AHP에 의한 탈리그닌 조건은 25℃에서 100시간 동안 H?O?(pH 11.5)로 반응시킨 것이 당화효율과 분해율이 가장 높았다. 이 조건하에서 생성된 당은 주로 glucose 및 xylose로 구성돼 있으며, 당화율과 분해율은 무처리재에 비해 각각 260%와 350%의 증가를 나타냈으며 이 같은 조건은 1% NaOH와 20% 과초산으로 처리한 목분의 그것과 대비될만한 것이었다. 탈리그닌 현사시 목분자화 우수균주로서 Aspergillus fumigatus KC-1을 선발하였다. 본균의 탈리그닌 현사시목분 1%를 탄소원으로하여 45℃에서 진탕배양시 4-5일째 섬유성물질 분해 효소생성이 최고치에 달했다. 본균의 효소에 의한 현사시 목분의 가수분해시 1% NaOH와 20% 과초산으로 탈리그닌한 목분의 가수분해율이 가장 높았고 최종산물로 glucose와 약간의 cellobiose 및 xylose가 검출되었다. A. fumigatus KC-1을 이용하여 1% 탈리그닌 현사시 목분을 함유한 배지로 45℃에서 3일간 진탕배양한 결과 건조중량으로서 4.1g/l의 균체를 얻었고, 목분당화액 (glucose 및 xylose) 함유배지로 Candida lusitaniae IFO 1019를 30℃에서 60시간 진탕배양하여 건조중량으로서 5.6g/l의 균체를 얻었다. 이들의 단백질 함량은 각각 33.1% 및 42.5%로서 대두박에 필적하였고 아미노산조성에 있어서는 lysine 함량이 특히 높을뿐더러 각종 아미노산 함량도 균형있게 조성돼 있었다. AHP 처리가 in sacco 반추위 소화율이 미치는 영향을 검토한 결과 현사시목분에 AHP 처리를 함으로서 40-60%의 lignin을 제거할 수 있었고 결과적으로 neutral detergent fiber (NDF), acid detergent fiber (ADF) 및 cellulose의 함량을 상대적으로 증가시켰고, 반추위 고형물, NDF, ADF 및 cellulose 소화율도 유의적 (P〈0.05)으로 증가하였다. 반추위 내용물의 outflow rate를 0.05/h로 가정하여 effective degradibility를 구한 결과 무처리 목분과 처리 목분들간에 현저한 (P〈0.05) 차이가 인정되었다. AHP 처리 사료를 급여한 면양의 일당증체량 및 사료 요구율이 무처리사료를 급여한 면양에 비해 유의적 (P〈0.05)으로 증가하였다. AHP 처리 사료의 고형물과 단백질의 가용성획분 (a) 및 분해가능획분 (b) 함량이 증가하는 듯한 경향을 보였으며 획분(b)의 분해율(k)과 lag time (1)에서는 유의적 (P〈0.05) 으로 개선되었다. Mobile nylon bag 을 이용한 유우 전체 소화관의 소화율에서는 AHP 처리사료의 조단백질 소화율이 약간 높은 수치를 나타냈다.

For the evaluation of woody waste as the possible foodstuffs, the sawdust of aspen delignifed with alkaline hydrogen peroxide (AHP) were hydrolyzed with cellulolytic enzymes and the single cell protein was produced with Candida lusitaniae. Finally runimal digestibility in sacco and nutritive value of AHP-treated aspen have been also examined. Approximately one-half of aspen lignin and most of the hemicellulose present in wood were removed when the wood sawdust was reacted at 25? for 100 hrs in alkaline solution(pH 11.5) of 1% peroxide (AHP). The rate of decomposition as well as the saccharification efficiency were enhanced up to 350% and 260% respectively in comparison with those of controll. This enhancement is comparable with that pretreated with 1% sodium hydroxide and 20% peracetic acid successively. Aspergillus fumigatus KC-1 was selected as the most active producer of cellulolytic activities among the 256 strains of cellulose-decomposing microorganisms screened in our laboratory. All the examined cellulolytic activites in a culture filtrate of A. fumigatus KC-1 grown on 1% aspen sawdust pretreated with AHP reached a maximum within 4-5 days. The sawdust of wood delignified with 1% NaOH and 20% peracetic acid succesively recorded the highest hydrolysis rate in the tests of enzymatic saccharification. The major end product of hydrolysis of aspen wood with the cellulolytic enzymes obtained from A. fumigatus KC-1 was glucose with small amount of cellobiose and xylose. A. fumigatus KC-1 was cultured at 45? for 3 days in the medium containing 1% delignifed aspen wood and 4.1g/l cell mass was harvested. For the production & SPC, Candida lusitaniae IFO 1019 was cultured in the medium containing enzymatic hydrolysates of sawdust at 30?, 60hrs. 5.6g/l of cell mass was obtained. Protein content in these cell masses were 33.1% and 42.5% respectively. These values could be comparable to that of soybean meal. Various kinds of amino acid were present in these single cell proteins, especially the highest amount of lysine was marked. The object of the current research was to evaluate the influence of alkaline hydrogen peroxide (AHP) treatment on in sacco digestion of aspen. Three non-lacting ruminally cannulated Holstein cows were fed a diet containing of 90% forage and 10% concentrate on a dry basis. The AHP treatment delignified 40-60% of lignin in aspen, resulting in increased neutral detergent fiber (NDF), acid detergent fiber (ADF) and cellulose. Disappearance rates of dry matter (DM), NDF, ADF and cellulose of aspen significantly (P<0.05) increased with AHP treatment from 12h incubation. The effective degradability of DM (EDDM), NDF (EDNDF), ADF (EDADF) and cellulose (EDCE) were determined using in sacco nylon bag technique. A ruminal outflow rate of 0.05/h was assumed. The EDDM were 14.2, 45.1, 44.9 and 79, the EDNDF 13.9, 47.2, 47.3 and 72.7, the EDADF 13.9, 41.7, 44.7 and 72.5, and the EDCF 14.3, 47.5, 47.3 and 71.1 for untreated, treatment A(T-A), treatment B(T-B) and treatment C(T-C), respectively. For all parameters measured, AHP treatment of aspen resulted in higher (P<0.05) digestion coefficients than untreated one. The results of this study show that AHP treatment to aspen can be used as an effective method in improving the nutritive value of aspen sawdust for ruminants. Two experiments were conducted to evaluate the nutritive value of a diet containing 20% alkaline hydrogen peroxide (AHP) treated aspen sawdust for ruminants. In experiment 1, sheep fed treated aspen (treated) had significantly (P<0.05) higher averagy daily gain and improved feed/gain rates compared to animals fed untreated aspen (untreated). Apparent NDF, ADF, organic matter and cellulose digestibilities were significantly (P<0.05) higher for the treated diets. Average apparent NDF and ADF digestibilities were significantly (P<0.05) increased with treated diet while lower tendencis were shown in DM, organic matter and cellulose digestibilities with untreated diet. In experiment 2, the soluble and fractional sizes (a+b) of DM and crude protein (CP) tended to increase with AHP treatment. The treated diet improved (P<0.05) the degradation rates (k) of fraction b as well as discrete time lag before degradation began in the rumen (1) of DM and crude protein (CP). Slightly higher tendency were shown with for whole tract CP disappearance of treated diet as estimated by mobile nylon bag procedure. Results of these experiment suggest that AHP treated aspen can be utilised as a energy source for ruminants.
