We had shown that yeast contains a 25-kDa protein (TSA, thiol specific antioxidant protein) which provides several enzymes with protection against the MCO system containing a mercaptan, but not against the MCO system containing the nonsulfhydryl reducing equivalent. Immunoblotting experiments reveal that rat tissues, especially brains, also contain a 25-kDa protein which can be specifically recognized by antibodies derived against the yeast protein. The yeast protein does not possess catalase, glutathione peroxidase, superoxide dismutase, or iron chelation activites. In this study we attempted to clarify the role of the TSA protein in vivo and in vitro. The results are as follows : The TSA from s. cerevisiae specifically prevents not only the inactivation of enzymes also the fragmentation of protein, incorporation of carbonyl groups into protein, strand breaks in pBluscript plasmid DNA, and the formation of 8-hydroxydeoxyguanosine (8-OH-dG) in calf thymus DNA when induced by the thiol/Fe??/O? MCO system. Ascorbate/Fe??/O? MCO system mediated damage of enzymes could be prevented by the antioxidant protein only in the presence of reduced thiol. It seems that the antioxidant activity of the TSA protein against a MCO system requires thiol as a reducing equivalent to restore its catalytic activity. Two proteins from the yeast, thioredoxin and thioredoxin reductase, when presented with NADPH and the yeast antioxidant protein prevented the inactivation of glutamine synthetase and damage of DNA by the ascorbate/Fe??/O? MCO system. This system also removes hydrogen peroxide effectively. Those results suggested that NADPH-dependent thioredoxin system is a physiological electron donor for TSA and reactivated the antioxidant protein by reversible disulfide-thiol exchange. A TSA was purified from human red blood cells. The TSA exists as a predominat protein in human erythrocytes, which showed distict TSA activites in the presence of dithiothritol or thioredoxin system as a reducing equivalent. The human TSA completely inhibited visible absorption spectral change of oxyhemoglobin, DNA cleavege, and the preoxidation of RBC membrane by a nonezymatic thiol/Fe??/O? MCO system. The complete cDNA encoding thiol specific antioxidant protein was isolated from a human brain cDNA library. An open reading frame was identified and found to encode a polypeptide of 197 amino acids with a Mr of 21,729. Expressin of the human antioxidant cDNA in E. coli yielded a functionally active protein. Oxidative DNA damage, as measured by the 8-OH-dG level, was increased in global brain by ischemia/reperfusion insult. Ischemia/reperfusion insult to gerbil brain did not cause any significant induction of antioxidant enzymes, such as superoxide dismutase, catalase, or TSA. However, catalase activity was slightly decreased. These results suggest that ischemia/reprfusion insult to gerbil brain cause the DNA damage via the production of reactive oxygen species and that the antioxidant enzyme systems were not induced to prevent oxidative damage to DNA. Any loss of antioxidant activity was not found chemical modification of amino acid residues such as arginine, histidine and lysine of the yeast TSA. N-ethylaleimde in the presence of thiols or N-bromosuccimide completely inhibited TSA activity. Cys47, Trp82, and Trp172 conserved highly in a family of proteins that exhibit sequence identity of 20% to 95% with TSA were replaced by Ala and Phe and expressed the mutant TSA proteins (C47A, W82F, and W172F). C47S was completely inactive. W82F and W172F showed partial activity as 80% and 20% of the activity of wild type TSA. The growth rate of E. coli strain expressed with wild type yeast TSA was significantly higher than that of E. coli in the presence of hydrogen peroxide. The growth rates of E. coli containing C47A and W82F TSA gene was similar to those of E. coli without TSA gene and E. coli containing wild type TSA gene. The eggect of yeast TSA on the conversion of xanthine dehydrogenase form to xanthne oxidase form was not observed in vitro and in vivo.
