Pl3K는 IGF 뿐만 아니라, EGF, PDGF, HGF, neurotrophin등 여러종류의 성장 인자에 의해 공통적으로 활성화되는 것으로도 알려져 있어 세포증식의 중심적 신호의 역할을 한다. 전년도에 본 연구실에서 수행한 결과에서 밝힌TCDD가 IGF 수용체의 발현감소는 곧 Pl3K의 활성변화와 그에 따른 세포증식 및 세포사멸 억제 신호로 작용할 것이라고 추정하고 본 과제에서는 TCDD에 의한 Pl3K 및 그 upstream의 IRS-1의 활성변화를 조사하였다.

In this study, we have investigated molecular bases of 2,3,7,8-tetrachlorodibenzo-p-dioxin (TCDD)-induced inhibition of neural cell proliferation using SK-N-SH human neuroblastoma cells as a model cellular system. We measured whether TCDD altersphosphorylation status of retinoblastoma (Rb) protein since Rb protein is permanently present in the cells no matter what cellsare in Go or cycling and its phosphorylated form releases gene regulatory proteins that favor cell proliferation. TCDDdecreased the phosphorylation of Rb in a time-dependent manner. The phosphorylation status of Rb is regulated by various cell cycleinhibitors. p53, one of the inhibitor, was not altered by TCDD either in mRNA nor protein level. In a consequence, p21 which is a down stream target of p53 was not changed. However, the level of p27, a cell cycle inhibitor was significantly increased, indicatingthat p27 plays an important role in decreased proliferation of neuronal cells by TCDD. In addition, the phosphorylation of insulin receptor substrate (IRS), a IGF-lR downstream signaling molecule, was increased by the treatment with TCDD. Also, TCDD increased Pl3 Kinase activity, which is known to phosphorylate Akt, an apoptosis inhibitor. The results indicate that inhibition of neuronal cell proliferation by TCDD is possibly mediated through increased induction of p27 not by p53 and that inhibition of cell death by TCDD may be due to activation of Pl3 Kinase.
