MCP-1은 또한 수용체인 chemokine receptor2(CCR2)과 상호작용함으로써 죽상경화를 발전시킨다. 연구목적 및 연구방법 본 연구에서는 MCP-1에 의해 유도되는 신혈관생성의 기전을 in vitro (내피세포의 증식), ex vivo(백서 동맥환 분석), in vivo (chick CAM assay) 실험을 통하여 알아보자 하였다. 또한, MCP-1과 CCR2의 결합을 특이적으로 저해하는 저해제를 개발하기 위하여 random phage display peptide library에서 peptide를 선택하여 automatic DNA sequencer로 시퀀싱하였다. 연구결과 내피세포의 증식과 백서 동맥환 분석, CAM assay 결과 MCP-1이 신혈관생성을 유도함을 확인하였다. CAM assay 결과 MCP-1에 의한 혈관생성능력은 강력한 신혈관생성 요인인 VEGF (VEGF-$A_{189}$)와 VEGF-$C_{388}$의 mRNA 발현을 증가시켰고 혈과내피세포와 단핵구에서 VEGF의 분비를 증가시켰다. 또한, CAM assay에서 MCP-1에 의한 신혈관생성은 VEGF 저해제인 $Flt_{2-11}$에 의하여 완전히 억제되었다. Rho protein 저해제인 Clostridium difficile toxin B와 RhoA를 특이적으로 저해하는 C3 transferase 또한 MCP-1에 의한 신혈관생성을 완전히 차단하였다. HMG-CoA reductase 저해제인 simvastatin은 MCP-1의 신혈관생성능을 완전히 차단하였고 VEGF의 mRNA 발현도 저해하였다. PPARv 활성제인 rosiglitazone은 MCP-1에 의한 신혈관생성을 어느 정도 저해하였고, 반면 PPARα 활성제인 fenofibrate는 내피세포의 sprouting을 증가시키는 결과를 보였다. 사람 동맥 내피세포(human aortic endothelial cells; HAECs)에서 CCR2가 발현됨을 확인하였다. HAECs를 주기적으로 기계적 스트레스를 주었을 때 (24%, 1㎐ for 30 minutes to 24 hours)CCR2 발현이 유의적으로 증가하였다. MCP-1에 결합하는 피아지 클론을 선택하고 DNA 염기서열을 분석하여 펩타이드 서열을 유추한 결과 CCR2와 homology를 가짐을 확인하였다. 선택한 펩타이드 서열 중 일부를 합성하여 MCP-1과의 결합을 확인하였으며, flow cytometry endothelial cell migratin assay 결과 합성 펩타이드는 MCP-1의 chemotatic activity를 저해하였다. rat aortic ring assay와 CAM assay 결과 합성 펩타이드는 MCP-1에 의한 신혈관생성을 억제하였다. 결론 MCP-1에 의한 신혈관생성은 VEGF에 의존적이며, small GTPase, 특히 RhoA를 매개로 하는 신호전달체계를 가지는 것으로 보인다. 또한, 본 연구 결과 선택된 MCP-1 결합 펩타이드는 MCP-1과 CCR2의 네트워크를 차단함으로써 죽상경화를 예방하는 새로운 물질이 될 것으로 사료된다.

BACKGROUND Angiognesis is implicated in the growth of atheromatous plaque. Together with ether chemokines such as IL-8 and SDF-1, a CC chemokine monocyte chemoattractant protein-1 (MCP-1) is expressed in the atherosclerotic lesion and reportedly play a critical role in the regulation of angiogenesis in addition to their functions in leukocyte vascular trafficking and activation. MCP-1 plays a crucial role in the development of atherosclerosis by interacting with it's exclusive receptor CC chemokine receptor 2(CCR2). PURPOSE and METHODS In this study we investigated the mechanism of MCP-1 induced angiogenesis using in vitro (endothelial cell proliferation assay), ex vivo (rat aortic ring assay) and in vivo (chick CAM assay) angigenesis models Furthermore, to develop a specific inhibitor for the MCP-1 binding to CCR2, we used reiterated three independent selections employing random phage display peptide library. The peptide that bind to MCP-1 sequenced by automatic DNA sequencer. RESULTS The result of endothelial cell proliferation assay, rat aortic ring assay and CAM assay showed that MCP-1 induced angiogenesis. The estimated number of newly-formed vessel induced by MCP-1 in the CAM was comparable to that of induced by human recombinant VEGF, indicating that MCP-1 is at potent angiogenic factor. MCP-1 increased the mRNA amount of VECF (VEGF-$A_{189}$) and VEGF cytokine family, VEGF-$C_{388}$ in the cultured rat aortic rings and the VEGF secretion in cultured human aortic endothelial cells (hAECs) and THP-1 monocytes. Accordingly, the CAM assay showed that the MCP-1-induced angiogenesis was completely suppressed by VEGF-inhibitor $Flt_{2-11}$. A Rho protein inhibitor, Clostridium difficile toxin B, and a specific RhoA inhibitor C3 transferase also abolished the MCP-1-induced angiogenesis. Pretreatment of excised rat aortic rings and CAM with HMG-CoA reductase inhibitor, simvastatin, completely blocked the angiogeneic property of MCP-1 and decreased mRNA amounts cf VEGFs. The PPARv ligand rosiglitazone showed moderate inhibitory effects on the MCF-1 induced angiogenesis, while the PPARαa activator fenofibrate enhanced tile endothelial cell sprouting. The result of RT-PCR showed that HAECs express the CCR2. When HAECs were subjected to cyclic mechanical stretch (24%, 1 ㎐ for 30 minutes to 24 hours) using the computer-controlled Flexcell Strain system, the amount of CCR2 transcripts was significantly increased.Sequences of the selected peptides were found to have homology to CCR2, which was MCP-1 interacting domain in CCR2. It was also found that sequences of selected peptide have homology to CCR3. which was a well-known antagonist of MCP-1, Peptide was synthesized and the binding affinity of peptide to the MCP-1 was measured by the Surface Plasmon Resonance technique. Flow cytometry and the endothelial cell migration assay showed that the MCP-1 binding to CCR2 and chemotatic response was inhibited by the peptide. Furthermore, the peptide effectively inhibited the angiogenic activity of MCP-1 as shown bi the CAM assay. CONCLUSION Neovasculariztion induced by MCP-1 dependent on signaling pathways involve VEGF and small GTPase, most notably RhoA. In addition, our selected MCP-1 binding peptide was hoped to be developed as a lead molecule for prevention atherosclerosis by inhibiting MCP-/CCR2 network.
