인삼 사포닌의 작용 기전은 세포내 작용 부위에 따라 크게 2가지로 나누어 생각해 볼 수 있다. 그 하나는 세포막에 있는 각종 이온 통로나 세포막 수용체에 영향을 미치는 것이다. 다른 하나는, 인삼 사포닌은 지용성 물질이므로 핵내까지 단순 확산에 의해 자유롭게 통과하여 핵내의 각종 수용체나 전사 활성 단백질과 결합하여 효과를 나타내는 것이다. 인삼 사포닌은 구조적 유사성으로 인해 핵내 수용체중에서 특히 스테로이드 호르몬 수용체와 결합하여 스레로이드 호르몬 활성을 나타낼 것으로 기대하고 있으며, 이 현상이 스테로이드 호르몬 수용체에 대

본 연구팀은 인삼 사포닌에서 여성호르몬 작용을 기진 호르몬 성분을 검색하고자 유방암세포주인 MCF극 세포주에 에스트로젠에 의해 유도되는 리포터 유전자를 일시적으로 트랜스팩션하여 루시퍼라아제 효소의 활성을 측정하는 방법으로 호르몬 활성을 검색하였다. 에스트로젠 활성 효과는 고려 인삼의 총사포닌에서 뚜렸하게 나타났으며, 실제 이러한 결과는 미국 인삼의 총사포닌에서의 결과와는 대조되는 것으로 고려인삼의 우수성을 의미하는 것이다. 본 연구에서 조사한 것중에서 인삼의 단일 성분에서 에스트로젠 활성을 가장 강하게 나타낸 진센노사이드 Rbl에 대해서는 에스트로젠에 의해 유도되는 원암 유전자인 c-fos, autoregulation되는 에스트로젠 수용체 등에 대해 mRNA와 단백질 수준에서 분석하였다. 진센노사이드 dbl을 포함한 다른 진센노사이드에 대해 에스트로젠 활성을 보이지 않은 경우에도 유방암 세포주를 포함한 다른 암 세포에서 세포 성장의 추이를 관찰하였다. 진센노사이드 Rh2에서 유방암 세포 살상 효과가 가장 크게 나타났으며, 진센노사이드 Rh2의 경우 세포 살상 효과가 탁월하였다. 본 연구에서 조사한 ginsenoside Re, Rc, Rh2, Rf, Rbl에서는 스테로이드 호르몬의 다른 종류인 레티노익산, 안드로젠, 글루코코티코이드등의 활성은 전혀 나타나지 않았다. 이는 식물성 호르몬 제제의 내재적인 문제점의 하나로 지적할 수 있는 특이성 측면에서 다른 스테로이드 호르몬에 대한 cross reactivity는 배재할 수 있음을 의미한다.
인삼 사포님이 스테로이드와 유사한 작용을 나타낸다는 예측은 되고 있으나, 이에대한 확실한 연구결과는 아직 부족한 실정이다. 따라서 고려인삼의 총사포닌과 진센노사이드-Rbl이 에스트로젠 효능을 나타내고, 진센노사이드 ah2가 유방암 세포에 대한 억제 효과가 있음을 밝힌 본 연구 결과는 부작용이 큰 문제가 되고 있는 현 상황에서 여성 호르몬 제제의 대체 물질의 개발에 획기적인 전기가 될 것이고 이를 통하여 진센노사이드의 임상적 응용에 있어 과학적인 근거를 제공할 것으로 기대된다.

Ginseng is a popular herbal medicine that has been used for eve. 2,000 years in oriental countries. It has been reported that ginseng has a wide range of pharmacological activities in cardiovascular, endocrine, immune, and central nervous systems. Ginseng and its constituents have been used for their tonic, immunomodulatory adptogenic, anticancer, and anti-aging activities. These beneficial effects of ginseng have prompted tremendous effort to discover the pharmacology of its action through biochemical and molecular biology techniques.
Ginsenosides could exercise their activities at either the cellular membrane, inside the cell or evert in the nucleus. Ginsenosides may initiate their mode of action by binding to transmembrane membrane receptors and subsequently activating the associated downstream signaling pathways. Due to the amphiphilic nature of ginsenosides, ginsenosides might even intercalate into the cell membrane, thereby influencing membrane fluidity. It is also possible that the hydrophobic properties of ginsenosides favor their binding to intracellular steroid hormone receptors, such as receptors for glucocorticoids, progesterone, androgens, mineralocorticoid, and estrogen. Like steroids, ginsenosides are lipophilic], which enhancesl their ability to traverse cell membranes by simple diffusion and acquire access to the nucleus to control gene transcription by binding to specific intracellular receptors. A few studies shelved several biological effects resulting from steroid hormone receptor activation. Ginsengl has been recommended for the alleviation of the symptoms of menopause, indicating that some components of ginseng act as phytoestrogens and/or involve activation of the estrogen receptor Ginseng extracts are able to stimulate the growth of ER-Positive cells. However, there has been no conclusive scientific data to show that ghseng contains phytoestrogens. In this report, we show that total saponin and ginsenoside-Rh1 is an active estrogenic ligand as determined by measuring the transcription activation of ant estrogen-responsive reporter plasmid and an endogenous estrogen responsive gene. In addition, we have found that ginsenoside-Rh2 is extreml powerful in killin MCP-7 breast cancer cells. Our results will provide scientific basis for use of ginseng as phytoestrogen in various estrogen-related diseases such as breast cancer, osteoporosis, and postmenopausal symptoms.
