나노 침전법은 무엇인가? | Fessi의 연구팀(Fessi et al., 1989, 1992)에 의해 최초로 개발된 나노 침전법은 다양한 유기 용매에 고분자와 유용 약물을 용해시킨 후 이를 비용매인 물에 적하⋅분산시켜서 나노입자를 얻는 방법으로, 두 용매는 혼화될 수 있어야 하지만 고분자와 유용 약물은 비용매에서 용해되지 않아야 하는 것으로 알려져 있다[11]. 유화/용매 확산법과는 대조적으로, 독성 효과를 일으키거나 표면 특성에 영향을 미치는 첨가제 및 높은 온도, 장시간의 초음파처리 공정이 필요하지 않고, 모든 절차가 단일 공정으로 수행되며, 좁은 단분산 분포를 갖는 나노입자의 생산이 가능하다는 이점이 있다. | |
캡슐화는 유용 약물의 어떤 단점을 보완하기 위한 방법인가? | 천연 추출물(Natural extracts), 약물, 비타민, 항산화 물질(Anti-oxidant) 등과 같은 유용 약물은 일반적으로 빛, 산소, 수분, 온도 등의 외부요인으로부터 영향을 받아 변질되거나 쉽게 파괴될 뿐만 아니라 가공 및 유통과정 동안 안정성이 저하되어 활성이 감소하게 된다. 또한 유용 약물은 직접 섭취 시 낮은 투과성(Permeability)과 위장(Gastrointestinal track) 내의 산성조건, 효소(Enzyme) 등 다양한 요인들에 의하여 안정성이 감소하게 되어 이들이 보유한 생리활성에 제한을 주게 된다[3-4]. 이러한 유용 약물의 보호와 저장 안정성을 향상시키기 위한 방법으로 캡슐화(Encapsulation)가 가장 일반적인 방법으로 알려져 있으며[5-6], 캡슐화를 통하여 화학적, 생물학적 반응을 최소화하는 물리적 장벽을 제공해 줌으로써 외부 환경으로부터 보호하고 섭취 후 생체 내 원하는 곳에서 흡수, 이용될 때까지 활성이 유지될 수 있다. |
