Two or more hydrophilic polymers that are not soluble in each other at a particular concentration and temperature, but which have a positive spreading coefficient in solution, are used to form multi-layered polymeric microspheres. The multi-layer microspheres produced by the method are distinguished by extremely uniform dimensioned polymer layers and actual incorporation of a substance to be delivered into the polymer layers. In the preferred embodiment of the method, two polymers are dissolved in an aqueous solvent, the substance to be incorporated is dispersed or dissolved in the polymer solution, the mixture is suspended in an organic solvent or polymer/water mixture and stirred, and the solvent is slowly evaporated, creating microspheres with an inner core formed by one polymer and an outer layer formed by the second polymer.

[ What is claimed is:] [1.] A method for forming multi-layered polymeric microspheres of hydrophilic, water soluble polymers, the method comprising:(i) selecting first and second hydrophilic, water soluble polymers;(ii) dissolving the first hydrophilic, water soluble polymer in a first aqueous solution comprising solvent for the first polymer;(iii) dissolving the second hydrophilic, water soluble polymer in a second aqueous solution comprising solvent for the second polymer;(iv) mixing the hydrophilic, water soluble polymer solutions;(v) combining the mixed hydrophilic, water soluble polymer solution with a third solution selected from the group consisting of organic oil, organic solvent, and combinations thereof, in which the first and second hydrophilic, water soluble polymers are not soluble, thereby to cause phase separation of the hydrophilic, water soluble polymers, and to allow the surface tension and the interfacial tension of the hydrophilic, water soluble polymers to cause the formation of microspheres, each microsphere including an inner core of the first hydrophilic, water soluble polymer, and at least one distinct outer layer of the second hydrophilic, water soluble polymer surrounding the first hydrophilic, water soluble polymer core; and(vi) evaporating solvent from the combined mixture to promote phase separation of the polymers and formation of the microspheres. [6.] The method of claim 1 or claim 5 wherein a substance to be incorporated into the microspheres is added to at least the first hydrophilic, water soluble polymer solution in step (ii), and wherein the substance is incorporated into at least the first hydrophilic, water soluble polymer in the microsphere in step (v). [9.] The method of claim 6 wherein the substance to be incorporated is biologically active. The method of claim 1 or claim 5 further comprising forming additional polymer layers around the microspheres. The method of claim 1 or claim 5 wherein the first and second hydrophilic water soluble polymers are selected from the group consisting of polyamines, natural and synthetic polysaccharides, poly(oxalkylene)oxides, polyacrylamides and synthetic copolymers of hydrophilic and hydrophobic polymers. The method of claim 1 or claim 5 wherein the first and second hydrophilic, water soluble polymers each are biodegradable polyelectrolytes. The method of claim 1 or claim 5 wherein at least one of the first and second solutions includes water and a polar solvent. The method of claim 1 or claim 5 wherein the first and second solutions are identical, and wherein steps (i)-(iv) comprise mixing the first and second hydrophilic, water soluble polymers in a single solution. The method of claim 1 or claim 5 further comprising:(vii) cooling the combined mixture, thereby to gel the first and second hydrophilic, water soluble polymers within the microspheres.
