Ⅰ. 연구제목 지구환경변화 및 오염에 따른 남극생물의 유전자 변화 연구와 검색 물질 개발 Ⅱ. 연구개발의 목적 및 필요성 1. 연구의 필요성 환경오염, 지구온난화, 오존층 파괴 등 지구환경변화가 생태계와 생 명현상에 미치는 영향에 대한 인류의 관심과 우려가 증대되면서 오염과 환경문제에 대한 국제적인 협약과 규제가 강화되고 있다. 개발도상국과 후발공업국은 이러한 국제환경에서 상대적으로 불리한 입장에 처하고 있다. 우리나라도 위 문제에 대한 과학적 자료와 연구가 미흡한 상태라 할 수 있다. 따라서, 생명현상의 기본인 유전자와 핵산물질의 분석을 통하여 오염과 환경변화의 영향과 생태계 파급효과를 정확히 진단하는 첨단기술의 개발 필요성이 절실하다. 남극은 지구상 유일한 청정지역으로 환경변화가 생명현상에 가장 민감하게 반영되는 곳으로 환경변화의 영향을 연구하고 방법론을 평가, 정립하는 최적지라고 할 수 있다. 2. 연구의 목표 가. 환경변화(지구온난화, 오염, 자외선 증가 등)에 반응하여 발현되는 남극생물의 유전자 및 세포물질(유용한 생리활성물질, 신물질, 자외선 차단물질 등)을 규명함. 나. 남극생물에 대한 환경변화의 영향을 정량적으로 분석할 수 있는 생리,유전적 변화의 검색물질(biomarker)을 개발함. 다. 환경변화에 수반하는 생태계 군집구조와 종다양성 변화, 해양생물의 피해정도(생산성, 번식 및 생리활동 저하)를 분석함. Ⅲ. 연구개발의 내용 및 범위 가. 환경변화에 대응하여 체내에 생합성되는 세포물질을 규명 식물성 조류, 미생물, 해양저서동물 등 주요 남극해양생물을 대상으로 오염, 온도증가, 자외선 증가 등 환경변화에 따라 세포내에 생합성되는 특정 세포물질 (자외선 흡수물질, 지방산, 스테로이드, 펩타이드 등)을 HPLC, GC 기법으로 추출하고 정제한 후 그 구조를 분석한다. 나. 환경변화에 따른 유전자 발현 패턴의 변화분석, biomarker 개발 환경변화에 따른 남극생물의 유전자 발현 패턴의 변화를 분석하기 위하여 발현유전자 라이브러리(cDNA library)를 제작한다. 또한, 발현된 각 유전자를 규명하고 그 염기서열을 확보하기 위해 cDNA 라이브러리의 각 클론을 시퀀싱하고 Expressed Sequence Tags (ESTs) 데이터베이스를 만든다. ESTs 중에서 생물학적으로 흥미를 끄는 유전자를 선택하여 환경변화에 따른 발현량의 변화를 분석한다. 긍극적으로 환경조건의 변화, 환경 스트레스를 지시하는 유전학적 생물질 표지(biomarker)를 개발한다. 다. 오염과 환경변화가 남극생태계의 생산성, 군집구조, 종다양성에 미치는 영향 연구 오염과 환경변화(자외선 증가 등)에 따른 남극생물의 생리와 발생과정의 변화, 식물플랑크톤과 해조류 등 일차생산자의 생산성 저해, 생태계 군집구조와 종다양성의 변화 등을 분석한다. (본 연구는 처음 계약 시 연구 기간이 3년이었으나 이 후 2년으로 연구 기간이 단축되고 연구비 또한 크게 감액되었다. 따라서, 연구자들은 연구의 범위를 축소하여 환경 변화 요인으로 자외선 증가에 대해서만 연구를 수행하였다.) Ⅳ. 연구개발결과 가. 남극 오존층 파괴에 의한 자외선 증가가 식물플랑크톤에 미치는 영향 최근 남극 해양 생태계는 지속적인 오존층 파괴에 의한 자외선 증가 로 영향을 받고 있다. 오존층 파괴에 의한 자외선 증가로 인해 남극 해양 생물들의 성장 환경이 변화하는 등 여러 가지 증후들이 남극 해양 생태계에 나타나고 있다. 특히 빛을 에너지원으로 하고 빠르게 성장하는 일차생산자들인 식물플랑크톤들이 남극 해양의 환경 변화 에 가장 민감하게 영향을 받고 있다. 오존층 파괴에 의한 자외선 증가는 전체 남극 해양 생태계를 유지하기 위한 주요 탄소 공급원인 식물플랑크톤의 유전적, 생리적인 변화를 야기 시켜 생산력, 생물량 및 종조성의 변화를 가져와 전체 남극 해양 생태계를 변화시키고 있다. 앞으로 진행될 해양 생태계 변화를 예측하기 위해 앞으로의 자료와 비교할 수 있는 기본 자료의 축적이 필요하다. 식물플랑크톤들은 오랜 기간 극한 환경에 적응하여 왔기 때문에 미세한 환경변화를 쉽게 감지하고 감시하기 위한 생물학적 환경 변화 지표종으로 지속적인 표준 데이터로서 이용될 수 있을 것이다. 나. 중파 자외선에 노출된 남극 규조 Chaetoceros neogracile 와 Stellarima microtrias의 성장과 영양염 흡수율의 변화 남극 해산 규조 두 종을 배양하여 중파자외선이 chlorophyll a(chl a) 증가와 영양염 흡수에 미치는 영향을 실험하였다. 중파자외선은 남극 세종기지 주변의 한 지역에서 측정한 값의 100-600 % 정도 범위에 이르는 1-6 W m-2 의 광도로 조사하였고, 조사시간은 고광도(6 W m-2)에서는 2-60분, 저광도(1 W m-2)에서는 1-5시간을 주었다. 그 결과, 고광도 및 저광도의 중파자외선에 노출된 두 규조(Chaetoceros neogracile Van Landingham and Stellarima microtrias (Ehrenberg) Hasle & Sims)의 chl a는 중파자외선이 없는 대조군에 비교하여 증가폭이 매우 감소하였다. 또한 고광도의 중파자외선에 노출되면 회복력이 떨어져 영양염의 흡수율이 급감하는 것으로 나타났으며, 저광도에 한시간 정도 노출된 두 strain은 다시 배양하면 회복 및 성장을 통해 영양염의 흡수가 지속되는것으로 나타났다. 그리고 S. microtrias이 C. neogracile에 비해 자외선에 대한 생존 가능성이 더 높은 것으로 나타나, 세포의 크기가 자외선에 대한 내성에 밀접한 관계가 있는 것으로 생각된다. 이 실험적 증거를 통해 중파자외선 특히 고광도의 중파자외선은 남극해에서 규조의 생존율을 감소시키며, 미세조류에 의한 영양염의 감소율을 낮추는 원인이 된다고 생각된다. 또한 유해한 중파자외선의 증가는 남극 표층생태계에 서식하는 미세조류 군집을 제한하는 요인이 된다고 생각된다. 다. 중파자외선에 대한 남극산 해조류의 광합성 및 자외선 흡수물질 형성 반응 남극산 홍조의 자외선에 대한 광합성 반응은 서식처 광환경의 영향을 받아서 수심 얕은 곳에 생육하는 종일수록 자외선 내성이 크다. 또한 자외선에 대한 이러한 생리적 탄성의 차이는 자외선 흡수물질의 존재 여부와 함유량과 밀접한 관계를 지닌다. 라. 지구환경 변화에 따른 자외선량 증가에 의한 생물의 방어 전략으로서의 자외선 흡수물질 Mycosporin-like AminoAcid (MAA) 연구 환경 변화에 따른 자외선의 증가로 자외선 흡수물질을 합성하는 것으로 알려진 남극 생물의 자외선 흡수물질 (Mycosporin-like AminoAacid)의 함량 및 종류를 분석하였다. 우선 MAA의 분리 및 분석을 위한 최적 분석방법을 확립하였으며 Krill 및 Porphyra endiviifolium을 위시한 남극산 해조류를 중심으로 분석이 수행되었으며 MAA-ser이 가장 많이 발견되는 종류였다. 남극해 일원의 MAA 분석도 수행되었는데 MAA-Met가 가장 높은 함량을 보이는 종류로 나타났다. 마. 남극 해양 생물들의 지방 및 지방산 조성 분석 남극 생물의 저온 적응 기작의 이해를 위하여 지방 및 지방산 조성의 분석을 수행하였다. 저온성해양세균부터 Phaeocystis antarctica, Krill (Euphausia superba), 그리고 변온 생물 단계에서의 먹이사슬의 최고봉이라고 할 수 있는 어류 (Notothenia sp.)까지 지방 및 지방산 조성을 분석하였으며 EPA, DPA 및 DHA 등의 고도불포화지방산의 함량이 5 - 25 % 수준으로 거의 모든 생물에서 발견되었다. 바. Anticancer phospholipids for antarctic sponge 남극에서 채집된 대형 육방 해면(Hexactinellida)의 일종인 Scolymastra sp.로부터 유기용매 추출, partition, Flash column chromatography와 reverse phase column HPLC를 이용하고 human leukemia K-562 cell line에 활성을 지침으로 항암 활성을line으로 구조를 결정하였다. 사. Alpha-oxidase 분석 방법 개발 해양발광 세균 Photobacterium phosphoreum을 이용하여 해조류와 식물의 α-oxidase(fatty acid : oxygen dioxygenase, 1 - decarboxylating)의 활성 측정법을 개발하였다. 기질로는 pentadecanoic acid를 사용하였으며 1 nM to 10 μM 범위에서의 측정이 가능하다. 최적 pH 와 온도는 각각 pH 7.5와 25 oC 였다. 본 방법은 매우 빠르고 안전하며 쉬운 장점이 있다. 아. 남극 해양생물의 배아 발생에 미치는 자외선의 영향 남극 성층권의 오존감소에 따른 중파자외선(UV-B; 280 nm - 320 nm)의 증가는 남극생물과 생태계에 심각한 환경변화로 작용한다. 식물플랑크톤과 해조류는 일차생산력이 감소하고 해양저서생물의 배아와 유생도 영향을 받게된다. 본 연구는 남극해 연안의 대표적인 저서생물인 남극성게를 대상으로 중파자외선이 그 배아 발생에 미치는 영향을 조사하였다. 포배기 중기, 낭배기, 프리즘 단계 배아에 중파자외선을 쬐었을 때(1W/m2) 각 배아는 발생단계에 따라 받는 영향 정도가 달랐다. 낭배기와 프리즘 배아는 30분 정도의 자외선 조사에 잘 견디었으나 포배기 배아는 24시간 이내에 모두 죽었다. 자외선을 3시간 쬐었을 때는 위 세 발생단계의 배아 모두가 6시간 이내에 죽었다. 한편, 자외선 조사는 배아의 발달형태를 변형시켜 세 발생단계의 배아에서 중배엽세포 수가 증가하고 동물극 세포가 터지는 것이 관찰되었다. 자. Construction of gene expression library and expressed sequence tags(ESTs) with respect to UV-B irradiation 생물은 환경조건의 급격한 변화에 대응하여 특별한 유전자들을 발현하는 것으로 알려져 있다. 온도충격유전자들은 그 대표적인 것으로 외부 온도의 갑작스런 상승에 따라 발현하는 유전자들이다. 자외선에 대한 영향은 피부암과 관련하여 연구가 되었던 바, apoptosis에 관련한 몇몇 유전자(p53, Jun, JNK 등)가 알려져 있다. 지난 30년간 오존홀의 형성으로 중파자외선의 급격한 증가를 경험하고 있는 남극생물에서 이에 반응하는 유전자에 대한 연구는 거의 이루어지지 않았다. 본 연구에서는 남극성게의 낭배기 배아에 중파자외선을 쬐어준 후 나타나는 유전자 발현패턴의 변화를 분석하고자 발현 유전자 라이브러리(cDNA library)를 만들고 발현된 유전자를 확인하였다. 지금까지, 229개 발현유전자 클론을 시퀀싱하여 그 염기서열을 얻어 데이터베이스(ESTs)를 구축하였다. 발현유전자들 중에서 Cu-Zn superoxide dismutase, Catalase, Polyadenylate-binding protein, Speract, Octamer binding protein, profilin, Insulinoma rig protein 등의 상동유전자가 확인되었으며, 114개 유전자는 아직 알려지지 새로운 유전자였다. 차. 남극생물의 자외선 영향을 감지하는 Biomarker 선정과 그 검색 방법 개발 환경조건의 변화는 생물체에 영향을 주어 그에 반응하는 특정 유전자들의 발현을 유발한다. 이러한 상관관계는 역으로 생물체에서 발현된 유전자를 파악함으로써 어떤 환경조건이 변화했는가를 추적하는 것이 가능토록 한다. 독성물질에 반응하는 유전자들이 모두 망라된 DNA 칩은 유전자 발현 패턴으로써 미지의 독성물질을 판별하는데 이용되고 있다. 본 연구에서는 자외선의 영향을 지시할 수 있는 유전자 biomarker를 확립할 목적으로 남극성게의 배아에 중파자외선(UV-B)을 쬐어준 후 발현된 유전자를 다량 검색하고, UV-B에 반응하여 발현양이 변하는 유전자를 biomarker로 선정하였다. 먼저 자외선에 반응하는 유전자 ESTs 중에서 6개의 예비 biomarker 후보를 정하였다: Cu-Zn superoxide dismutase, Catalase, Polyadenylate-binding protein, Profilin, Insulinoma rig protein, Speract. 이후 UV-B가 조사된 배아와 정상의 배아에서 이 유전자들의 발현양을 비교함으로써 자외선에 대한 biomarker로서 Speract, Polyadenylate-binding protein, Insulinoma rig protein 유전자가 유용함을 확인하였다. Cu-Zn superoxide dismutase는 자외선을 쬐었을 때 발현이 오히려 줄어드는 역관계를 보여 역관계의 biomarker로서 사용할 수 있는 가능성을 보였다. 한편, 본 연구에서 유전자의 발현양을 측정하는 방법으로서 gene-scan을 이용하는 새로운 RT-PCR 방법이 개발되었다. Ⅴ. 연구개발결과의 활용계획 가. 남극생물의 자외선 흡수 물질 및 생리 활성 물질 MAA로 알려진 자외선 흡수물질을 여름철 자외선 차단제로 이미 쓰이고 있는 물질로서 산업적인 응용 범위가 넓을 것으로 예상된다. 다만, 본 연구의 후속 사업으로 MAA를 어떻게 다량, 경제적으로 확보할 수 있는가를 연구해야 할 것으로 판단된다. 그외 남극생물의 독특한 지방산 조성, 항암효과가 있는 인지질, alpha-oxidase 효소 등은 산업적 활용도를 보다 면밀히 분석하여 부가가치를 높일 수 있는 연구가 뒤따라야 할 것이다. 나. 자외선에 반응하는 유전자 및 유전자 biomarker. 본 연구에서 speract 유전자는 중파자외선에 반응하여 10배이상으로 발현이 증폭되는 것으로 나타나 중파자외선의 영향을 지시하는 biomarker로서 이상적인 특성을 보였다. 그 외, poly A bindin protein 유전자와 insulinoma rig mRNA등도 자외선에 반응하여 발현이 증가되었다. 따라서, 이들 유전자는 앞으로 자외선 영향을 평가하는 지시자로 광범위하게 사용할 수 있을 것이다. Speract 유전자가 자외선 이외의 어떤 환경 스트레스에 반응하는가는 아직 연구가 부족한 사항으로 환경독성의 일반적인 지시자로서 사용여부를 검증할 필요가 있다. SOD 유전자는 자외선 조사시 오히려 발현이 크게 감소되어 negative한 지시지로서 사용하기에 충분하다. 다. 자외선 증가에 따른 생태계 변화 예측 남극에서 CFCs에 의한 오존홀의 형성, 그에 따른 중파자외선량의 증가는 전적으로 산업화에 따른 인위적인 현상으로 자연현상과 거리가 멀다. 본 연구에서 자외선이 남극생물과 생태계에 미치는 영향을 분석한 자료는 온실가스 증가가 지속될 경우 자외선의 증가로 인하여 발생하는 생태계 종조성과 구조의 변화, 생산성의 저하를 예측하는 데 중요한 자료로 사용될 수 있다. 또한, 지구 자연환경 보존이라는 측면에서 본 연구의 결과가 시사하는 바는 크다.

Ⅰ. Title Responses of Antarctic marine organisms to global environmental changes and pollution; gene expression & biomarkers. Ⅱ. Needs and aims of the project 1. Needs While the concern on the effect of pollution, global warming, and ozone depletion on the earth ecosystem and the organisms increases, international conventions and regulations on these environmental issues have been tightened. Developing countries such as Korea are situated in relatively weak positions regarding these issues compared to the developed countries due to the lack of scientific researches and data. To overcome this situation, Korea needs to encourage studies on the effect of global environmental changes on the ecosystems and their lives and to develop state-of-the-art techniques to measure the effect. The Antarctica provides unique opportunity for the study of environmental change and its effect on the organisms and also for the study of developing new methods to assess the effect since it had remained free of pollution until recently so that the effect of environmental change can be reflected on the lives pretty well. 2. Aims A. Identification of genes and cellular materials that are responsive to the environmental changes such as global warming, pollution and ultraviolet increase. B. Development of new biomarkers that indicate the effect of the environmental changes on the organisms in a quantitative manner. C. Assessment of the harmful effect of the environmental changes on the community structure, species diversity, productivity, and reproduction. Ⅲ. Contents and scope of the study A. Identification of cellular materials that are newly synthesized in response to the environmental changes With the phytoplankton, algae, microorganisms, and marine benthic organisms of the Antarctic as target species, purification and analysis of biochemicals such as UV absorbing materials, lipids, steroids and peptides that are newly synthesized in the cell in response to pollution, global warming, and UV increase will be done. B. Assessment of changes of gene expression patterns in response to the environmental changes and development of genetic biomarkers indicating the effects cDNA library will be made with an antarctic organisms to analyse the changes of gene expression patterns in response to the environmental changes. An EST database will be constructed by random sequencing the cDNA clones. Of the ESTs, new genetic biomarkers will be selected that indicate the effect of the environmental changes on the organisms in a quantitative manner. C. Studies on the effects of pollution and the environmental changes on the production, community structure, species diversity of the Antarctic ecosystem. (This study focused on the issue of UV-B increase in the Antarctica due to ozone depletion) IV. Results of the study A. Effects of the Antarctic ozone depletion on the marine phytoplankton The Antarctic ecosystem has been affected by continuous ozone depletion. Increase of UV radiation due to the ozone depletion in the Antarctic has changed growth environments of the marine organisms, resulting in various effects on the Antarctic marine ecosystem wether adverse or not. Especially, fast growing primary producers, phytoplankton which use solar light as energy source are the most sensitively affected organisms against the UV increase. The UV radiation increase causes to change the whole Antarctic marine ecosystem because the productivity, biomass, and species composition of phytoplankton which are the major carbon producers for sustaining the ecosystem have been changed by genetic and physiological changes of phytoplankton cells. It is necessary to accumulate baseline data which will be compared with the future data to estimate the degree of changes in the marine ecosystem. Phytoplankton which have been adapted for a long time in the extreme environment can be used as continuos standard data and biological indicator to detect and monitor the environmental changes. We have reviewed recent research papers about effects of ozone depletion on Antarctic marine phytoplankton. B. Effects of UV-B radiation on growth rate and nutrient dynamics in Antarctic marine diatoms Chaetoceros neogracile and Stellarima microtrias Increase of Chlorophyll a(Chl a) and nutrients dynamics were investigated in cultures of two commonly occurring Antarctic marine diatoms exposed to a range of UV-B irradiations. Experimental UV-B exposures ranged on 1 or 6 W m-2 that were chosen to span the range of 100-600 % of peak UV-B exposure as measure at an Antarctic coastal site near King Sejong Station. UV-B exposure times ranged from 20 to 60 minute on high irradiance(6 W m-2) and ranged from 1 to 5 hours on low irradiance(1 W m-2). As a result, chl a of the two diatom strains(Chaetoceros neogracile Van Landingham and Stellarima microtrias (Ehrenberg) Hasle & Sims) were less increased under the low and high UV-B irradiances than the control group which was culture without UV-B radiation. The rate of nutrients uptake by the two strains were decreased abruptly at high UV-B irradiances because of decreasing rate of recovery rate. In contrast to high UV-B irradiance, the two strains which were exposed with low UV-B irradiance during one hour were increased concentration of chl a and continued uptake of nutrients. Also, S. microtrias appear higher capable of surviving than C. neogracile under UV-B radiation. It seems that volume of cell was closely related with tolerance of UV-B irradiance. This experimental evidence shows that exposure to UV-B radiation especially high irradiance of UV-B decreases diatom survival and causes to less decrease of nutrients concentration by microalgae in Antarctic water. Furthermore, evidence suggests that microalgal communities confined to near-surface waters in Antarctica will be harmed by increased UV-B radiation, thereby altering the dynamics of Antarctic marine ecosystems. C. Photosynthesis and formation of UV-absorbing substances in Antarctic macroalgae under different levels of UV-B radiation Photosynthetic sensitivity of Antarctic rhodophytes to UV radiation was influenced by the acclimation capability to the surrounding light climates showing that plants from shallow waters were more tolerant of UV stress than those from deeper waters. The differences in physiological plasticity in response to UV may be ascribed to the presence/absence or the relative amounts of UV-absorbing substances. D. Research on Mycosporin-like Amino Acid (MAA) Production by Antarctic organisms as a UV protection An optimal analysis method was established, through ODS and carbon-filtration, 3 HPLC. Antarctic organisms including krill and sea weeds such as Porphyra endiviifolium were analyzed and MAA-ser was the most abundant compound. MAAs including in the sea water at sites in Antarctic sea near Drake passage were also extracted and assayed, and MAA-Met was found as the major UV protection materials. E. Lipid and Fatty acid composition of Antarctic organism Composition of lipids and fatty acid from Antarctic organisms including psycrophillic marine bacterium, Phaeocystis antarctica, Krill(Euphausia superba), fish(Notothenia sp.) were analyzed for the low temperature adaptation mechanism. High content(5 - 25 %) of polyunsaturated fatty acid, such as EPA, DPA and DHA were found almost all Antarctic organisms F. Anticancer phospholipids for antarctic sponge From Scolymastra sp., a species of Antarctic Hexactinellida, an anticancer compounds against human leukemia K-562 cell line was purified through organic solvent extraction, partition, Flash column chromatography and reverse phase column HPLC, and the structure was elucidated with NMR as sn-1-10-methyl stearioyl sn-2-oleioyl Phosphatidyl Choline. G. Analysis of Alpha-oxidase isolated from sea weeds A new assay method of α-oxidase(fatty acid:oxygen dioxygenase, 1-decarboxylating) was developed using bioluminescence reaction system of marine luminous bacterium, Photobacterium phosphoreum. α-Oxidase was isolated from cucumber(Cucumis sativus) and Ulva pertusa, and pentadecanoic acid was used as a substrate, and the product, tetradecanal was analyzed with bacterial luciferase-coupled reaction. Initial light intensity was directly related to the concentration of tetradecanal in the range of 1 nM to 10 μM. Optimal pH and temperature were 7.5 and 25 oC, respectively. Optimal pentadecanoic acid concentration in standard assay of α-oxidase was 0.1 mM, and the Km value of pentedecanoic acid was 85 μM. This method is straightforward, rapid, convenient, and easy needing no treatment or extraction of reaction mixture. H. Differential effect of UV-B on the Antarctic sea urchin embryos depending on developmental stages Increase of UV-B (280 nm-320 nm) radiation in the Antarctica due to ozone depletion in the atmosphere has been a serious environmental problem for the Antarctic ecosystem and its inhabitants. It is known that UV-B irradiance on the phytoplankton can result in inhibition of photosynthesis thereby reduction of primary productivity. Marine benthic organisms that live in the sea floor can also be affected by UV-B radiation since many of them undergo planktonic embryogenesis and larval development in the surface water. In this study, we examined the effect of UV-B radiation on embryos with using the Antarctic sea urchin Sterechinus neumayeri as a model system. Embryos at mesenchyme blastula(mblastula), mid-gastrula, and prism stages were irradiated by UV-B(1 W/m2) for 10 minutes, 30 minutes, one and a half hours, and three hours. Then, their survivorships were monitored through the 24h incubation period following the irradiation. At all stages of embryos, it was observed that the number of mesenchyme cells increased and animal pole cells ruptured after irradiation. Survivorship curves showed that the mblastula embryos could be more vulnerable to UV-B irradiation than the mid-gastrula or the prism embryos. After 10 or 30 min of irradiation, all the mblastula embryos died within 24 hours, but almost all the mid-gastrula and the prism embryos survived. For 1.5 hr irradiation, the mid-gastrula embryos showed no affection up to for seven hours whilst the mblastula embryos underwent 80 % of death by that time. Three hour irradiation of UV-B was so harmful to all the stages of embryos that almost all died within 6 hours. I. Construction of gene expression library and expressed sequence tags(ESTs) with respect to UV-B irradiation The abrupt changes of environmental conditions provoke organisms to express a special set of genes. Heat shock protein genes are one of such examples of which expression increase at high temperature condition. Studies of UV effect on the organisms have been conducted in relation to skin cancer and revealed that several genes such as p53, Jun, JNK are involved in apoptosis after UV irradiation. In this study, we examined the genes expressed in the Antarctic sea urchin embryos when they were exposed to UV-B radiation in order to access the effect of ozone depletion and UV-B increase in the Antarctic. Total RNA was extracted from the UV-B irradiated embryos and mRNA was purified. cDNA library was constructed and each cDNA clone was sequenced. In total, 229 ESTs were obtained of which mitochondrial gene clones were 87, ribosomal gene clones 6, other nuclear known gene clones 21 and unknown gene clones 114. The known genes include Cu-Zn superoxide dismutase, Catalase, Polyadenylate-binding protein, Speract, Octamer binding protein, profilin, Insulinoma rig protein. J. Identification of genetic biomarker for UV-B irradiation from Antarctic organisms and development of a biomarker screening method Environmental changes or toxic chemicals affect animals to express a certain type of genes in coping with the external stresses. By use of the relationship between the external stress and the expressed genes, one can identify the nature of external stress with knowing the type of genes expressed in the target organism, so called genetic biomarkers. For example, a DNA chip was developed to identify the nature of toxic chemicals which was imposed onto the test organism. In this study, to obtain genetic biomarkers indicating the effect of UV-B, we examined the expressed genes from the Antarctic sea urchin when it was irradiated by UV-B. Among the ESTs in the previous section, 6 putative genes were selected first which include Cu-Zn superoxide dismutase, Catalase, Polyadenylate-binding protein, Profilin, Insulinoma Rig protein and Speract. In comparison of expression levels of the genes between the UV-B irradiated embryos and control embryos, Speract gene showed the highest ratio(13:1), and Polyadenylate-binding protein and Insulinoma rig protein gene showed a high ratio(1.6:1, 1.7:1 respectively) suggesting their good use as UV-B biomarkers. On the other hand, Cu-Zn superoxide dismutase(SOD) showed an inverse ratio(0.34:1) indicating the inhibition of its expression by UV-B. SOD could also be used as a biomarker of UV-B by its low level of expression. In this study, a new RT-PCR method was developed for measuring the gene expression level by application of the gene-scan method. Ⅴ. Future application of the study A. UV absorbing materials and other bio-active materials of Antarctic organisms UV absorbing materials of organisms known as mycosporine-like amino acids (MAA) are now being used as ingredients of sun block creams. Since a few MAA were identified in this study, it needs to develop methods on how to obtain MAA in large quantity at low cost. Other cellular materials such as lipids, anti-cancer phospholipids, and alpha-oxidase would be possible candidates for industrialization. B. Genes and genetic biomarkers responsive to ultraviolet In this study, new genetic biomarkers that indicate the effect of ultraviolet were found. Speract of sea urchin was found to be an ideal biomarker for ultraviolet irradiation since its expression level increased more than 10 fold when the organism was irradiated by UV-B. Polyadenylate binding protein and Insulinoma Rig protein genes could be also used as biomarkers for UV. Superoxide dismutase(SOD) could be another type of biomarker for UV because its expression was badly suppressed: only about 30% of the control. ESTs database constructed from cDNA of UV-B irradiated sea urchin embryos could be valuable source of information on the genes responsive to ultraviolet. C. Prediction of change of ecosystem due to ultraviolet increase Effects of UV-B increase on phytoplankton, algae, and other marine organisms were studied. These results could be used for prediction of ecosystem change due to UV-B increase in the Antarctic. In addition, these results could be cited in discussion of protecting the global environment.
