A closure device for sealing a percutaneous puncture in a wall of a body passageway is disclosed, the closure device including a plug configured to engage a surface of the puncture; a toggle configured to engage an internal surface of the body passageway; an elongate filament configured to associate the plug with the toggle; a locking member configured to engage the plug; and a tamper comprising at least two lumens, wherein a first lumen of the at least two lumens is configured to deliver a pharmaceutical agent to the plug.

The closure device of claim 1, wherein the tamper is a cylindrical tamper tube. 3. The closure device of claim 1, wherein at least one of the at least two tamper lumens is a channel that runs along a portion of a surface of the tamper. 4. The closure device of claim 1, wherein a third lumen of the at least two tamper lumens is configured to deliver a pharmaceutical agent to the plug. 5. The closure device of claim 1, wherein the tamper comprises four or more lumens. 6. The closure device of claim 1, wherein the plug is a resorbable collagen sponge or foam. 7. The closure device of claim 1, wherein the tamper comprises an access opening that is fluidically coupled to one of said lumens. 8. The closure device of claim 4, further comprising a drug delivery member to deliver the pharmaceutical agent to the third lumen. 9. The closure device of claim 8, further comprising an extension tube connected to an access opening through which the drug delivery member may deliver the pharmaceutical agent to the first lumen. 10. The closure device of claim 1, wherein the first lumen and the second lumen of the tamper are substantially parallel to each other. 11. The closure of device of claim 1, wherein one of the at least two tamper lumens is configured to deliver a pharmaceutical agent to a desired location at the puncture. 12. The closure device of claim 11, wherein one of the at least two tamper lumens is configured to deliver a pharmaceutical agent to a desired location on the plug. 13. The closure device of claim 1, wherein the tamper engages the locking member at the distal end of the tamper such that the tamper causes the locking member to engage with the plug so as to seal the puncture. 14. The closure device of claim 1, wherein the elongate filament associated with the plug, the toggle, the locking member, and the tamper. 15. The closure device of claim 1, wherein the tamper defines a length that extends from the proximal end to the distal end, wherein the length is such that an entirety of the tamper is removable from the deployment body to tamp the locking member against the plug. 16. A closure device for sealing a percutaneous puncture in a wall of a body passageway, the closure device comprising: a plug configured to engage a surface of the puncture;a toggle configured to engage an internal surface of the body passageway;an elongate filament configured to associate the plug with the toggle;a locking member configured to engage the plug;a tamper having a) a first lumen that is configured to receive a guidewire therethrough, b) a second lumen that the elongate filament passes through, and c) a third lumen that is configured to deliver a pharmaceutical agent to the plug; anda drug delivery member to deliver the pharmaceutical agent to the third lumen. 17. The closure device of claim 16, wherein the tamper is a cylindrical tamper tube. 18. The closure device of claim 16, wherein at least one of the lumens is a channel that runs along a portion of the surface of the tamper. 19. The closure device of claim 16, wherein the plug is a resorbable collagen sponge or foam. 20. The closure device of claim 16, further comprising an extension tube connected to an access opening through which the drug delivery member may deliver the pharmaceutical agent to the first lumen. 21. The closure of device of claim 16, wherein the tamper engages the locking member at the distal end of the tamper such that the tamper causes the locking member to engage with the plug so as to seal the puncture. 22.
