. coli 균주를 구축하였다.
구축된 재조합 E. coli 균주를 설정된 최적 조건에서 진탕 배양한 후 얻어진 배양액을 4℃에서 원심분리(15,000Xg, 20 min) 한후 상등액을 50% (NHD2SO4로 염석하고 투석하여 AA-2G 생산을 위한 조효소액으로 사용하였다. 재조합 균주인 E.
기본 배지인 LB 배지에 각종 탄소원을 1.0% 첨가한 후 37℃ 에서 진탕 배양하여 전술한 방법에 따라 효소 활성 및 균 생육 도를 측정하였다. 그 결과 탄소원으로는 glucose를 사용하였을 때 가장 높은 활성을 나타내었다(Table 1).

이론/모형
coli 균주 그리고 분리 균주인 Paenibacillus sp. JB-13을 각각 배양하여 다음과 같은 방법으로 CGTase 활성을 측정하였다.

5%까지 첨가하여 측정하였다. 그 결과 0.5%일 때 가장 높은 활성을 나타내었으나 0.3%에서 0.5%까지의 차이가 미미하므로 0.3%를 최적 농도로 결정하였다(Fig. 1C).

특히 AA-2G는 AA와 달리 Fe2+ 혹은 C* iF의 존재 하에서도 안정하며 ascorbate oxidase에 대해서도 저항성을 갖는다(23). 또한 AA에 비해서 내열성과 내광성(耐光性)이 크게 증대되어 100℃, 30분 동안의 가열에도 분해되지 않고 또한 직접적인 환원 활성을 가지고 있지는 않지만 세포라든지 생체에 투여되면 그곳에 존재하는 a-glucosidase에 의해서 환원 형 AA와 glucose로 분해되어 AA와 마찬가지로 항 괴혈병 작용(24)이라든지 collagen의 합성 촉진(22), 백내장 예방 작용, 면역 촉진 작용(25) 등의 생리 활성을 발휘하며 AA에서는 발견되지 않았던 항체 생산 증강 작용이 있다는 것도 밝혀졌다. 뿐만 아니라 AA-2G는 중성 용액 중에서 안정하기 때문에 주사제의 조제 및 세포 내에서의 AA의 생리적 기능 연구도 가능하다.
3). 또한 생산된 AA-2G 반응 산물 20 ㎖에 1 unit a-glucosidase를 처리하여 AA와 glucose로 분리됨을 확인할 수 있었다. 이는 AA-2G가 생체 내에 존재하는 a-glucosidase에 의해 AA와 glucose로 분리되어 AA의 생리적 기능을 나타냄을 반증하는 결과이다.
coli 균주를 접종하여 37P에서 9시간 진탕배양하였다. 배양액을 원심분리하여 상등액과 균체로 분리하여 CGTase 활성 측정 결과 상등액에서는 활성이 나타나지 않았으며 균체 파쇄액에 활성이 확인되어 본 균주가 생산하는 CGTase는 세포 내 효소로 판단되었다.
재조합 균주에서 정제된 CCTase를 AA-2G 생산 조건에 적용하여 AA-2G를 생산하고 HPLC를 사용하여 분석한 결과 AA-2G 표준품과 합성된 AA-2G는 동일한 peak를 나타내었다(Fig. 3). 또한 생산된 AA-2G 반응 산물 20 ㎖에 1 unit a-glucosidase를 처리하여 AA와 glucose로 분리됨을 확인할 수 있었다.

확보된 cgt gene의 염기서열은 GenBank (NCBI; http://www. ncbi.nlm.nih.gov)에 수록되어 있는 CGTase 생산 Bacillus 속 균주의 cgt gene과 최대 98%의 상동성을 나타내었다(결과 미제시).
후속연구
재조합 균주와 기존 균주의 CGTase 활성을 비교한 결과 재조합 균주는 배양14시간째 640 urms/ml의 활성을 가져 기존 균주에 비해 70%의 활성을 나타내었다. 확보된 cgt gene을 directed evolution 전략 (10)으로 추가적인 연구를 통해 그 기능을 개선할 수 있는 기초를 마련하였다고 사료된다.
