독성이 없으면서도 탁월한 예방과 치료효과를 주는 Influenza virus에 대한 항바이러스제 개발의 필요성은 오래 전부터 인류의 숙원이 되어 왔는데 이는 보통 선인의 경우 1년에 한, 두차례 또는 두, 세차례 이들 바이러스의 공격을 받으며 이들 바이러스에 감염될 경우, 이 바이러스에 의해 유발되는 감기나 유행성 독감 그 자체보다 이 질환으로부터 야기되는 합병증에 의해 많은 사망자가 발생하고 또한 이들 질환이 만병의 원인이 되기 때문이다(1). 역사적으로 인플루엔자의 중요성은 격심한 합병증에 대한 피해로부터 유래되고 있다. 이와
독성이 없으면서도 탁월한 예방과 치료효과를 주는 Influenza virus에 대한 항바이러스제 개발의 필요성은 오래 전부터 인류의 숙원이 되어 왔는데 이는 보통 선인의 경우 1년에 한, 두차례 또는 두, 세차례 이들 바이러스의 공격을 받으며 이들 바이러스에 감염될 경우, 이 바이러스에 의해 유발되는 감기나 유행성 독감 그 자체보다 이 질환으로부터 야기되는 합병증에 의해 많은 사망자가 발생하고 또한 이들 질환이 만병의 원인이 되기 때문이다(1). 역사적으로 인플루엔자의 중요성은 격심한 합병증에 대한 피해로부터 유래되고 있다. 이와 같은 사실은 1918-1919년에 있었던 유행으로 세계적으로 2천만명 이상이 희생된 "스페인 인플루엔자"의 피해에서 너무나 잘 인식되어 왔던 것이다(2). 인플루엔자의 주요 합병증은 폐와 관련된 하기도, 순환계, 신경계에 영향을 미친다. 이중 폐와 관련된 합병증이 가장 중요한 부분을 차지하게 된다. 인플루엔자 유행의 크기는 범세계적 유행, 일시적 유행, 국지적 유행, 산발적 발생 등으로 구분할 수 있으며 과거 90년간 범세계적 대유행은 5회에 이르고 이때마다 바이러스의 항원 변이가 크게 일어났었다. 통상 바이러스의 소변이(Antigenic Drift)시 집단 인구의 10-20%, 대변이(Antigenic Shift)시 40-50% 이상 감염을 일으키기도 한다(2). 인플루엔자 바이러스는 A, B, C로 분류되는데 그 중 인플루엔자 바이러스는 소변이와 대변이를 일으킨다. 따라서 미국, 일본 등 선진국에서는 오래전 부터 상기 질병에 대한 치료연구와 더불어 새로운 의약품 개발에 많은 투자를 해 왔다. 이러한 연구 활동 결과 몇몇 화학요법제 및 항생물질들이 개발되었으나 이들 항생제 및 화학요법제들은 정도의 차이가 있을 뿐 그들 자신이 지닌 약리학적 특징으로 일어나는 독작용(toxic reaction)과 항원성의 획득으로 유발되는 과민반응 등 여러 가지 부작용과 호흡기 질환에 관여하는 바이러스 자체가 가지는 다양한 내성기작으로 말미암아 사용에 제한을 받고 있는 많은 경우 아직도 독성이 많거나 효능이 만족할 만한 수준에 이르진 못하고 있다(3). 따라서 범세계적으로 부작용이 최소화 되고 사용상 안전성이 보장되는 새로운 항바이러스제의 탐색 및 개발에 박차를 가하고 있으나 화학 합성제나 항생제 자체가 가지고 있는 한계성으로 말미암아 큼 효과를 보지 못하고 있는 실정이며 효능이 탁월한 무독성 약품의 개발이 절실히 요청되고 있다(3).

In order to search for anti-influenza virus type A agents from Korean traditional prescriptions(herb complexes) and Korean medicinal herbs, we selected 63 traditional prescriptions and 100 medicinal herbs, based on a review of the Korean traditional medicine books. Both methanol extracts and boiling-water extracts were tested, by means of the Haemagglutination Inhibition Test(HIT). 32 new prescriptions of active medicinal herbs, based on Chiljung theory, also were tested by means of HIT. Three of the 63 traditional prescriptions and four of 100 medicinal herbs, which were extracted by methanol, CM-22, CM-26, CM-48, MM-40, MM-57, MM-72 and MM-110(see explanation of nomenclature below) showed strong efficacy against influenza virus type A. CM-22, CM-26 and CM-48 showed anti-influenza virus type A activity at the range of 3.13mg/ml to 25mg/ml and 3.13mg/ml to 12.5mg/ml, respectively. MM-40, MM-57, MM-72 and MM-110 showed antiviral activity at the range of 3.13μg/ml to 12.5mg/ml, 780μg/ml to 6.25mg/ml, 780μg/ml to 3.13mg/ml, 97.5μ-390μg/ml, respectively. Two of the 63 traditional prescriptions and four of 100 medicinal herbs, which were extracted by water, CW-34, MW-1, MW-40, MW-88 and MW-100 showed strong efficacy against influenza virus type A. CW-14 and CW-34 showed anti-influenza virus type A activity at the range of 1.56mg/ml to 100mg/ml and 780μg/ml to 100mg/ml, respectively. MW-1, MW-40, mW-88 and MW-100 showed antiviral activity at the range of 97.5μg/ml to 100mg/ml 195μg/ml to 100mg/ml, 48.7μ/ml to 100mg/ml and 48.7μg/ml to 100mg/ml, respectively. NPW-6, NPW-25 and NPW-31, which were combined with individual active medicinal herbs and were extracted by boiling-water, showed anti-influenza virus activity at the rage of 390μg/ml to 100mg/ml, 195μg/ml to 100mg/ml and 195mg/ml to 100mg/ml, respectively. NPM-5, NPM-31 and NPM-32, which were combined with individual active medicinal herbs and were extracted by methanol, showed anti-influenza virus type A at the rage of 390μg/ml to 1.56mg/ml and 390μg/ml to 3.13mg/ml. In order to determine cytotoxicity of each extracts, chicken red blood cells were incubated with the various concentrations of extracts of Korean traditional prescriptions, Korean medicinal herbs and new prescriptions. Only MW-40 of water extracts, which had a strong antiviral activity, showed cytotoxic effect against red blood cells at the range of 50mg/ml to 100mg/ml whereas CW-14 and CW-34, MW-1, MW-88, MW-100 did not show cytotoxic effect against red blood cells. CM-22 and CM-48, CM-26, MM-40, MM-57 and MM-72, MM-110 showed cytotoxic effect against red blood cells at the range of 6.25mg/ml to 100mg/ml, 3.13mg/ml to 100mg/ml, 6.25mg/ml to 100mg/ml, 1.56mg/ml to 100mg/ml and 195μg/ml to 100mg/ml, respectively. NPW-6, NPW-25 and NPW-31 did not show cytotoxic effect against red blood cells at all whereas NPM-5. NPM-31 and NPM-32 showed cytotoxic effect at the range of 1.56mg/ml to 100mg/ml, 3.13mg/ml to 100mg/ml and 3.13mg/ml to 100mg/ml, respectively. These results of HIT indicated that Korean traditional prescriptions, Korean medicinal herbs and new prescriptions might be inhibit either attachment of virus to cell surface receptor or penetration of virus into cell during the initial stage of infection. CW-14, CW-34, MW-1, MW-40, MW-88, MW-100, CM-22, CM-26, CM-48, MM_40, MM-57, MM-72, MM-110, NPW-6, NPW-25, NPW-31, NPM_5, NP-31 and NPM-32 are considered as being useful for the development of antiviral drug against influenza virus type A. In establishment of condition for anti-influenza virus activity in vivo, influenza virus type A were passaged 3 and 4 times in Balb/c mouse and had 4 HA unit titers on the 5days, 7 days after infection, respectively. The titer of the allontonic fluid in EID/sub 50/ was 2.6X10/sup 22//ml. Inhibitory effects of fractions on influenza virus type A showed that 40% MeOH fraction of MM-40, H₂O fraction of MM-55 and 20% fraction of MM-63 had very strong antiviral activity at the range of 78μg/ml to 156μg/ml, 1.56mg/ml to 100mg/ml, 12.5mg/ml to 50mg/ml, respectively. In the cytotoxicity of fractions against red blood cells, 40% MeOH fraction of MM-40 and 20% fraction of MM-63 showedcytotoxicity at the range of 78μg/ml to 10mg/ml, 50mg/ml to 100mg/ml, respectively whereas H₂O fraction of MM-55 did not show cytotoxicity.
