Ⅰ. 제 목 미세조류를 이용한 CO2의 생물제품화 기술 개발 Ⅱ. 연구개발의 목적 및 필요성 우리나라는 에너지의 95%를 수입에 의존하고 있으며 산업구조 및 에너지사용에 있어 화석연료 의존 비율이 높고, 최근 급속한 경제성장으로 온난화가스 배출량이 빠른 속도로 증가추세에 있으므로 CO2 저감 등을 포함한 기후변화협약의 향후 진행 추이에 대하여 적극적인 대응이 필요하다. 미세조류를 이용하여 CO2를 고정화하는 공정이 개발되면 대기로 방출되는 CO2량을 감소시키는데 따른 미래의 직접적인 혜택 (탄소세 도입에 따른 경제적 부담)이외에도 미세조류는 단백질, 탄수화물 등이 풍부하여 사료로 활용 가능하므로 본 공정이 실용화되면 산업체 배출 가스로부터 사료를 보다 저렴한 비용으로 조달할 수 있을 것으로 기대된다. 본 공정이 실용화되면 사료 수입량의 상당 부분을 줄일 수 있을 것으로 기대된다. 미세조류를 이용하여 CO2를 고정화하는 공정은 연소 가스의 전처리 과정이 원칙적으로 필요없고 공정의 운전 조건이 상온, 상압이어서 장치의 설치에 필요한 투자 및 운전 비용이 다른 공정에 비해 크게 낮다. 또한 태양 광을 주 에너지원으로 활용하므로 에너지 소모량이 극히 적으며 그 결과 공정의 운전에 따른 CO2 방출 량도 적다. 따라서 적절한 공정이 개발되면 높은 경제성을 가질 수 있을 것으로 기대된다. 일반 식물과 마찬가지로 조류는 신진대사를 통하여 공기중의 탄산가스를 탄수화물이나 단백질 등 다른 물질로 변환시킨다. 조류 중에서도 Botryococcus braunii는 탄화수소의 생산력이 매우 높은 식물이며 그 조성은 Crude Oil과 같아서 Hydroprocessing을 통하여 얻어지는 생산물의 조성은 휘발유(67%), Kerosin(15% ), 디젤(15%)과 잔류유(3%)로 되어 있다. 이 조류(Botrycoccus)를 이용한 공정은 탄산가스를 제거함과 동시에 탄화수소를 생산하는 미래기술로의 포텐셜이 매우 높아서 기초 과학적인 문헌만 소개되고 있을 뿐이며 공정의 개발은 선진국 일부 회사에서 차세대의 기술선점을 위하여 극비리에 추진하고 있는 중이다. Botryococcus braunii의 이와 같은 특성으로 인하여 현재 미국, 독일, 일본 등의 선진국에서는 이를 이용하여 탄산가스를 제거함과 동시에 신 에너지를 생산하기 위한 공정개발을 활발하게 전개시키고 있다. 한편 이 공정에서도 다른 생물공정과 마찬가지로 질소, 인 등의 영양성분이 필요하므로 이에 대한 공급원으로 축산폐수 및 발효폐액과 같이 영양성분은 다량 함유되어 있으나 그로 인하여 현재 처리상의 큰 문제가 되고 있는 문제폐수들을 자원으로 활용할 수 있어, 이 공정은 이산화탄소의 저감효과와 함께 수질오염물질을 자원화하며 동시에 신 에너지원으로서의 포텐셜이 매우 높은 전방향적인 환경기반기술의 성격을 가지고 있다. Botryococcus braunii를 이용하는 본 공정의 개발에서 현재 세계적으로 핵심 목표가 되고 있는 것은 크게 나누어 두 가지 방향으로서, 조류의 생장속도를 높이기 위한 기술의 개발과 광반응기의 효율을 높이기 위한 반응기 제작기술이나 이 두 가지 문제점은 어느 정도 상호 교접되는 관계가 있다. 그러므로 문제의 핵심은 고성장 조류의 확보와 고성장을 보장할 수 있는 반응기와 이의 운전기술 개발이며, 생성되는 탄화수소를 세포 외부에 집적시키는 Botryococcus braunii의 특성상 후자의 경우에는 반응기내에서 이들 생성물을 얼마나 신속히 세포로부터 제거하여 기질과 광을 신속히 공급되게 하는 것과 광반응기의 일반적인 문제점인 광원 또는 광의 전달 경로에 발생되는 Fouling문제를 제거하는 것이다. 본 연구는 이산화탄소의 저감기술 확보와 동시에 화석연료를 대체할 재생 생물연료 (renewable biofuels)의 생산, 조류의 배양과 동시에 생산물을 회수할 수 있는 동시 추출공정의 개발, 부수적으로 수질정화에 큰장애물이 되고 있는 질소와 인과 같은 영양 염류의 제거와 미래에?이 있다. Ⅲ. 연구개발의 내용 및 범위 1. 사료화 연구 (주관 연구기관) ○ CO2 고정화용 최적 미세조류 종의 도출 및 배양조건 확보. - LNG 연소가스로부터 직접 CO2 고정화 가능한 토속 미세조류 종 분리 - 배양 배지 조성의 최적화 - CO2 고속 고정화 광 생물반응기 개발 ○ 실증 규모 CO2 고정화 공정 (1일 3m3 연소가스 처리) 시스템 개발 - 150L 광 생물반응기의 옥외 시험 - 광 생물반응기의 CO2 고정화 능 시험 - 광 생물반응기의 개선 방안 도출 (온도 조절) ○ 미세조류의 기능성 사료로 활용 기술 개발 - Chlorella KR-1의 영양 성분 분석. - Chlorella KR-1의 소화 효율 향상을 위한 전처리 기술 연구. - 연소가스 고정화 공정에서 생산된 미세조류의 닭에 대한 독성 시험. 2. 연료화 연구 (공동 연구기관) ○ 단위 핵심공정 개발 - 고효율 광반응기 개발 - 분리용 최적 용매 선정 - 독자적 우수 조류 선별 ○ 통합 공정의 완성 - 고농도 조류 배양기술 개발 - 탄화수소 연속 생산 시스템 완성 ○ 탄화수소 연속 생산 공정 최적화 및 상업화 연구 - 연속공정의 최적화 연구 - 질소/인 함유 폐수를 이용한 처리효율 검토 - 파일럿 설계 및 상업화 연구 Ⅳ. 연구개발결과 1. 사료화 연구 ○ 우수 토속 미세조류 종의 확보 연소가스로부터 직접 CO2 고정화가 가능한 우수 토속 미세조류 종을 국내 토양으로부터 분리하였다. 분리 균주의 동정 연구를 통해 Chlorella 종임을 확인하고 Chlorella KR-1으로 명명하였다. 동 균주는 외국의 특허 균주에 비해 연소가스의 주 독성 성분인 SOx와 NOx에 대해 탁월한 내성을 보여 국내외에 특허 출원 및 등록을 마쳐 지적 재산권을 확보하였다. Chlorella KR-1은 현재 LNG 연소가스로부터 CO2를 직접 고정화하는 연구에 활용되고 있다. ○ 고속 CO2 고정화용 광 생물반응기의 개발 및 실증 운전 고속 CO2 고정화용 광 생물반응기를 개발 연구를 수행하여 150L 판형 반응기 (30L/반응기)와 180L 관형 반응기 (13L/반응기) 시스템을 설계 제작하였다. 동반응기는 태양 광을 주 에너지원으로 하는 에너지 절약형 고속 CO2 고정화 시스템으로 순 CO2 제거율 (= 공정의 운전에 따른 CO2 제거 양/ " CO2 발생양)이 3.3으로 공정의 운전에 따른 CO2의 순수 제거율이 높았고 단위 면적당 CO2 고정화 속도는 40g CO2/m2-day로 기존 광 생물 반응기에 비해 1.5배 이상이었다. ○ 클로렐라 KR-1의 기능성 사료 첨가제로 활용 기술 연구 클로렐라 KR-1은 단백질과 불포화 지방산 함량이 높아 기능성 사료로서 활용 가치가 높은 것으로 나타났으며 다만 두꺼운 세포벽으로 인해 소화 효율이 약 60%로 낮은 점이 사료 첨가제로서 활용에 장애 요인으로 밝혀졌다. 검토된 전처리, 가공 기술중 효소 처리(0.01% pancreatin)가 소화 효율을 높이는데 가장 효과적이었으며 소화 효율은 30% 이상 높아졌다. 닭을 모델 가축으로 Chlorella KR-1 배합사료의 급이시 육질의 향상 효과가 있었다. 2. 연료화 연구 - 독자적 우수 균주 선별 - 탄화수소 생산능을 가지는 조류를 담수로부터 국내 최초로 독자개발 성공 - 이상 동시 추출법 - 조류성장 보다 탄화수소 회수율 측면에서 dihexyl ether를 동시분리용 최적 용매로 선정 - 최적조업조건: 지수성장기의 중기에 60 ml dihexyl ether 투여 - 배양액과 유기용매층 사이의 좁은 경계면에서 단지 폭기에 의한 불충분한 교반으로 인해 18∼32%의 낮은 탄화수소 회수율 획득 - 이단 재 순환 추출법 - 탄화수소 회수율이 개선된 이단 재 순환 추출법에서는 조류성장 측면에서 옥탄을 최적용매로 선정 - 이단 재 순환 추출법을 이용한 동시추출을 통해 이상 동시추출법의 2배 이상의 탄화수소 회수율 (57%) 획득 - 회분배양후 후속분리공정으로 이단in일때 6시간 추출을 통해 60% 이상의 탄화수소 회수율 획득 - 후속 분리 공정후 바이오매스의 재 배양을 통해 바이오매스의 재 이용 가능성 확인 - 초기 N/P 농도가 조류성장과 탄화수소 생산에 미치는 영향 - 조류성장과 탄화수소 생산은 질소결핍에 의해 강하게 저해 받음 - 인 결핍을 제외하고는 인은 조류성장과 탄화수소 생산에 거의 독립적임 - 충분한 질소원 공급을 통해 고농도배양 (> 7 g dry biomass/L) 가능 - 축산폐수를 이용한 조류배양 - 12일 회분 배양결과, 8.5 g /L 바이오매스, 0.95 g/L 탄화수소 생산, 2750 mg/L nitrate 제거 - nitrate의 완전제거를 위한 축산폐수의 최적의 희석비는 33% (축산 폐수:지하수=1:2) - 고효율 광반응기 개발 - 순환유동식 광생물반응기를 설계 제작하여 B. braunii를 배양한 결과 기존 최적화된 bubble column 광 생물반응기에서 보다 성장속도가 약 3배 빠르게 나타났다. - 미세조류의 성장 속도는 광도를 13∼114μE/m2·s까지 변화시켜준 결과 광도가 높을수록 성장 속도가 빨라졌다. 이로써 미세조류 반응기에서 광 투과율을 높여주는 연구가 필요함을 알 수 있다. - 실험 기간내 구(bead)가 없는 경우 Biofouling이 배양 3일째부터 발생하여 급진전하였으며 구를 충진한 경우 최하 겉보기 충진율 5%까지도 Biofouling이 발생하지 않았다. - 광생물반응기의 배지로 소화 탈리액을 이용한 결과 Chu13 배지와 유사하여 새로운 적용 분야가 될 수 있으리라 추정되었다. Ⅴ. 연구개발 결과의 활용계획 1. 생물학적 CO2 고정화 공정은 저 비용의 에너지 절약형 CO2 고정화 공정으로써 산업체에서 배출되는 폐기 CO2를 고 부가가치의 유용 물질로 전환시키는 특성을 갖는다. 현재 CO2 처리 기술의 실용화에 가장 큰 장애 요인이 높은 처리 비용인 점을 고려할 때 미세조류를 이용하여 CO2를 고부가가치의 기능성 사료로 활용하려는 본 연구의 목표는 비교적 가까운 시기에 실용화 가능 할 것으로 판단된다. 다만 현재의 연구 결과를 실용화 공정에 적용하기 위해서는 먼저 시범 실증 연구가 필요하다. 현재 실증 연구에 참여할 기업을 물색중이며 본 연구에 참여 의사가 있는 기업을 확보하면 시범 사업을 추진할 계획이다. 2. - 기후변화협약 관련 무역규제 대응 기술로 활용 - 선진국 선도기술의 대등연구 개발 - CO2 저감 신기술 개발 - 질소, 인 제거 신기술로 활용 - 환경친화적 대체에너지 신기술 개발 - 미세조류를 이용한 유용자원 미래기술 개발

Utilization as feedstuff Biological methods, particularly the use of microalgal photosynthesis, have several merits such as mild conditions for CO2 fixation and no requirements for further disposal of trapped CO2. Carbon fixed by microalgae is incorporated into carbohydrates or lipids, therefore, energy, chemicals, or foods can be produced from the algal biomass. The direct use of flue gas reduces the cost of pretreatment yet imposes extreme conditions on the microalgae such as high concentrations of CO2 (10-15%) and the preence of inhibitory compounds like NOx and SOx. The levels of CO2 found in flue gas could be inhibitory to algal growth. The objective of this work is to isolate a suitable algal strain for the direct CO2 fixation from flue gas and the optimization of the culturing condition of the strain. Since the utilization of algal biomass is very important for the biological CO2 fixation process to be economically feasible, the research should be carried out to make the microalgae to be value-added product. 2. Utilization as fuel The aim of this research was to develop the biological CO2 fixation technology as well as the renewabl biofuels-producing technology from the microalgae Botryococcus braunii. In this study, we carried out the development of in situ extraction process, removal process of nitrogen/phosphorus in piggery wastewater, and high performance photobioreactor. Ⅲ. Scope and Contents 1. Utilization as feedstuff ● Isolation of new algal strain and the optimization of culturing condition for direct CO2 fixation from flue gas. - Isolation of algal strain which can fix CO2 from LNG flue gas. - Optimization of the chemical composition of culturing medium - Development of the photobioreactor ● Operation of Bench-scale CO2 fixation system. - Outdoor test operation of 150L photobioreactor. - CO2 fixation experiment from flue gas using the photobioreactor. - Study for the improvement of the photobioreactor. ● Utilization of KR-1 as feedstuff - Determination of the nutritional values of Chlorella KR-1. - Pretreatment for the enhancement of digestibility of KR-1. - Application of KR-1 as feedstuff for poltury. 2. Result and Recommendations 1. Utilization as feedstuff - A suitable algal strain which has high tolerances to CO2 and other toxic compounds like SOx and NOx has been isolated in this work. The tolerances of the strain to SOx and NOx could be greatly enhanced by controlling the culturing conditions. The strain was also successfully applied to fix CO2 directly from actual flue gas emitted from the boilers. - Bench scale photobioreactor systems have been built to culture Chlorella KR-1 in large scale. The reactors have been operated using sun light as an energy source. The CO2 fixation rate of the photobioreactor is 40g CO2/m2-day which is 50% higher than those of the reactors studied in other laboratories. The net CO2 fixation efficiency (defined as "CO2 fixed during the operation/ CO2 generated due to the operation of the system") is 3.3 which indicates that the system is favorable in terms of the CO2 balance. - The protein and lipid contents of Chlorella KR-1 cells cultured with either CO2 enriched air (10%) or LNG flue gas were compared. The digestibility of KR-1 was rather low due to its thick cell wall. Some pretreatment methods to enhance the digestibility are studied. The nutritional quality of a protein in Chlorella KR-1 was also evaluated by analyzing its amino acid profile. The fatty acid content of KR-1 grown with LNG flue gas was also determined. KR-1 has high levels of palmitic acid (16:0) as well as oleic (18:1) and linoleic acids (18:2), which is considered as one of the essential fatty acids. However the digestibility of KR-1 was too low to be used directly. Therefore, it is necessary to carry out research to provide appropriate processing methods to enhance the digestibility. 2. Utilization as fuel ○ The isolation of domestic strains of the green microalgae Domestic strains producing hydrocarbon were isolated. New strains was similar to B. braunii in the morphology of cell and charateristics of produced-hydrocarbon. ○ Two-phase extraction - Two-phase extraction was performed with dihexyl ether in a viewpoint of recovery rather than algal growth. - Optimum operation conditions were the addition of 60 ml dihexyl ether at mid-growth phase in two phase column - The poor recovery (18∼20%) of hydrocarbon from two-phase bubble column seems to be caused by insufficient mixing between two phases. ○ Two-stage cell-recycle extraction - Because dihexyl ether is expensive ($ 230/500 ml) and required more separation stages due to its higher boiling point (228 ℃) than octane (120 ℃), octane was selected as optimum solvent in two-stage cell-recycle extraction culture. - The recovery in two-stage cell-recycle extraction culture was two times higher than that in two-phase extraction culture. - When two-stage cell-recycle extraction was applied to downstream separation after batch culture, the hydrocarbon of over 60 % in the cell suspension was recovered for 6 h at 150 ml/mim of cell-recycle flow rate ○ Effects of initial N/P concentration on algal growth and hydrocarbon production - The algal growth and hydrocarbon production were intensely inhibited by nitrate-deficiency - Initial concentration of phosphate do not significantly affect the algal growth and production yield of hydrocarbon except P-deficient supply. - High cell density (> 7g dry biomss/L) was obtained by sufficient nitrate supply - Algal culture in piggery wastewater - Concentration of biomass and hydrocarbon reached 8.5 g/L and 0.95 g/L, during 12 d. - 2750 mg/L of nitrate could be removed by algal cultivation for 12 day. - Optimum dilution ratio for the removal of whole nitrate in piggery wastewater was 33 % (piggery wastewater : ground water = 1:2) - Development of a high performance photobioreactor - Anti-biofouling photobioreactor was developed in KIST, when B. Braunii was cultured in that, growth rate of it showed about 3 times higher than that of bubble column photobioreactor. - The more light intensity is to photoreactor in a range of 13∼114μ E/m2 ·s, the higher microalgae growth rate is. It showed that to design better photobioreactor, light must be transmitted efficiently to photobioreactor. - In case of photobioreactor developed in KIST without bead, after 3 days culture time, biofouling occur rapidly in wall of the photobioreactor. With bead 5%(V/V), biofouling do not do in all experimental days. - In the event of wastewater after dewatering sludge used as a culture fluid, microalgae growth rate showed somewhat similar to Chu 13 used as a culture fluid. It can be new research field as re-usage of wastewater.
