A process is disclosed for reducing or removing endotoxins from compositions containing therapeutic active substances extracted from natural sources by genetic engineering and/or biotechnology. For that purpose, the compositions are treated with chromatographic materials. The natural sources are disintegrated, the thus obtained fractions are, if required, centrifuged, filtered or treated using affinity chromatography methods, the fractions are pre-incubated in an aqueous salt solution and detergents, are treated with anion exchange materials, then washed with another salt solution. The active substances are eluted from the anion exchanger then further purified in a manner known per se.

[ We claim:] [1.] A process for the depletion or removal of endotoxins from preparations containing active ingredients designated for therapeutic use which are obtained from natural sources by genetic engineering and/or biotechnology by treatment with chromatographic material, comprising the sequential steps of:lysing said natural sources and, optionally, centrifugation of the lysed natural sources, filtration of the lysed natural sources, or subjecting the lysed natural sources to affinity chromatography, in order to remove coarse cell debris, to effect lysed material;incubating said lysed material with an aqueous salt solution and detergents to effect a separation;subjecting the separation to anion exchange chromatography by applying the preparation to an anion exchange material, washing the anion exchange material with an aqueous salt solution, different from the aqueous solution with which the lysed material was incubated, and eluting from the anion exchange material said active ingredients;purifying the eluted active ingredients.
