이론/모형
Cloning host로써 Novagen에서 구입한 Escherichia coli DH5α와 BL21(DE3)을 Luria-Bertani (LB) broth (Difco, USA)에서 37℃로 배양하였으며, pET-22b(+)를 cloning vector로 사용하였다. Yeast (Pichia angophorae CBS5830와 Saccharomyces cerevisiae (JENICO, Korea))를 alcohol 발효 균주로써 150 rpm, 30℃ 조건 하에서 Horn 등[14]의 방법을 사용하여 배양 하였으며, 에탄올 생산에 사용한 배지는 0.02% urea와 0.006% NaH2PO4를 basal medium으로 하였으며 배지의 pH는 Mcilvaine [19]의 방법에 따라 조정하였다. 그리고 각각의 균주는 glycerol을 이용하여 -80℃에서 보존하였다.
Adams로 부터 받아 사용하였으며, endo-1,3-β-glucanase (accession number: AF013169)의 증폭을 위한 주형으로 사용하였다. 모든 분자생물학적 실험방법은 Sambrook 등[24]의 방법을 따랐다. Glucanase의 ORF를 각각 NdeI과 XhoI site가 존재하는 forward primer (5’-GGCCCATATGA AAAAAGAAGCCTTGTTGTTTC-3’)와 reverse primer (5’-GGCCCTCGAGACCACTAACGAATGAGTAAAC-3’)를 사용하여 PCR로 증폭하였다.

성능/효과
Fig. 1에 나타낸 것처럼 host E. coli의 배양과정 중 IPTG를 이용한 대량유도 발현이 성공적임을 확인하였으며, 약 33 kDa의 대량발현 된 재조합 β-1,3-glucanase는 대부분 inclusion body의 형태로 cytoplasm내에서 존재함을 확인하였다.
HPLC를 사용한 분석에서도 TLC의 결과와 마찬가지로 standard 물질로 주입한 glucose와 retention time이 같은 peak가 존재하였으며, 다른 retention 시간에서 나타나는 peak들이 있는 것으로 보아 고분자의 laminarin이 glucose 및 glucose 이외의 올리고당으로 분해 되었음을 알 수 있었다(Fig. 3).
Laminarin 분해산물을 유일한 C-source로써 P. angophorae를 접종한 후 배양 과정 중 생산된 에탄올은 laminarin분해 반응에 첨가된 laminarin의 농도에 따라서 생산량이 증가하였다(Table 1). 효소반응을 위해 첨가된 laminarin의 농도가 0.
따라서 보다 많은 에탄올을 생산하기 위해서는 laminarin과 β-1,3-glucanase의 반응 시간, 최적 조건 및 기질과 효소의 첨가 비율 등과 같은 반응 조건을 확립하여 P. angophorae 균주가 에탄올 생산에 사용할 수 있는 glucose를 최대한 많이 laminarin으로부터 생산함이 중요하다는 것을 확인할 수 있었다.
따라서 본 연구에서는 미생물을 alcohol 발효에 사용할 수 있으며, 반응에 첨가되는 유해물질이 없는 방법을 위하여, 해조류 유래의 laminarin이라는 다당을 사용하여 β-1,3-glucanase과 반응하여 분해 산물이 alcohol 발효에 이용할 수 있는지를 확인한 결과 재조합 β-1,3-glucanase는 laminarin을 glucose 수준의 단당을 부분적으로 생산하였으며, 이러한 분해 단당을 이용한 alcohol 발효과정을 통하여 bio-ethanol을 생산할 수 있는 가능성이 있음을 증명하였다.
배양 후 gas chromatography로 확인한 결과 S. cerevisiae에서는 β-1,3-glucanase에 의한 laminarin의 분해 산물을 C-source로써 사용하여 생산한 에탄올의 함량이 배지 성분내에 매우 소량이었으며, P. angophorae의 경우 S. cerevisiae보다는 많은 양의 에탄올을 생산함을 알 수 있었다(data not shown).
그러나 48시간 이후에는 생산량의 변화에 차이를 보이지 않았다(Table 1). 이와 같은 결과는 laminarin의 분해 농도가 높을수록 P. angophorae가 알코올 발효반응에 사용할 수 있는 분해되어 생산되는 glucose의 함량이 많다는 것을 알 수가 있었다. 하지만 동일한 비율의 laminarin 분해산물과 glucose를 이용하여 각각의 에탄올 생산량을 비교하였을 때에는 glucose보다는 현저하게 낮은 수율의 에탄올 생산량을 laminarin 분해물에서 나타났다.
이와 같은 결과는 laminarin이 β-1,3-glucanase에 의해 생산되는 효모 이용성 glucose의 농도가 비교대상의 glucose의 농도보다 낮음을 알 수 있었으며, TLC 실험 및 HPLC 분석에서도 보여주고 있듯이 laminarin 분해 산물에는 glucose이외의 분해 다당 또는 단당류가 존재함을 알 수가 있었다(Table 1).
전개용매를 따라 이동한 spot들의 위치를 분석한 결과, 정제한 β-1,3-glucanase이 첨가되지 않고 liminarin만을 첨가한 control 용액의 spot은 전개용매에 의한 이동이 거의 없었으나, β-1,3-glucanase와 반응한 laminarin 용액은 다양한 종류의 저분자 올리고당 또는 단당의 형태로 분해됨을 알 수 있었으며 glucose의 Rf치와 동일한 spot도 존재하고 있었다(Fig. 2).

해조류 유래의 다당류는 이러한 공정으로써 glucose 수준의 단당 형태의 저단위체로의 분해가 어려우며 또한 분해된 단당류를 이용한 alcohol 발효 효율이 매우 낮다는 문제가 있다[3,16]. 따라서, 해조류 유래의 다당류를 효과적으로, 그리고 alcohol 발효가 용이한 형태의 단당체 형태로의 분해 기술의 개발을 위한 연구가 필요하다.
