The present invention relates to the identification of a novel metalloprotease in gram positive microorganisms. The present invention provides the nucleic acid and amino acid sequences for the metalloprotease. The present invention also provides host cells having a mutation or deletion of part or all of the gene encoding the metalloprotease. The present invention provides host cells which further comprises a nucleic acid encoding desired heterologous proteins such as enzymes. The present invention also provides cleaning compositions, animal feeds and compositions used to treat a textile that include the metalloprotease of the present invention.

A genetically engineered gram positive microorganism having a non-reverting mutation or deletion of part or all of the nucleic acid encoding a metalloprotease, said non-reverting mutation or deletion resulting in the inactivation of the metalloprotease proteolytic activity, wherein the nucleic acid of the metalloprotease comprises the sequence shown in SEQ ID NO: 1 prior to mutation or deletion. 2. The gram positive microorganism according to claim 1 wherein said microorganism is a member of the family Bacillus. 3. The microorganism according to claim 2 wherein said Bacillus is selected from the group consisting of B. subtilis, B. licheniformis, B. lentus, B. brevis, B. stearothermophilus, B. alkalophilus, B. amyloliquefaciens, B. coagulans, B. circulans, B. lautus and B. thuringiensis. 4. The microorganism of claim 3 wherein said microorganism comprises nucleic acid encoding a heterologous protein. 5. The microorganism of claim 3 wherein said microorganism comprises nucleic acid encoding a homologous protein. 6. The microorganism of claim 4 wherein said heterologous protein is selected from the group consisting of hormones, enzymes, growth factors and cytokines. 7. The microorganism of claim 6 wherein said heterologous protein is an enzyme. 8. The microorganism of claim 7 wherein said enzyme is selected from the group consisting of a protease, a carbohydrase, a lipase, an isomerase, an oxidase, a reductase, a transferase, a kinase and a phosphatase. 9. A genetically engineered Bacillus subtilis having a mutation or deletion of part or all of the nucleic acid encoding a metalloprotease wherein the nucleic acid prior to mutation or deletion encoded the metalloprotease having the sequence shown in SEQ ID NO: 2, and wherein said mutation or deletion is non-reverting and results in the inactivation of the metalloprotease proteolytic activity. 10. The Bacillus subtilis of claim 9, wherein said Bacillus has been further engineered to comprise a heterologous enzyme. 11. The Bacillus subtilis of claim 10, wherein the enzyme is selected from the group consisting of proteases, amylases, cellulases and lipases. 12. The Bacillus subtilis of claim 9, wherein the nucleic acid encoding the metalloprotease has been deleted from the Bacillus chromosome. 13. A genetically engineered Bacillus comprising a mutation or deletion in a metalloprotease gene which results in the inactivation of the production of a proteolytically active metalloprotease having the amino acid sequence of SEQ ID NO.2 or a proteolytically active metalloprotease having an amino acid sequence with 95% sequence identity to SEQ ID NO. 2. 14. The genetically engineered Bacillus of claim 13 further comprising a nucleic acid encoding a heterologous protein. 15. The genetically engineered Bacillus of claim 14, wherein the heterologous protein is an enzyme. 16. The genetically engineered Bacillus of claim 13, wherein the deletion is in the coding region of said gene. 17. The genetically engineered Bacillus of claim 13 further comprising a mutation or deletion in a gene selected from the group consisting of apr, npr, epr and mpr. 18. The genetically engineered Bacillus of claim 13, wherein said Bacillus is selected from the group consisting of B. subtilis, B. licheniformis, B. lentus, B. stearothermophilus, and B. amyloliquefaciens.
