Ⅱ. 연구개발의 목적 및 필요성
연료전지는 화학반응에서 발생하는 자유에너지를 전기 에너지로 직접 전환시킬 수 있는 장치이다. 현재까지 가장 널리 알려지고 가장 성공적인 연료전지로는 1834년 Grove에 의해 발명된 수소/산소형 전지로서 1960년대에 이르러 미국 유인우주선 계획의 일환으로 중점적으로 연구되고 실용화된 바 있으며 일반적으로 anode에 수소를, cathode에 산소를 사용한다. 이러한 연구 및 응용을 통하여 2000년 현재 세계 각국에서는 alkaline fuel cell (AFC), phospholipic acid fuel cell (PAFC), solid oxide fuel cell (SOFC), solid polymer electrolyte fuel cell (SPEFC), molten carbonate fuel cell (MCFC) 등 다양한 형태의 연료전지를 개발중이거나 상용화 하고있다. 그러나 대부분의 연료전지는 작동시 고온이 필요하며 (e.g. SOFC : 800 - 1000) ℃; AFC, SPEFC: 100℃) 고온 유지를 위해서는 천연 가스나 methane 등이 부가적으로 소비된다.
이상에서 기술한 연료전지는 수소 등의 간단한 화합물은 연료로서 사용할 수 있으나 폐수 중의 복잡한 유기물인 탄수화물, 지질 단백질 등은 일반적인 연소이외에는 복잡한 산화-환원반응을 거쳐 전자가 이동하며 산화하기 때문에 간단한 화학 촉매를 이용하는 연료전지에서 사용하지 못한다. 이러한 유기물을 연료전지의 연료로 사용하기 위해서 biocatalysts 즉 미생물을 도입한 것이 미생물연료전지이다. 미생물을 사용함으로써 각종 복잡한 형태의 유기 화합물을 전지의 연료로서 사용할 수가 있으며 온화한 조건에서 전력생산이 가능하다. 1911년 Potter가 효모 배양액 (Saccharomyces sp.)과 미생물이 존재하지 않는 살균한 배지의 전압 차를 발견한 이후 이 전압 차의 발생원인이 미생물 때문이라는 것이 여러 연구에 의하여 확인되었다. 미생물 연료전지의 다른 장점으로는 미생물의 기질로 사용될 수 있는 다양한 유기물의 미생물에 의한 소비를 통하여 전기를 생산할 수 있다는 점이다. 이후 유기물을 연료로 이용하는 생물연료전지의 개발에 많은 연구가 이루어 졌다. 생물에서 전자전달은 세포 내막에서 이루어지며, 이 내막은 세포벽 등 절연성 구조의 안쪽에 위치하기 때문에 생물의 전자전달 과정에서 직접 전자를 전극에 전달할 수 없다. 따라서 유기물을 산화하여 생장하는 미생물로부터 효과적으로 전자를 전극으로 전달하기 위해 양친매성이며 저분자 형태의 인공 전자전달매개체 (electrochemical mediator)를 사용하여야 미생물 연료전지를 구축할 수 있었다. 생물연료전지는 화학 연료전지와 더불어 우주선에서 발생하는 폐기물을 효율적으로 처리하기 위하여 1965-70년에 걸쳐 미국 NASA에서 집중적으로 연구되었으나 상대적으로 화학 연료전지에 비하여 전력 생산 능력이 낮고 불안정한 출력형태가 얻어진 것으로 나타났다.
1960년대 이후 여러 연구에 의하여 보다 효율적인 형태로 발전해 왔으며 1993년 Allen 등은 포도당을 연료로 하고 Proteus vulgaris를 이용하여 electrochemical mediator (2-hydroxy-1,4-naphthoquinone)를 첨가시 0.4mA의 전류를 5일 이상 안정적으로 생산하는 20ml용량의 생물연료전지를 제작하였다. 여기서 포도당을 전력으로 전환시키기 위해서는 위에서 언급한 electrochemical mediator의 첨가가 필수적이다. 이 mediator를 대체적으로 난분해성으로 2차 오염의 원인이 될 수 있으며 전극에 점착되어 효율을 떨어뜨리는 수 도 있다. 현재까지 생물연료전지의 biocatalysts로 Escherichia coli, Proteus vulgaris, Saccharomyces cerevisiae 등이 이용되었다 이들은 폐수 중의 다양한 유기물을 대사 할 수 있으나, 매개체를 반듯이 사용해야하는 단점이 있다. 따라서 이와 같은 매개체를 사용하지 않아도 전력을 생산 할 수 있는 미생물 연료전지의 개발이 이루어진다면 다양한 유기 폐수를 별도의 전처리 없이 직접 연료로 사용할 수 있을 것으로 판단할 수 있었으나 현재까지의 기술로는 불가능한 것으로 여겨졌다..
최근 혐기성 생태계에서 산소 대신, 제2철을 전자수용체로 이용하여 생장하는 세균이 많이 보고되고 있다. 이들은 물에 녹지 않는 전자수용체[Fe(III)]를 세포 안으로 운반하지 않고 세포 밖에서 환원시키기 위해 세포외막 (outer membrane, OM)에 cytochrome을 보유하고 있는 것으로 보고되었다. 따라서 이와같은 외부의 전자수용체에 직접 전자를 전달하는 미생물을 사용, 미생물 연료전지를 구성하면 별도의 매개체 없이 전지를 운전할 수 있을 것으로 판단하였다.
무매개체 미생물 연료전지의 구성기술은 유기폐수의 자원화를 통한 효과적이며 환경 친화적인 폐수처리공정 기술 개발이 가능하며, 폐수 처리시 발생하는 활성오니의 감소를 유도 할 수 있고, 폐수 처리와 동시에 발생하는 전력으로 인한 에너지 절감효과가 있다. 또한 최초 개발에서 비롯된 기술 선도 및 미생물의 응용범위 확대에 따른 사회의 인식 변화 및 생물산업 활성화가 가능하다. 따라서 보다 다양한 폐수를 이용하여 전력을 생산 할 수 있도록 중의 다양한 유기물을 이용할 수 있는 무매개체 미생물연료전지를 개발하고 이에 대한 구체적인 조사 및 응용 연구를 본 과제의 목적으로 하였다.
Ⅲ. 연구개발의 내용 및 범위
1. 전기 화학적 활성 세균 (electrochemically active bacteria)의 특성 연구
일반적인 유기 폐수에 함유된 복합적인 유기물을 생물연료전지를 통해 산화시키기 위하여 금속염 환원세균인 Shewanella putrifaciens IR-1을 model strain으로 사용하여 전기 화학적, 생리학적 특성조사를 실시하였다.
특히 금속염 환원세균의 전기 화학적 특성을 조사하고 생물연료전지에의 적용성을 검토하였으며 위의 연구를 통해서 습득한 자료를 생물연료전지를 구축하기 위한 기본적인 자료로 활용하였다.
2. 효율적인 생물연료전지 시스템의 구축
전극재료의 변경, channeling이 발생하지 않는 연료전지 구조변경 등 물리화학적 방법을 통하여 효율적으로 전자가 전극으로 전달될 수 있도록 한다. 특히 효율적인 연료전지의 구조개선, 예를 들어 전극-미생물간의 친화력 증가, 효과적인 연료 공급 및 안정적 작동을 위한 기밀설계 등을 통해 구조개선을 실시하였다. 또한 미생물 농도, 전극의 재질 및 넓이 등 기본조건의 변화에 따른 기질 소모량, BOD 감소량, 전력 생산량 등을 조사하여 주어진 조건에서 효율적으로 운전되는 시스템을 구성하였다.
이상의 연구에서 습득한 자료를 바탕으로 하여 미생물 및 연료 등 생물연료전지의 최적화가 유지되도록 세포외적 조건을 computerized하고 적정 가동시간을 조사하고 장시간 가동한 생물연료전지의 기능상 문제점을 보완하였다.
3. 무매개체 미생물연료전지 기술의 응용 범위 확대 및 효율 증대 방안 조사
위에서 습득한 결과를 바탕으로 무매개체 생물 연료전지의 응용범위를 조사하고 발전 효율을 증가시킬 수 있는 방안을 실험적으로 조사하였다. 특히 생활 폐수를 위시한 각종 폐수의 이용성을 확인하고 미생물 연료전지를 이용한 전기 화학적 활성 세균의 농화 배양 및 기질 특이성, 전극에 대한 부착상태, 폐수처리능력, 전극에서의 미생물 분리 및 동정 등을 시행하였다..
Ⅳ. 연구개발결과
1. 금속염 환원 세균에 대한 기본특성조사 및 기본 연료전지 구축
Cyclic voltammetry를 통한 전기화학적 활성세균의 전기 화학 활성도 조사법 확립을 확립하였고 기본형 생물연료전지의 제작을 완성, 0.65 V, 0.1mA의 전기가 생산됨을 확인하였다.
2. 효율적인 생물연료전지 시스템의 구축
생물 연료전지 시스템을 이용한 전혀 새로운 전기 화학적 활성 세균의 농화배양법 개발을 개발완료 하였고 이를 이용, 다양한 기질 자화 능력을 갖는 미생물을 연료전지 내에서 농화배양하여 전기를 생산하였다. 또한 이 기술을 응용하여 다양한 폐수를 연료로 전기를 생산하는 무매개체 생물연료전지를 개발하였다.
3. 시스템 최적화 및 다양한 폐수를 직접 연료로 사용하는 생물 연료전지의 개발
생물 연료전지의 구조개선을 통한 최적운전조건을 확인하였다 또한 전기 화학적 농화배양시 전극에 부착되는 초미세세균 (nano bacteria)를 조사하였다. 생물 연료전지의 연속 작동 실험 및 다양한 폐수를 연료로 적용, 생물연료전지의 폐수 처리능 실험을 수행하였고 이를 바탕으로 다양한 분야에 적용할 수 있는 무매개체 생물 연료전지를 원천기술로서 개발하였다.
Ⅴ. 연구개발결과의 활용계획
1. 새로운 에너지 생산형 대규모 폐수처리장치의 개발
Scale-up 과정등 후속 연구를 통한 새로운 에너지 생산형 폐수 처리 system의 개발

연구기간 중 접촉한 미국, 일본, 영국등의 연구기관과 공동으로 본 연구에서 부족한 부분 에 대한 정보를 교환하고 전극, 이온교환막등, 각 부품의 국산화 시도 및 공동 개발

In the present study, construction and application of a novel mediator-less microbial fuel cell using electrochemically active bacteria were investigated. Direct electron transfer from different Shewanella putrefaciens strains and its mutants to an electrode was examined.
In the cyclic voltammetry, anaerobically grown cells of S. putrefaciens strains showed electrochemical activities. Microbial fuel cells were constructed using the S. putrefaciens strains. The concentration of lactate as the electron donor in the anode compartment determined the current generation capacity and potential development in the microbial fuel cell. When the high concentration of the bacteria (0.47g dry cell weight / l) and an electrode that has large surface area (apparent area: 50 cm2) were used, relatively high Coulombic yield (over 3 C for 12 h) was obtained
Using a three-electrode cell the strain IR-1 was able to cultivate. In the cultivation system, the positively poised working electrode worked as an artificial electron acceptor.
On the basis of these results, the microbial fuel cell was used to enrich electrochemically active microbes using wastewater from a starch processing factory as the electron donor with activated sludge or sludge of the anaerobic digester as the bacterial source. Within 4 weeks a current of 0.2 mA was generated with a resistance of 1k in sequential-batch operation mode. The COD of anode reaction mixture treated with anaerobic sludge decreased from 1942 ppm to 55 ppm after a month. Coulomb has been maintained almostly 10 Coulomb(C). The COD change and Coulomb production with aerobic sludge were 1100 ppm to 69 ppm and 20 Coulomb, respectively. Starch processing wastewater treatment was tested after enrichment of electrochemical active bacteria. The COD and TOC removal was more than 95% and 87%, respectively. The microbial fuel cell performed optimally at pH 7 -8, and the optimum temperature was 37C. Current generation was influenced by the presence of respiration inhibitors. The Uncoupler (DNP) and ATPase inhibitor (DCCD) showed the inhibition of current production. P-CMPS (Fe-S cluster inhibitor), HQNO(Cytochrome b site inhibitor) and Antimycin A (electron transport chain II inhibitor) also decreased the current generation. However, the terminal oxidase inhibitors, cyanide and azide did not inhibited.
Based on the previous experimental data the microbial fuel cell format was revised to decrease the channelling effect and contact resistance between electrode and the connection lead. Several attempts were made to overcome those problems, and the application of electron collector was selected for the construction of the microbial fuel cell. A 17 ml of newly constructed cell and an artificial wastewater were used for the further experiment. Various operational parameter such as different feeding rate of fuel and resistance were examined and the fuel cell system was optimized.
The anode from the enriched fuel was also examined microscopically. Low vacuum electron microscope showed bacterial clumps loosely associated with the electrode and biofilm onto the electrode. Scanning electron microscopic observation of the biofilm revealed particle of ultramicrobacterial size as well as of normal bacterial size. The smaller particles were found to possess bilayer membrane structure by transmission electron microscope. Confocal scanning laser microscopic observation of the enriched electrode showed that Gram negative and positive bacteria formed microcolonies throughout the bulk and biofilm at all depths.
When the bacteria concentration of the anode was analysed by the conventional colony count method, the biomass enriched on the electrode was 4 orders of magnitude less than that predicted by chemical analyses of the enriched electrode. Chemical analyses of the enriched electrode showed that one gram of the electrode generated biomass containing 131.759.8mg protein and 4.330.24mg DNA. These figures were used to calculate the number of bacteria present as 6.34 0.6 x 1011/g electrode assuming that the bacterial cells contain 55% protein and 1.8% DNA. Most of the Fe(III)-reducing isolates were electrochemically active. Based on these findings it was hypothesized that electrochemically active bacteria are wide spread, and that their electrochemical activity is related to a form of anaerobic respiration.
An obligately anaerobic bacterium designated as EG3 was isolated from a microbial fuel cell. The isolate was Gram-positive, motile and rod (2.8-3.0㎛ long, 0.5-0.6㎛wide). The partial 16S rRNA gene sequencing and analysis of the fatty acid profile suggested that EG3 clustered with Clostridium sub-phylum and exhibited the highest similarity (98%) with Clostridium butyricum. The temperature and pH optima for growth were 37℃ and 6.8-7.4, respectively. Clostridium butyricum EG3 grew in phosphate buffered basal medium (PBBM) with glucose as a sole electron donor. Other simple carbohydrates were also used by the isolate. Growth was faster when the medium was supplemented with FeOOH than without FeOOH at the initial phase, but the final cell yield was similar. These results suggest that Fe(III) ion is utilized as an electron sink. The major products of glucose and glucose/FeOOH metabolism were lactate, formate, butyrate, acetate, CO2, H2. Cyclic voltammetry showed that Clostridium butyricum EG3 cells were electrochemically active. The fuel cell was able to generate the current of 0.21mA and 54 Coulomb at the glucose consumption of 10mM. It is a novel characteristic of strict anaerobic gram-positive bacterium.
