Human Immunodeficiency virus (HIV)에 의한 감염의 결과로서 나타나는 여러가지 현상들 중 가장 큰 특징은 감염자가 면역결핍증(Immunodeficiency State)의 상태로 서서히 진행된다는 것이다. 통계역학 조사에 의하면 감염자들의 대부분은 일단 뚜렷한 증세가 없는 상태 (asymptomatic phase)를 1-10년간 유지하다가 발병기 (ARC, AIDS)로 전환하게 되어 사망하게 된다. 바이러스의 성장속도와 발병기는 매우 밀접한 상관관계가 있는 것으로 알려져 있다. 즉 asymptomatic phase 동안은 혈액이나 세포조직내의 바이러스 농도가 매우 낮다가 발병기로 전환하면서 바이러스의 양이 급증하게 된다는 것이다. 이러한 HIV의 잠복기와 만성 감염현상(latency/persistence)을 이해하는 것은 현대 HIV 생물학이 풀어야 할 가장 큰 숙제 중의 하나이다. 본 연구팀들은 HIV의 잠복기가 어떻게 유지되고 활성화되는가에 관하여 분자유전학적, 바이러스학적, 세포생물학적 방법론을 사용하여 연구하였는 바 다음과 같은 결과들을 얻었다. 첫째, in vitro laterncy model들을 U1과 ACH2 세포주들을 사용하여 HIV의 잠복기가 어떻게 유지되고 활성화되는가를 분석한 결과, U1에서는 Tat 단백질이 제한되어 HIV가 잠복기에 있음을 밝혔다. 또한 잠복기의 HIV를 활성화시키는 기작의 하나로 HCMV의 IE1 단백질이 HIV LTR에 미치는 영향을 조사한 결과 세포전사인자인 NF-kB가 중요한 역할을 함을 밝혔다. 둘째, AIDS 사망률(mortality/morbidity)을 증가시키는데 관여하는 인자들 중 Cytomegalovirus (CMV)는 가장 중요한 바이러스 인자로 알려져 있다. 이 바이러스가 생산하는 중요 전사인자인 IE2와 IE1에 관하여 조사한 결과, IE2는 암억제 단백질인 Rb와 접촉하며 IE1은 세포전사인자인 AP-1과 NF-kB를 활성화시켜 유전자 발현을 증가시킴을 밝혔다. 세째, HIV 잠복기 과정에서 큰 역할을 하는 것으로 알려진 monocyte/macrophage의 HIV 감염을 myeloid 세포의 감염이라는 차원에서 연구하였다. 특히 early myeloid line인 HL-60을 모델로 조사한 결과 바이러스는 HL-60에서 높은 빈도로 잠복기로 들어가며, 세포분화의 양상에 따라 바이러스 생산에 큰 차이가 있었다. 또한 HL-60에서 나오는 바이러스는 감염성을 상실하는 것을 관찰하였다. 본 연구의 수행으로 얻어지는 결과들은 AIDS의 잠복기 병리현상을 규명하는데 도움이 될 뿐만아니라, 궁극적으로 AIDS의 치료와 예방에도 기여할 수 있으리라 기대된다.

HIV infection of humans is characterized by a slow progression to an immunodificient state. It has been shown by long-term follow-up studies that a significant portion of HIV-infected individuals remain asymptomatic, without evident pathology, for many years. It appears that there is a tight correlation between viral replication and the development of AIDS. The well-accepted model of viral kinetics in HIV-infected individuals predicts that during the early phase of infection, virus replicates repidly in the absence of antibodies to HIV, causing an early viremia. This is followed by a long period of latency, clinically characterized as the asymptomatic phase, in which viral production is very low or undetectable. In general, the progression to AIDS coincides with an increase in virus titer in the peripheral blood. One of the central questions in the biology of HIV is to understand the mechanism of such viral latency/persistence. The major goal of this research project was to investigate how HIV latency is established, maintained and activated at molecular levels. Our results can be summarized as follows. 1. We studied the molecular mechanisms of viral latency/persistence using in vitro cell line models, U1 and ACH2. Our data suggest that U1 cells seem to have suboptimal levels of Tat protein, while Tat is not the primary factor for low level expression of HIV-1 of human cytomegalovirus. Our data suggest that the cellular transcriptional factor NF-kB plays a central role in the IE1 transactivation of the HIV LTR. 2. We investigated the interaction between HIV and HCMV whose infection has been shown to increase morbidity and mortality in AIDS patients. First, it was demonstrated that the tumor suppressor protein Rb directly associates with IE2 of HCMV which is an extremly potent transcriptional activator of HCMV, IE2. In addition, we found that IE1 activates various viral and cellular promoters by upregulating cellular transcriptional factors, NF-kB and AP-1, probably through the protein kinase pathway. 3. We studied the effects of HIV infection of early myeloid progenitors on hematopoiesis. We used the human promonocystic cell line HL-60 as a model system to study HIV-1 infection of early myeloid cells. It was observed that latent of low level infections by HIV-1 establish at a high frequency in this cell line, as 40% of subclones derived from a fully infected population were negative for RT and p24. Furthermore, different pathways of promonocystic differentiation vary in their ability to stimulate HIV-1 replication. Finally, HIV-1 produced from these early myeloid cells contained unusual characteristics, such as a loss of infectivity. Data obtained from our work should help us to understand HIV latency and its activation, and eventually to design strategies that may prevent or slow down the progression to clinical AIDS.
