제안 방법
MB에서 확인된 아밀라아제 생산량을 증가시키기 위하여 물리적 요인에 변화를 주어 RSM을 진행하였다. 물리적 요인들과 아밀라아제 생산의 상관관계를 확인하기 위하여 물리적 요인으로는 접종원의 부피(inoculum volume), 수소이온농도, 배지의 부피(medium volume)를 선정하였으며, 선정된 3가지 물리적 요인들을 독립변수로 하여 Minitab 16을 통한 중심합성계획법을 설계한 후, 조건이 각기 다른 20개의 실험군을 각각 3번의 반복실험을 거쳐 평균적인 아밀라아제 생산량을 확인하였다(Table 1).
이후 4 °C에서 냉각하여 ELISA Reader를 이용하여 575 nm에서 흡광도를 측정하였고, 멸균된 MB를 첨가한 혼합액을 blank로 사용하였다. 또한, 사전에 측정된 0.2% 말토오스(maltose)를 이용한 표준곡선(standard curve)을 바탕으로 확인된 광학밀도(optical density, OD)를 대입하여 환원당의 양을 계산하였다(data not show).

미생물의 세포농도는 ELISA Reader (Molecular Devices, VersaMax™ and SpectraMax® 340PC384, Menlo Park, CA)를 이용하여 600 nm에서 흡광도를 측정하였다.

대상 데이터
