고분자를 혈액적합성 재료로 사용할 경우 필연적으로 야기되는 재료 표면 위에서의 혈장단백질의 흡착과 이로부터 유도되는 혈소판의 점착을 억제하기 위하여, 본 연구에서는 비수용성 친수성/소수성/이온성 삼성분계 공중합체를 합성한 후 이를 기존의 고분자에 대한 코팅제, 첨가제 아울러 공중합체의 형태로 적용하여 각각의 혈액적합성을 평가하고자,
① 새로운 항응혈성재료의 합성 및 벌크/표면 특성분석
② 신 재료의 코팅제 및 첨가제로서의 응용연구
③ 초기 체외 혈액적합성 평가를 통한 재료/표면의 선정 과정을 수행하여 다음과 같

③ 초기 체외 혈액적합성 평가를 통한 재료/표면의 선정 과정을 수행하여 다음과 같은 결과를 얻었다.
가) Molecular cilia 개념(PEO)과 음이온 표면(SO₃)의 복합된 Negative cilia 모델과 Phosphorylcholine polar
group을 함유한 생체유사막 모델을 복합시켜 인지질함유 Negative cilia형의 신개념 혈액적합성 공중합체를 (친
수성 block에 (술폰산화) PEO (metha)acrylate를, 소수성 block에는 octadecylmethacrylate, 이온성 block에는
methacylolyloxyethyl phosphorylcholine)를 최적 합성하고 이들 공중합체의 합성분석과 특성평가를 분광학적 방
법 및 여러 가지 분석방법을 이용하여 수행하였다.
나) 공중합체의 생물학적 특성평가위해 혈소판 점착 실험과 혈장 단백질 흡착, 그리고 박테리아 점착 등의 in-
vitro 실험을 통해 체외 혈액적합성을 평가하였고 체내 기능평가를 위해 A-A rabbit 모델을 이용하여 폐색시간
(occlusion time)을 측정하였다. 그 결과 새로운 혈액적합성 MPC/PEG1KMA(PEG1K-SO₃MA)/OMA의 친수성/소수성 공
중합체를 개발하였고 공중합체의 기존 의료용품 및 인공장기에 코팅제 및 가공첨가제로의 응용 가능성이 확인되
었으며 체내외 혈액적합성 평가를 통하여 이들 표면의 우수한 혈전 형성 억제 효과를 확인하였다.
다) 비닐계 단량체들로부터 소수성인 methy1 metyl methacrylate (MMA), 친수성인 methoxy polyethylene oxide
monomethacrylate (MPEOMA; PEO 분자량, 1000), 음이온성기를 가지는 vinyl sulfonic acid sodium salt (VSA)를
선정하여 이들로부터 PEO 사슬과 음이온성기를 동시에 곁가지 사슬로 가지는 MMA/MPEOMA/VSA 삼성분계 공중합체
를 합성하고, 표면특성과 혈액적합성을 MMA/MPEOMA 혹은 MMA/VSA 공중합체와 비교하였으며, 이 공중합체를
polyurethane (PU) 튜브에 도포하여 소구경 인공혈관으로써의 가능성을 타진하여 보았다.
라) 합성한 공중합체들을 도포한 PU 표면의 물접촉각을 측정한 결과, 공중합체 내 MPEOMA 조성이 증가할수록 PU 표
면에 도포된 공중합체의 물접촉각이 감소함을 관찰할 수 있었고, Zeta-potential 측정 결과 VSA 단량체가 증가할
수록 공중합체의 표면의 음이온성이 증가함을 관찰할 수 있었다. 공중합체 도포 표면의 in vitro 혈액적합성 실
험 결과 MMA/MPEOMA/VSA (8/1/1) 공중합체가 혈장단백질 흡착과 혈소판 점착 억제에 매우 효과적이어서 혈액적합
성이 상당히 우수하다고 판단되었다.
마) 인공혈관용 PU 튜브 (diameter, 5mm)를 본 연구실에서 직접 제조한 튜브제조장치를 이용하여 제조하였으며, 공
중합체의 튜브 내부 도포에도 동일한 방법을 이용하였다. 제조한 PU 인공혈관의 물성 평가를 위해, 컴플라이언
스(compliance) 측정장치 또한 본 연구실에서 직접 고안 제작하였다. 합성된 공중합체는 PU 튜브의 내부에 균일
하게 도포됨을 관찰하였으며, 도포된 공중합체는 PBS 내에서 매우 안정함을 관찰하였다. 인공혈관을 제조하여
ex vivo 동물실험(A-A shunt)을 수행하였으며, in vitro 실험결과와 마찬가지로 MMA/MPEOMA/VSA (8/1/1) 공중합
체가 도포된 PU tube가 혈액적합성이 매우 우수함을 관찰하였다. 이로써 MMA/MPEOMA/VSA 공중합체는 혈액적합성
이 요구되는 수많은 의료용품 및 기구에 효율적으로 사용할 수 있을 것으로 판단된다.
바) 다당류를 포함하는 생체유사막 표면인 trisystem Ⅰ(poly(HBA choline-co-VM7A-co-DEAEMA)과 trisystem Ⅱ
(HBA choline-co-VM7A-co-MMA))의 공중합체를 합성하였고, 이들의 표면과 벌크 특성을 비교하여, 친수성 부분
과 소수성 부분의 이론적인 molar monomer composition과 실제 합성된 중합체에서의 monomer composition이 거
의 일치함과 합성된 공중합체들이 모두 물에 어느 정도 이상의 안전성을 나타내고 있음을 알 수 있었다.
사) 공중합체의 분자량은 기존의 trisystem Ⅰ에서의 15,000 전후의 분자량에서 trisystem Ⅱ는 70,000으로서의 증
가를 나타내었고 표면 특성에서는 소수성 아크릴계DEAEMA나 MMA가 상당량 도입되고 적은 분율의 친수성 VM7A가
도입됨에도 불구하고 본 공중합체들은 극도의 친수화를 나타내고 있음으로 보아, comb-like 타입의 본 공중합체
에서 water phase에서 long chain을 가진 VM7A가 노출되었음에 기인하고 있다.
아) in vitro 혈액적합성 평가에서는 공중합체에서의 HBA-choline과 VM7A의 몰비가 증가함에 따라 혈소판 점착이 억
제되어 우수한 혈액적합성을 보여 주었고, 혈소판의 활성화도 억제됨을 알 수 있었다.
자) 개를 모델로 하여 in vivo 실험에서는 이식된 PU 그라프트가 완벽한 개통을 보이고 있었으며 실험에 사용된 개
는 2주까지 생존하였으며, 본 공중합체들이 가진 양친매성으로 인해 고분자코팅 재료뿐 아니라 기계적 물성의
보강으로 인하여 혈액 적합성 재료로도 이용할 수 있다.

New blood compatible polymeric materials have been developed for blood contacting devices. These copolymers consist of hydrophilic poly(ethylene glycol)/sulfonated poly(ethylene glycol) (metha)acrylate, hydrophobic components of octadecyl (metha) acrylate and phosphatidyl cholin moiety, which have been prepared by conventional radical polymerization. The structures of synthesized copolymers were confirmed by ????H-NMR, ????C-NMR and elemental analysis. Physical results obtained from viscosity measurement and thermal analysis demonstrated the characteristic of new copolymers. Modified surface properties have been characterized by water contact angle measurement and ESCA analysis.
Biological responses on the obtained copolymers have been evaluated by measuring platelet adhesion, protein adsorption, bacterial attachment in-vitro. In-vivo rabbit A-A shunt experiment have been performed to evaluate the whole blood compatibility using copolymer coated shunts. In vitro platelet adhesion, plasma protein adsorption and bacterial attachment demonstrated MPC modified surfaces reduced in-vitro protein, platelet and bacterial attachmen as compared with control surfaces. In-vivo occlusion time with MPC copolymer extended longer than that of control PU surface, which is well correlated with in-vitro low platelet adhesion, protein absorption and bacterial attachment.
The results attest to the usefulness of these new materials as surface modifying agents such as a coating or additive to improve the blood compatibility of blood contacting materials. New water-insoluble copolymers were synthesized by random copolymerization of methyl methacrylate (MMA), methoxy polyethylene oxide monomethacrylate (MPEOMA; PEO mol. wt., 1000), and vinyl sulfonic acid sodium salt (VSA) monomers with different monomer composition. The synthesized MMA/MPEOMA/VSA copolymers were characterized by 1H-nuclear magnetic resonance spectroscopy and gel permeation chromatography.
They were studied as possible means to produce blood-compatible surfaces by spin coating onto polyurethane (PU) or polymethyl methacrylate (PMMA) substrates (films or tube). The surface properties of MMA/MPEOMA/VSA copolymers were compared with those of MMA/MPEOMA or MMA/VSA copolymers with similar MPEOMA or VSA composition, by the measurements of water contact angle and zeta potential. For in vitro blood compatibility evaluations, the behavior of the adsorption of blood proteins (albumin, a-globulin, fibrinogen, and plasma proteins) and platelet adhesion on the copolymer-coated surfaces was compared by electron spectroscopy for chemical analysis and scanning electron microscopy, respectively. It was observed that the MMA/MPEOMA/VSA copolymer with monomer molar ratio 8:1:1 was particularly effective in preventing both protein adsorption and platelet adhesion on the surfaces. Same results were also observed from ex vivo animal study (A-A shunt using dogs). The MMA/MPEOMA/VSA (8/1/1) copolymer-coated PU thbe (diameter, 5mm) was particularly effective in preventing platelet adhesion on the surfaces. probably owing to the combined effects of highly mobile PEO and negatively charged group in aqueous solution. The MMA/MPEOMA/VSA copolymer can be used as a blood-compatible coating material applicable to various blood-contacting devices as well as vascular grafts.
The insertion of the material would bring the formation of thrombogenity and the polymer is no exception. So, when the artificial blood vessel is inserted into the body, the coagulation occurs, which prohibits the artificial blood vessel to function. To solve this problem, several methods had been studied, like the use of modified surface or use of blood vessel-like materials. Recent studies have indicated that oligosaccharide and phospholipid derivated materials independently can function as excellent hemocompatible materials.
Hydrophilic oligosaccharide, hydrophobic phospholipid and DEAEMA, polycaprolactone and MMA were copolymerized to synthesize the natural-like blood vessel. With this material, the blood compatibility was evaluated to see whether this material is adequate to apply as artificial blood vessel.
The polymerization was done using DMF and DMSO respectively to see how the outcoming polymers are different. The copolymers chemical structure was confirmed by NMR, DSC and IR. The sample surfaces were characterized with AFM and DCA. AFM were used to obsrved the morphology difference between the hydrated and unhydrated surface and DCA to see hydrophilicity fo the samples. The samples were also tested in several different solvents including water to confirm their water solubility.
The human platelets were used for in vitro blood compatibility test above sample films. Right after drying in the oven, the samples were hydrated. Then incubated platelets were added and left for 2 hours in the incubator. After that, the samples were rinsed with PBS and then immobilized with 2.5% glutaraldehyde for a while. All the samples were washed with PBS and dehydrated using ethyl alcohol series (50%∼100%) for the SEM observation. In vivo, we have prepared two kinds of vascular grafts, 3mm in diameter. These prostheses placed in canine femoral vein and detected blood pressure in the graft. Phospholipid and oligosaccharide is very interesting materials and these copolymers have to studied further to synthesize a perfect hemocompatibility materials.
