Bei diesem Versuch wurde der Einfluss von 2.4-D, von deren Propylenglycolbutylattherester (PGBA) und deren Isooctylester (IO) auf die foetale Entwicklung und auf das neonatale Wachstum und Uberleben untersucht. Dosierungskonzentrationen von 2.4-D bis zu einer tolerierten Maximaldosis von 87.5 mg/kg/Tag oder molare Aquivalente von PGBA oder IO wurden an trachtige Sprague-Dawley-Ratten vom 6.-15. Tag der Trachtigkeit verabreicht. Am 20. Tag der Trachtigkeit wurden die Jungen dutch Kaiserschnitt zur Welt gebracht und makroskopisch untersucht, gemessen und gewogen. Nach routinemassigen Vorbereitungen wurden die weichen Gewebe und Skelette untersucht. Anzeichen der Embryotoxizitat und Foetotoxizitat wie vermindertes Korpergewicht des Foetus, subcutanes Odem, verzogerte Ossification von Knochen, Lumbalrippen und Costae fluctuans wurden bei hohen Dosierungen beobachtet; insgesamt genommen standen die Reaktionen in Beziehung zur Dosierung. Teratogene Wirkungen wurden jedoch bei keiner Dosierung beobachtet. 2.4-D beeinflusste nicht Fruchtbarkeit, Trachtigkeitsdauer, Lebensfahigkeit oder Laktation. PGBA und IO hatten keinen Einfluss auf Fruchtbarkeit und Trachtigkeitsindizes, aber die hochsten Dosierungen verminderten die Lebensfahigkeit und setzten die Laktationsindizes herab. Neonatales Wachstum und Entwicklung wurden dutch Verabreichung wahrend der Trachtigkeit nicht beeintrachtigt.
On a evalue les effets du 2.4-D, de l'ester propylene-glycol-butyl-ether du 2.4-D (PGBE) et de l'ester isooctylique du 2.4-D (IO) sur le developpement foetal et sur la croissance et la survie neonatales. Des doses de 2.4-D allant jusqu'au maximum tolere de 87.5 mg/kg/jour ou des doses equivalentes, en molecules-grammes, de PGBE ou d'IO ont ete administrees a des femelles gravities de rats Sprague-Dawley entre le 6e et le 15e jour de grossesse. Les foetus extraits par cesarienne le 20e jour de la grossesse ont fait l'objet d'un examen general et ont ete mesures et peses. On a examine les tissus mous et les squelettes apres preparation de routine. Des indices de toxicite embryonnaire et foetale, par exemple une insuffisance de poids du foetus, des oedemes sous-cutanes et un retard d'ossification des os, des cotes lombaires et des cotes flottantes, ont ete observes dans les groups qui avaient recu une dose elevee; consideres dans l'ensemble, ces indices etaient en correlation avec le dosage. Aucun effet teratogene n'a toutefois ete constate, quelle qu'ait ete la dose administree. Le 2.4-D n'a pas influe sur la fecondite, la gestation, la viabilite ou la lactation. Le PGBE et l'IO n'ont pas influe sur les indices de fecondite et de gestation, mais les doses les plus elevees ont fait baisser les indices de viabilit6 et de lactation. La croissance et le developpement neonatals n'ont pas ete alteres par le traitement administre pendant la grossesse.
