A separator that uses centrifugation to fractionate a suspension such as blood comprises a separation container and a buoy. The buoy is carried in the separation container and has a tuned density that is configured to reach an equilibrium position in a suspension. The guide surface is carried on the buoy upper surface and is inclined to an accumulation position near a buoy perimeter. The buoy suspension fractionation system can be used in a method of isolating a fraction from a suspension, and in a method for re-suspending particulates for withdrawal.

1. A method for separating a material with a buoy fractionation system, comprising: placing the material in a container having:a first buoy portion defining a first height and having a first side in physical contact with the wall of the container and defining a first apex above which the material is initially received,a second buoy portion defining a second height and having a second side and defining a second apex, wherein the second apex of the second buoy portion is spaced a fixed distance apart from the first apex of the first buoy portion, anda third buoy portion extending from the second apex of the second buoy portion to the first buoy portion interconnecting the first buoy portion and the second buoy portion and maintaining the spacing of the first buoy portion and the second buoy portion at the distance; andapplying a centrifugal force to the material to fractionate the material and push a selected fraction of the material toward the first buoy portion such that the selected fraction of the material is received within a volume between the first buoy portion and the second buoy portion, wherein the first height of the first buoy portion is substantially smaller than the second height of the second buoy portion. 2. The method of claim 1, further comprising: withdrawing at least the selected fraction from between the first buoy portion and the second buoy portion through a withdrawal port near the first apex that is in fluid communication with a withdrawal passage formed through the third buoy portion and an inlet port from the volume between the first buoy portion and the second buoy portion. 3. The method of claim 2, wherein withdrawing at least the selected fraction from between the first buoy portion and the second buoy portion further comprises withdrawing through a buoy withdrawal passage defined through at least a portion of the second buoy portion that is in communication with the withdrawal port through the withdrawal passage and the inlet port that is defined through a collection surface in the second buoy portion. 4. The method of claim 3, further comprising: placing a separation container containing the first buoy portion, the second buoy portion, and the third buoy portion in a centrifuge device;wherein applying a centrifugal force includes operating the centrifuge device with the separation container placed therein. 5. The method of claim 2, wherein withdrawing the at least selected fraction concentrates particulates of the material at an accumulation position. 6. The method of claim 5, further comprising: conveying the particulates along the first side of the first buoy portion to between the first buoy portion and the second buoy portion. 7. The method of claim 6, further comprising: conveying the particulates along the second side of the second buoy portion from between the first buoy portion and the second buoy portion. 8. The method of claim 5, wherein concentrating particulates includes concentrating at least one particulate from a platelet, a stromal cell, white blood cells, or combinations thereof. 9. The method of claim 5, further comprising: agitating the concentrated particulates to at least partially re-suspend the concentrated particulates within a portion of the selected fraction prior to withdrawing the portion of the selected fraction containing the re-suspended concentrated particulates. 10. The method of claim 9, further comprising: forming a bubble between the first buoy portion and the second buoy portion;wherein agitating includes moving the container with the formed bubble to at least partially re-suspend the concentrated particulates. 11. The method of claim 1, wherein the material placed in the container comprises a volume of whole blood. 12. The method of claim 1, wherein the material comprises multiple components; wherein applying the centrifugal force to the material causes the component to separate into at least two fractions of varying densities. 13. The method of claim 12, further comprising: allowing the first buoy portion and the second buoy portion to reach an equilibrium within the fractions; andpositioning at least a portion of one of the two fractions within a collection space between the first buoy portion and the second buoy portion. 14. The method of claim 1, wherein the first side is inclined substantially towards a single portion of the perimeter of the first buoy portion. 15. The method of claim 1, wherein the second side is inclined substantially towards a single portion of the perimeter of the second buoy portion.
