AA의 lipoxygenase 대사물질은 다시 phospholipaseC를 활성화시켜 dizcylaglycerol(DG)을 생성시키고 그 결과 protein kinase C (PKC)가 활성화되어 세포의 spreading이 일어난다.본 연구는 따라서 첫째로 HeLa 세포가 gelatin 기질에 spreading하는 과정의 조절에second messenger로서 중요한 AA가 어떠한 인지질로부터 유래되는 지를 규명한바,HeLa-gelatin 부착과정에서 여러 이지질 중 단지 phosphatidylcholine만이 감소하며 다양한lysophospholipid 중 단지 lysophosphatidylcholine 만이 spreading 과정에서 생성되는 것으로 보아 phosphatidylcholine이 PLA2의 활성화는 세포질 calcium의 농도변화 및 세포질 pH의 알칼리화에 기인하지 않으며, 또한 pertussis 혹은 cholera toxin-sensitive G protein 역시 PLA2 활성화와는 무관한 것으로 나타났다. AA와 DG은 PKC를 활성화 시킴으로서 그기능을 나타낸다. PKC는 지금까지 11가지의 isoform이 알려져 있으며 각각의 PKCisoform은 세포내 분포, cofactor의 필요성, 활성화 기작 등에서 차이를 나타냄으로 각 PKCisoform은 세포내에서 서로 다른 기능을 할것으로 추정된다. 따라서 어떠한 PKC isoform이 HeLa-gelatn adhseion 과정에 관여 하는지를 밝힌바, HeLa 세포에서 발현되는 PKCsubspecies를 isozyme에 특이적으로 반응하는 각각의 항체를 사용하여 Western blotting으로 확인한 결과 α,γ,ε,ξ,λ그리고ι등이 발현됨을 알았다. HeLa 세포가 gelatin 기질에spreading하는 과정에서 cytosolic PKC가 membrane으로의 translocation을 검증하여 밝혔다. HeLa 세포가 gelatin에 spreading하는 과정에서 단지 PKCε만이 translocation됨을알았고, PKC의 활성화는 cytosol에서 particulate membrane으로의 translocation을 수반함으로, HeLa 세포에 발현되는 PKC isoform 중 PKCε isoform이 특이적으로 활성화 됨을 았았다.이와같은 연구결과는 세포와 기질간의 상호작용 과정에서 신호전달 경로를 이해하여세포가 ECM에 반응하는 기작에 대한 기초를 제공하며 또한 세포와 기질의 상호작용에 의한 생물학적 현상들 즉 ECM에 의한 세포의 성장과 분화의 조절, 배발생과정, 암의 전이 과정의 조절,그리고 상처치유 등의 과정을 이해하는 기초를 제공할 것으로 기대된다.

ECM receptors alsogive rise to a variety of signaling events. This sutdies were, therefore, performed tounderstand signal transduction initiated by integrins during cell-substratum adhesion.HeLa cells attach to various substrata but subsequent spresding is specific to collagenor gelatin. The spreading is initiated by the activation of phospholipaseA2(PLA2)which produce arachidonic acid (AA) as a consequence of cell surface collagenreceptor clustering.This study examined the mechanism of PLA2 activation and which phospholipidsare hydrolyzed by PLA2 to release AA. The levels of phosphatidylcholine decreases,among various phospholipids, during attachment and spreading of HeLa cells.Lysophosphatidylcholine is the only lysophospholipids formed during HeLa cell adhesionindicating that clustered collagen receptors activate PLA2 to hydrolyzephosphatidylcholine to AA and lysophosphatidylcholine. Among various molecularentities which are known to regulate PLA2 activation, it appears that PLA2 activation isnot mediated by either changes in Ca2+ levels, alkalinization of cytoplasmic pH,activation of protein kinase C or pertussis or cholera toxin-sensitive G proteins.The spreading of HeLa cells requires the activation of protein kinase C (PKC).Membrane-bound PKC was activated during cell attachment and in response to theactivation of a series of lipid second messengers turned on by the ligation of β1-integrin collagen receptors. HeLa cells express the α,γ,ε,ζ,λ and ιisozymess ofPKC as determined by western blotting with specific antibodies. Only PKCε began toredistributed from the cytosol to the membrane during cell adhesion. Most of the PKCε in cells that were in suspension was in the cytosolic fraction. During cellattachment to a gelatin matrix, all of the PKCε moved out of the cytosol, with mostgoing to the membrane fraction. The conventional PKCα and-γisozymes weretranslocated from the cytosol to the membrane only when phorbol ester was present ata concentration that increases the rate and extent of cell spreading. Under normalconditions, i.e.in the absence of phorbol ester, PKCε appears to be the PKC isozymeresponsiblefor the regulation of HeLa cell adhesion to the extracellular matrix.
