1. 제목 남극해 유용생물자원 개발 연구 2. 연구개발 필요성 및 목표 남극해는 막대한 양의 크릴자원과 저온에 적응한 독특하고 다양한 생물자원을 보유하고 있다. 본 연구과제는 궁극적으로 남극해에서의 크릴 어획활동을 증진시키기 위한 기본자료를 축적함과 아울러 새로운 유용생물자원을 개발하여 고부가가치 상품을 창출할 수 있는 기술을 개발하기 위하여 수행되었다. 가. 필요성 (1) 신어장 개척 (2) 크릴 어획쿼타량의 증대 (3) 크릴 어업관리 정책에 대한 조언과 정책방향 제시 (4) 남극생물에서 저온 특유의 생리활성물질, 유용 유전자의 개발 나. 목표 (1) 연구의 최종 목표 (가) 남극해에서 해양조사를 실시하여 크릴의 분포 및 자원량 파악, 그리고 이들에 영향을 미치는 해황 조건을 파악함으로써 우리 어선들이 효율적으로 크릴을 어획할 수 있도록 사전 정보를 제공함. (나) 남극생태계를 총괄적으로 파악하고 생태계 내에서 크릴의 역할과 지위를 이해함으로써 크릴의 어획량을 합리적으로 결정하는데 필요한 과학적 근거 자료를 제공함. (다) 크릴자원량 변동 과정을 파악하여 미래 자원량을 예측하고 이를 크릴자원 관리정책에 반영함. (라) 남극생물 특유의 유용물질, 유용유전자를 개발하여 우리 나라 생명공학 산 업을 진흥시킴 (2) 1단계(1998- 2000)의 연차별 연구 목표 제1단계 연구에서는 크릴의 주요 어장인 남쉐틀랜드군도 해역에서 크릴 분포를 조절하는 요인과 기작의 파악을 염두에 두고 크릴의 분포 및 자원량 산정 연구를 일반 해양조사와 수행하였다. 아울러 남극생물에서 유용한 물질을 개발하기 위해 후보종을 선정하여 기초 연구를 수행하였다. 3. 연구개발의 내용 및 범위 본 연구과제의 3차년도 연구 과제로 2차년도와 비슷한 연구지역을 대상으로 남쉐틀랜드군도 북쪽, 엘리펀트섬 주변 해역, 브랜스필드 해협에서 크릴 자원량 조사를 일반 해양조사와 함께 수행하였다. 추가로 엘리펀트 섬 동쪽 해역에서도 자원량을 파악하였다. 크릴 분포를 조절하는 요인과 남극해 하계 기간의 크릴 자원 변동을 파악할 수 있는 시계열 자료를 얻고자 하였다. 또한, 유전학적 방법에 의한 크릴의 계군분석, 생태계 구조 연구도 수행되었다. 한편, 남극생물의 유용물질을 분석하기 위해 세균의 저온활성 효소, 남극어류와 무척추동물의 부동 물질에 대한 기초연구를 수행하였다. 가. 남쉐틀랜드 군도 부근 해역에서 크릴 자원과 해양 물리 환경 조사 3차년도 연구에서는 남쉐틀랜드군도 북쪽 해역 및 엘리펀트 섬 주변, 브랜스필드 해협에서 크릴의 분포 및 자원량을 조사하였으며 부가적으로 엘리펀트 섬 동쪽 해역에서도 음향 관측을 실시하였다. 크릴의 분포 위치 및 예측된 자원량을 1, 2차 년도 결과와 비교하여 차이점을 분석하였다. 수온 염분 자료에 의해 해양물리 환경을 기술하고 수괴를 구분하였다. 나. 남쉐틀랜드 군도 주변 해역의 식물플랑크톤 생물량과 분포 양상 남쉐틀랜드 군도 주변 해역에서 크릴의 먹이가 되는 식물플랑크톤의 생물량 을 세포 크기별로 나누어 측정하고 분포 양상을 기술하였다. 다. 남쉐틀랜드 군도 주변 해역의 영양염과 식물플랑크톤 생물량 분포 2000년 1월 남쉐틀랜드 군도 주변 해역의 수괴를 구분하고 수괴에 따른 영 양염과 식물플랑크톤 생물량의 분포, 일차생산력을 수괴에 따라 고찰하였다. 라. 크릴 분포와 자원량 변동을 조절하는 요인 지난 3년간 크릴 자원 조사 결과를 정리하여 고밀도 크릴군이 발견될 가능 성이 높은 지역을 구분하고 크릴 분포를 조절하는 후보 요인을 제시하였다. 조사 기간동안 크릴 자원량 연변동을 변동 주기 예측식과 비교하고 변동을 초래하는 예 상 요인과 앞으로 조사 계획을 제안하였다. 마. 미토콘드리아 유전자 COI 염기서열을 이용한 남극크릴 계군의 유 전적 분석 지난 3년간 남쉐틀랜드 군도 부근 해역에서 매해 채집된 크릴들이 또 인도 양 해역에서 채집된 크릴이 유전적으로 구별되는 계군인지 여부를 조사하기 위하여 크릴 DNA의 CO1 유전자를 분석하고 결과를 제시하였다. 바. 남극성게의 채액량, 지질과 단백질의 함량, 아미노산 조성을 분석하였다. 사. 남극에서 분리한 저온성 세균 유래 DNA 중합효소 개발 남극에 서식하는 저온성 세균확보하고 이를 대량 배양하여 저온에서 활성을 갖는 새로운 DNA 합성효소를 분리, 정제하고 그 특성을 분석하였다. 아. 남극어류 결빙방지 단백질의 유전자 검색 결빙방지 단백질 유전자를 확보하고 이를 산업적으로 이용할 목적으로 연중 남극대구 시료를 채집하고 간과 이자에서 전체 RNA를 추출, 분석하였다. 4. 연구개발 결과 가. 남쉐틀랜드 군도 부근 해역에서 크릴 자원과 해양 물리 환경 조사 2000년 12월 3일부터 12월 28일까지 총 2,810 n mile의 정선에서 음향관측을 실시하였다. 계산된 자원량은 남쉐틀란드 군도 북부 해역에서 1,343,000 톤 (area : 9,470 mile2, CV : 21.5%), 엘레판트 섬 주변에서 758,000 톤 (area : 6,308 mile2, CV : 39.355%), 브랜스필드 해협의 중앙부에서 463,000 톤 (area : 3,316 mile2, CV : 21.24%, Leg 2), 640,000 톤 (area : 4,621 mile2, CV : 21.19%, Leg 3), 그리고 엘레판트 섬의 동쪽 해역에서 379,000 톤 (area : 4,653 mile2, CV : 17.93%) 이었다. 남쉐틀란드 군도 북부 해역에서 고밀도의 크릴군은 조사 지역 중앙부의 대륙사면 부근의 넓은 지역에서 관측되었다. 엘레판트 섬은 섬을 둘러싼 모든 대륙사면에서 고밀도를 보였는데 특히 북동쪽의 사면지역에서 전체 조사 지역 가운데 가장 높은 분포를 나타내고 있었다. 한편, 브랜스필드 해협의 중앙부는 킹조지 섬 남쪽의 사면 부근에서 높은 밀도를 보였다. 남쉐틀란드 군도 북쪽 해역에서 1999/2000 조사와 비교할 때, 강도가 약해진 Winter Water와 강도가 비슷한 Circumpolar Deep Water가 나타났다. Drake Passage와 브랜스필드 해협이 연결되는 남북 방향의 정선에서는 해협의 중앙부에 V-형의 밀도 분포와 남단에 Weddell해의 영향에 의한 Antarctic Slope Front Water가 나타났다. 해협의 동서 방향에서는 100 m 상층부는 Antarctic Surface Water에 의해 thermocline이 형성되었으나 하층은 거의 성층화된 구조를 보였다. 한편, T/S diagram에 의해 수괴를 분석한 결과 5개의 수괴가 있는 것으로 나타났으며, 부력 주파수는 200 m 상층부는 2 4 cycles/hr로 나타났으니 200 m 이하에는 거의 1 cycle/hr 미만으로 안정도가 약한 것으로 나타났다. 나. 남쉐틀랜드 군도 주변 해역의 식물플랑크톤 생물량과 분포 양상 2000년 12월 18일부터 26일까지 남 쉐틀랜드 군도 주변 해역에서 크릴 자원 량 조사와 연관되어 총 38개의 연구 정점에서 식물플랑크톤의 분포 양상과 생물량 (chl a)이 조사되었다. 전 연구정점의 100 m 이상의 상층부에서 측정된 Integrated chl a 농도(0-100 m)는 6.77 - 122 mg chl a m-2 범위(평균 60.6 mg chl a m-2)를 보였다. 50 mg chl a m-2 이상(Integrated chl a 농도, 0-100 m), 1 mg chl a m-3 이상(surface chl a 농도)의 식물플랑크톤 생물량을 보인 지역은 남 쉐틀랜드 군도 의 북쪽 대륙붕을 경계로 남쪽 해역에 존재하였다. 이는 이 지역에 nanoplanktontic(<20 m) 식물플랑크톤인 Cryptomonas sp.와 Phaeocystis sp. (motile stage)가 표층에 우점하고 있었기 때문이었다. 연구 지역의 북서쪽 해역에 >20 m의 microplanktontic 식물플랑크톤(주로 돌말류)이 우점하고 있었다. 연구기 간 동안 연구 해역에서 20 m 이상의 식물플랑크톤과 이하의 편모식물플랑크톤이 전체 식물플랑크톤 현존량의 15%, 85%을 각각 차지하고 있었다. 남 쉐틀랜드 군도 남쪽 해역의 브랜스필드 해협 연구지역의 평균 chl a 값(82.9 mg chl a m-2)과 북chl a 값(50.4 mg chl a m-2)을 비교 할 때 브랜스필드 해협에서 32.4 mg chl a m-3(1.6 배) 정도 높은 값을 보였다. 식 물플랑크톤 생물량과 일차소비자인 크릴과의 연관 관계를 이해하기 위해서는 식물 플랑크톤의 종조성, 생물량, 생산력과 관련된 주요 요인 즉, 생물학적 요인(동물플랑 크톤 포식, 박테리아 분해, 원생동물 분포), 화학적 요인(영양염, 철 이온 농도), 물 리적 요인(수온, 염분 및 수괴안정), 해빙의 분포 등과의 관계를 규명하는 연구가 지속적으로 이루어져야 할 것이다. 다. 남쉐틀랜드 군도 주변 해역의 영양염과 식물플랑크톤 생물량 분포 2000년 1월에 남극 남쉐틀랜드군도(South Shetland Islands) 주변해역에서 수심 200 m 까지 수온, 염분, 영양염, 엽록소, 일차생산력 등을 측정하였다. 표층수 온은 남쉐틀랜드군도 북쪽 드레이크해협(Drake Passage)에서 높았고 남극반도 북동 해역에서 낮았다. 반대로 염분은 드레이크해협에서 낮았고 남극반도쪽으로 갈수록 점차적으로 증가하여 남극반도 북동해역에서 최고 값을 보였다. 표층해수의 영양염 농도는 대체로 드레이크해협에서 낮은 값을 보였고 남쉐틀랜드군도 인접해역에서 높은 값을 보였다. 엽록소 농도는 드레이크해협과 남극반도 인접해역에서 낮았고 킹조오지섬(King George Island) 북쪽해역에서 높았다. 연구해역은 T-S diagram의 특징적인 형태에 따라 4개 해역으로 구분된다; 드레이크해협, 브랜스필드해협 (Bransfield Strait), 혼합 해역(Mixed zone), 웨델해(Weddell Sea) 해역. 이들 4개 해역들은 각각 특징적인 물리, 화학, 생물학적 특성을 보였다. 식물플랑크톤 생물량 과 일차생산력은 드레이크해협과 웨델해 해역에서 상대적으로 낮았고 브랜스필드해 협과 혼합 해역에서 높았다. 드레이크해협 해역에서 식물플랑크톤 생물량과 일차생 산력이 낮은 것은 식물플랑크톤 성장에 필요한 철이 충분히 공급되지 않았기 때문 이고, 웨델해 해역에서는 상대적으로 낮은 수온과 수심 200 m 이상의 깊은 표층혼 합 때문이라고 생각된다. 브랜스필드해협과 혼합 해역에서 높은 식물플랑크톤 생물 량과 일차생산력을 보이는 것은 비교적 안정된 수층구조와 높은 철 농도 때문인 것 으로 생각된다. 라. 크릴 분포와 자원량 변동을 조절하는 요인 크릴의 분포는 수괴 특성, 수심, 지형과 같은 물리적 기작과 먹이생물의 양 과 분포같은 생물현상의 상호작용에 의해 결정되는 것으로 보인다. 지난 3년의 크 릴 자원량 분포를 동시에 보면 높은 밀도의 크릴군이 발견될 가능성이 높은 지역을 몇 군데 지적할 수 있다. 지형적인 이유나 수괴 충돌 때문에 해수의 흐름이 느려지 기 때문에 크릴, 또 경우에 따라 크릴과 먹이 생물의 집적이 이루어지는 곳이 아닌 가 추정된다. 크릴의 자원량은 심한 연변동을 보이며 우리의 최근 자료도 발표된 자원량 변동 주기 예측식과 비교적 잘 일치한다. 이러한 자원량 변동은 가입량의 차이만으로 설명하기 어려우며 먹이생물의 증감보다는 다른 물리적 환경요인의 변 화가 더 결정적인 역할을 하는 것으로 추측된다. 마. 미토콘드리아 유전자 COI 염기서열을 이용한 남극크릴 계군의 유전적 분석 남극 크릴은 남극대륙을 감싸고 있는 남극해 전체에 분포한다. 남극해에서 이 생물종은 그 자원량이 수 억톤에 달한다고 알려져 있어 원양어업의 대상생물로 서 세계 각국의 관심을 끌고 있다. 남극 크릴자원의 관리 및 이용에 기반이 되는 것이 계군분석이다. 남극 크릴의 계군 구조에 대해서 현재 제안되고 있는 대표적 인 가설 두 가지는, 첫째 남극해의 크릴 전체가 하나의 계군이라는 가설과 둘째 남 극 대륙 주변의 각 지역에 독특한 계군이 존재한다는 가설이다. 본 연구에서는 크 릴의 주요어장의 하나이며 주 산란지로 알려진 우리 나라 세종기지 근처 남쉐틀랜 드군도 해역의 크릴 자원 변동을 파악하기 위한 기초단계로서 이 해역에 출현하는 남극 크릴의 계군을 분석하려고 하였다. 또한, 인도양측 남극해 크릴과 남쉐틀랜드 군도 크릴를 비교하려고 하였다. 이를 위하여 미토콘드리아 DNA 염기서열 분석이 라는 유전학적 기법을 이용하여 1998, 1999, 2000년에 남쉐틀랜드군도에서 채집된 크릴과 1999년 인도양에서 채집된 크릴 계군을 분석하였다. 크릴의 치토크롬 유전 자 단편 660개 염기서열을 파악하고 시료 간 서로 비교한 결과 25개 위치에서 서로 다른 염기가 발견되었다. 그러나, 남쉐틀랜드군도에서 채집된 크릴은 연도별로 일 관성 있는 염기서열의 차이를 보이지 않았다. 즉, 1998, 1999, 2000년도에 남쉐틀랜 드군도에 출현한 크릴은 유전적으로 하나의 계군에 속할 수 있음을 시사하였다. 한편, 인도양측 남극해의 크릴은 단 하나의 위치에서 남쉐틀랜드군도 크릴과 일관 성 있는 차이를 보였다. 이것이 계군을 구별지을 수 있는 증거가 될 수 있는가는 여러 개체의 크릴을 더 조사해야 할 것이다. 또한, 남쉐틀랜드 군도에 출현하는 크 릴 계군이 언제나 일정한지 여부도 3-4년 더 조사해야 명확한 결론을 내릴 수 있을 것이다. 바. 남극성게의 채액 분석 남극 성게의 체액에서 유용물질을 발굴하기 위한 기초조사로, 성게 체액 중 화학원소의 함량, 지질과 단백질의 함량, 아미노산 조성을 분석하였다. 성게 체액 의 이온 함량은 다른 해양 무척추동물과 비슷하였다. 단백질과 지질의 함량은 현 저하게 낮았으며, 아미노산 중에서 글라이신의 비중이 높았다. 체액에 아미노산이 상당량 함유되어 펩타이드 같은 작은 분자가 존재할 가능성을 시사하였다. 사. 남극에서 분리한 저온성 세균 유래 DNA 중합효소 개발 저온에서 최적 생육을 보이는 호저온성 균주를 남극해양에서 분리하여 생화학적 특성 및 16S rRNA 염기서열로부터 Shewanella gelidimarina에 속하는 균주로 동정하고 S. gelidimarina L93으로 명명하였다. 본 균주에서 생산되는 저온성 DNA polymerase를 ammonium sulfate precipitation, phosphocellulose column chromatography, BioScale Q2 ion exchange chromatography 및 gel permeation chromatography를 통하여 SDS-PAGE 상에서 단일 band로 정제하고 그 특성을 조사하였다. 아. 남극어류 결빙방지 단백질의 유전자 검색 결빙방지 단백질 유전자가 발현되는 남극대구의 간에서 순도가 높은 전체 RNA가 추출되었다. 유전자 검색을 위해 cDNA 합성에 이용될 degradation이 비교적 적은 RNA가 얻어졌다. 한편, 남극 대구 이자에서 추출한 전체 RNA는 저온 활성의 새로운 효소단백질 유전자를 검색하기에 적합할 것임을 보였다. 5. 연구개발 결과의 활용계획 남쉐틀랜드군도 해역의 크릴 자원 분포와 자원량 파악이 3년에 걸쳐 이루어 졌다. 두 해의 크릴 자원량이 10배 정도 차이가 나고 있어 크릴 자원의 연변동이 매우 심함을 보였다. 앞으로 몇 년 더 조사해야만 이 해역의 크릴 자원에 대한 정 확한 이해를 가져올 수 있을 것으로 판단된다. 그 자료는 다음과 같은 활용이 가 능할 것이다. 또한, 작년부터 시작된 남극생물의 생리 특성에 대한 기초조사 자료 는 생명공학 산업의 발달에 기여할 것이다. 가. 생명공학 산업의 발달 남극해 해양생물자원은 저온 환경에서 필요한 독특한 물질의 원천으로 생각되고 있어 이러한 물질의 연구 개발은 새롭게 떠오르고 있는 생명공학 산업에 필요한 기초 재료를 제공하게 될 것이다. 본 연구의 결과는 저온과 관련된 생명공학 산업의 발달에 크게 기여할 것으로 생각된다. 나. 원양어업의 발달 자원개발에 회의적이고 무조건 보수적으로 치닫고 있는 남극 조약국들에게 적절하고도 과학적인 자원분석과 관리기법을 개발함으로써, 남극해 유용생물자원을 최대한 이용할 수 있는 방안을 제시하여 우리 나라 원양어업에 새로운 어장을 제공하게 될 것이다. CCAMLR와 같은 국제회의에서 남극해 연구 결과를 제시함으로써 남극해 수산자원에 대한 우리 나라의 지속적인 관심을 표명할 수 있고, 어업기득권의 확보와 어획 쿼타의 증대에 본 연구자료를 활용할 수 있을 것이다. 남극해 수산자원에 대한 경쟁이 치열해짐에 따라 조업활동을 구속하는 조치가 추가로 채택될 가능성이 높으며 본 연구결과는. 남극해 생태계의 파악 및 크릴자원의 변동 특성 규명 크릴 자원량은 지난 3년간의 연구에서 보듯 연변동이 매우 심하다. 해양생태계에서 한 생물종의 변동은 먹이 연쇄과정에서 살펴보아야 하므로 크릴 자원에 대한 명확한 이해는 생태계의 종합적 연구로서만이 가능하다. 본 연구는 한국해양연구원 극지연구본부의 남극생태계 특성 파악 연구와 상호 보완적인 연구로서, 남극해 생태계를 총괄적으로 이해하는데 기여할 것이며, 이에 따라 크릴 자원의 변동 특성과 기작을 규명하게 될 것이다.

Needs and aims of the project The Antarctic ocean is known to contain a huge amount of krill stock and a lot of unique living resources adapted to the cold environment. The present research has been conducted to establish basic information and database on the antarctic krill to assist our fishing activities in the Southern Ocean and to develope techniques to make good use of the unique antarctic organisms. A. Needs (1) Development of new fishing grounds (2) Increased quota for the antarctic krill (3) Advice to krill fisheries management policy (4) Development of low temperature specific bioactive molecules and genes B. Aims (1) Final aims of the project 1) To study the distribution and abundance of the major fisheries species, Antarctic krill, and the regional oceanography in order to provide our fishing operators with advice for efficient fishing activities in the Southern Ocean. 2) To provide scientific data for reasonable determination of krill catch amount by way of comprehensive study of the antarctic ecosystem and the role of krill within the antarctic food chain. 3) To fomulate krill fisheries management policies with understanding of processes of krill stock fluctuations and with prediction of the stock. 4) To promote nation"s biotech industries through development of bioactive materials and genes from the antarctic organisms. (2) Research goals of the first period (1998-2000) Most basic and urgent items were selected for the first period. The aims of the first period are to study the distribution and abundance of antarctic krill in the seas around South Shetland Islands which are the one of the major fishing grounds for krill and to perform basic studies for development of useful materials from the antarctic organisms. 3. Content and scope of the project for the year 2000 This year, the third year of the first period, studies on the distribution and abundance of antarctic krill in the seas north, east and south of the South Shetland Islands were to be performed in conjunction with an oceanographic survey. In addition, genetic analysis of krill stocks, ecosystem analysis were also to be performed. Baseline studies for development of useful molecules from the antarctic organisms were to be carried out with selected candidate species. A. Distribution and biomass of Antarctic krill and the hydrography around the South Shetland Islands To estimate krill biomass, a hydroacoustic survey using EK500 scientific echo sounder was conducted on Antarctic krill from 3 to 28 December 2000, in the northern part of the South Shetland Islands and around the Bransfield Strait. The estimated biomass and distribution of krill was compared with the results of our two past surveys. Physical oceanography of the study area is described based on temperature-salinity data and water masses are identified. B. Antarctic phytoplankton biomass and distribution pattern around the South Shetland Islands during December 2000 Size-fractionated phytoplankton biomass and distribution pattern was determined around the South Shetland Islands. Chlorophyll concentration in the surface water and integrated chlorophyll in the upper 100 m are presented. C. Distribution of nutrients and phytoplankton biomass around the South Shetland Islands, Antarctica Phytoplankton biomass, nutrients and primary productivity were examined in relation to water masses around the South Shetland Islands during January 2000. D. Factors controlling the distribution and interannual biomass fluctuation of Antarctic krill Our krill survey data from the past 3 seasons are compared and high density krill patch areas are identified. Major factors controlling the distribution of krill are preliminarilly examined. Biomass estimates from the past surveys are compared with the predicted biomass cycle. Items for further research are suggested. E. Genetic analysis of antarctic krill stocks by use of mitochondrial DNA sequences Mitochondrial DNA sequences of krill were analyzed to deterime if the krill stocks occurring near the South Shetland Islands every year and also the krill from the Indian Ocean Sector are genetically distinguishable. F. Biochemical composition of body fluid from Antarctic sea urchins. As a preliminary study to develop the potential bioactive molecules from Antarctic invertebrates, the elemental compostion, the level of protein and lipid and the amino acid composition were determined from the body fluid of Antarctic sea urchins. G. Cold-adapted DNA polymerase from psychrophillic Antarctic bacterium A psychrophillic Antarctic bacterium was isolated and cultured. Cold-adapted DNA polymerase was extracted and purified. H. A study on the genes for the antifreeze protein from Antarctic fish In order to obtain and study the genes for antifreeze protein, RNA was extracted and analyzed from the Antarctic fish. 4. Results A. Distribution and biomass of Antarctic krill and the hydrography around the South Shetland Islands Acoustic/oceanographic survey was conducted along the 2,810 n mile transect around the South Shetland islands from 3 to 28 December 2000. Estimated krill abundance in the survey area was as follows ; South Shetland Islands - 1,343,000 Mt (area : 9,470 mile2, CV : 21.5%) Around Elephant Island - 758,000 Mt (area : 6,308 mile2, CV : 39.355%) Central Bransfield Strait - 463,000 Mt (area : 3,316 mile2, CV : 21.24%, Leg 2) - 640,000 Mt (area : 4,621 mile2, CV : 21.19%, Leg 3) East Elephant Island - 379 Mt (area : 4,653 mile2, CV : 17.93%). Higher krill aggregations were largely found on the continental slope at northern central part of the South Shetland Islands. In the case of Elephant Island, higher krill patches appeared around the almost continental slope. Especially, very strong krill patches among entire study area were found at the north-eastern of the island (Leg 3). Higher krill patches in the Bransfield Strait were found on the continental slope at southern part of King George Island. As compared with results of 1999/2000 survey, strength of Winter Water decreased but strength of Circumpolar Deep Water remained about the same in the northern part of the South Shetland Islands. Density field with V- shape was found in north eaastern Bransfield Basin along north- south transect. Also Antarctic Slope Front Water due to Weddell Sea appeared at the extremely southern part of the transect. In the east-west transect of the strait, strong thermocline was formed upper 100 m depth due to Antarctic Surface Water. But all water was stratified at lower 100 m depth. In the mean time, water masses due to T/S diagram were classified into 5 typees. The buoyancy frequencies of water column were about 2 4 cycles/hr upper 200 m depth, about under 1 cycle/hr at lower 200 m depth. B. Antarctic phytoplankton biomass and distribution pattern around the South Shetland Islands during December 2000 Surface and integrated (0- 100 m) chl a concentrations were measured around the South Shetland Islands (total of 38 stations) during December, 2000. Mean total integrated chl a concentration during the whole cruise period was 60.6 mg chl a m-2, ranging from 6.77 to 122 mg chl a m-2. Dominant phytoplankton was nanoplanktonic (<20 m) phytoflagellates such as Cryptomonas sp. and Phaeocystis sp. (motile stage). Nanoplanktonic phytoplankton accounted for more than 85% of the total chl a concentration. The mean integrated chl a in the Bransfield Strait region was 82.9 mg chl a m-2 which was 1.6 times higher than in the Drake Passage stations (50.4 mg chl a m-2). Higher chl a concentrations (>50 mg chl a m-2, >1 mg chl a m-3) were found in southern part of continental shelf of the South Shetland Island. We would speculate that the phytoplankton dynamics in the study area would be controlled by various physical, chemical, and biological factors such as upwelling (nutrient supply), front formation, iron limitation, vertical stability, sea-ice distribution, and grazing pressure by microzooplankton. C. Distribution of nutrients and phytoplankton biomass around the South Shetland Islands, Antarctica Temperature, salinity, nutrients, chlorophyll- a, and primary production were measured within the upper 200 m water column in the area around the South Shetland Islands in January, 2000. Surface temperature was relatively high in the Drake Passage north of the South Shetland Islands and low in the northeastern area of the Antarctic Peninsula. In contrast, surface salinity was low in the Drake Passage and increased toward the Antarctic Peninsula, reaching the maximum value in the northeastern area of the Antarctic Peninsula. Surface nutrients was low in the Drake Passage and high in the area near the South Shetland Islands. Surface chlorophyll- a was also low in the Drake Passage and near the Antarctic Peninsula and high in the area of the northern King George Island. The study area could be classified as four geographical zones based on the characteristic shape of the T/S diagrams; the Drake Passage, the Bransfield Strait, the mixed zone, and the Weddell Sea. Each geographical zone showed apparently different physical, chemical, and biological characteristics. Phytoplankton biomass and primary production were relatively low in the Drake Passage and the Weddell Sea and high in the Bransfield Strait and the mixed zone. The low availability of iron could be responsible for the low phytoplankton biomass and primary production in the Drake Passage. However, the low phytoplankton biomass and primary production in the Weddell Sea could be explained by the low temperature and deep surface mixing down to 200 m. The high phytoplankton biomass and primary production in the Bransfield Strait and the mixed zone appeared to be sustained by a relatively high iron concentration and the stable water column. D. Factors controlling the distribution and interannual biomass fluctuation of Antarctic krill The interaction between physical features (water mass, depth, bottom topography) and biological processes (abundance and biomass of food organisms) appears to control the distribution of Antarctic krill. The areas where high density krill aggregations are likely to be observed could be identified from our past surveys. It is speculated that there are areas where krill and/or food organisms are retained by the sluggish water formed due to topography and the mixing of water masses. Our krill biomass estimates from the past surveys fluctuate considerably between years and agree reasonably well with the predicted krill biomass cycle. Such fluctuation cannot be explained by the variation of recruitment and seems to be governed by the variability of the major physical environmental factors rather than the annual variation of phytoplankton biomass or the simple re-distribution of krill population. E. Genetic analysis of antarctic krill stocks by use of mitochondrial DNA sequences Antarctic krill occurs all over the Antarctic ocean that surrounds the Antarctica. Because of its huge biomass, krill is being paid world- wide attention as a new fishery species. As for the management and utilization of the krill resource, its stock identification should be done first. Two hypotheses are proposed for the population structure of the krill: the first is that all the krill in the Antarctic ocean comprise a single stock since they migrate around the Antarctica along the circum-antarctic current and the second is that there are many distinct stocks separated in each region of the Antarctic ocean. In this study, we tried to separate the krill stocks occurring near the South Shetland Islands that is one of the major spawning grounds of krill and an important fishing ground, and also those from the Indian sector of the Antarctic ocean. For this, we analyzed a mitochondrial gene cytochrome C oxidase subunit I of 13 krills sampled in 1998, 1999, and 2000 from the South Shetland Islands and in 1999 from the Indian sector. Of 660 nucleotides of the gene segment sequenced, 25 nucleotide differences were observed among the samples. Among the yearly krill samples from the South Shetland Islands, there was no consistent differences, suggesting that all three year krills belong to one single stock. Between krills of the South Shetland Islands and those of the Indian sector, only a single consistently different nucleotide was observed. It seems to be too early to make any conclusion on the population genetic structure of the antarctic krills. Long term monitoring of krill stocks in the South Shetland Islands and further analysis of krills from different sectors of the Antarctic ocean would be required before any conclusion is made. F. Biochemical composition of body fluid from Antarctic sea urchins As a preliminary study to develop the potential bioactive molecules from Antarctic invertebrates, the elemental compostion, the level of protein and lipid and the amino acid composition were determined from the body fluid of Antarctic sea urchins. Ion concentrations were similar to those of other marine invertebrates. The level of protein and lipid was fairly low. The body fluid of Antarctic sea urchin was overwhelmingly rich in Glycine, which may play a role as an antifreeze molecule. G. Cold-adapted DNA polymerase from psychrophillic Antarctic bacterium A psychrophillic bacterium was isolated from Antarctic marine sediment and identified as Shewanella gelidimarina species based on the biochemical properties and 16S rRNA sequence, and designated as Shewanella gelidimarina L93. DNA polymerase produced by this strain was purified through ammonium sulfate precipitation, phosphocellulose column chromatography, BioScale Q2 ion exchange chromatography, and gel permeation chromatography to single band on SDS- PAGE, and basic properties of this enzyme were investigated. H. A study on the genes for the antifreeze protein from Antarctic fish In an attempt to secure and study the genes for antifreeze proteins, total RNA was extracted from the liver of the Antarctic fish. The extracted RNA was fairly pure enough to make cDNA, although degradation occurred in same samples. Total RNA was also extracted from the pancreas of the fish, which seemed to be useful for identification of any new enzyme genes. 5. Planned uses of the research outcomes Up to the third year of the first period of this project, krill surveys have been conducted near the South Shetland Islands. The results show that the amounts of krill stocks can fluctuate by an order of magnitude between years. Therefore, further surveys are needed for better understanding of krill resources in this survey area. Such results would be helpful for the distant water fishery industry. In addition, new research on the physiological characteristics of the antarctic organisms which started last year would be helpful to promote biotech industries. A. Promotion of new biotech industries Antarctic marine organisms are the unique resources for bioactive molecules working in the cold environment. Studies on these molecules would constitute basis for development of biotech industries which are presumed to be the leader of 21 centrury industries. The results of the current project could contribute to the development of low temperature specific biotech companies. B. Assistance to the distant water fisheries industry Appropriate scientific management scheme needs to be presented to conservation oriented Antarctic Treaty nations for the full use of the fisheries resources. Pursuit of national policies need to be backed by science at international conventions and research results are the most important tools in securing the quota. Debate over the far seas fisheries resources has been continuing at various international conventions with CCAMLR being one of the most influential and prestigious for the antarctic living resources. The research results of this project help our nation to maintain a major position in CCAMLR and to secure the privilege of fishing in the antarctic seas. As the antarctic fishing activities are becoming stronger, it is previsioned that new measures regulating the Antarctic fisheries will be introduced. The results of this project will be helpful to prepare for such coming event. C. Understanding of the Antarctic ecosystem and fluctuation of krill biomass As the results of the past surveys suggest, annual variation of krill biomass in the study area is very high. To understand the krill resource better, it is necessary that the entire ecosystem of the antarctic ocean is examined. The present research will complement the "Research on the characteristics of the Antarctic ecosystem" which is being carried out by the Polar Research Center of Korea Ocean Research and Development Institute and help to make clear understanding of the antarctic ecosystem and also of the fluctuation of krill stocks.
