목적; 본 여구과제의 최종목표는 NK 세포의 세포독성능 제어를 통한 발암기전 규명과 암 치료 및 예방법 개발의 모색에 있다. 본 여구를 통하여 NK 세포의 세포성 면역반응이 어떻게 조절되어 표적특이성이 결정되는가를 규명하고자 하였고, 암세포들은 또한 어떻게 NK 세포의 면역감시기능을 피해가고 있는지를 파악하여 암세포의 발암기전에 대한 정보를 얻고자 하였다. 이러한 정보를 역이용하면 NK 세포의 면역반응을 숙주에 유리한 방향으로 조절할 수 있는 방안을 모색하는데 많은 도움이 될 것이고 NKd세포를 이용한 항함요법개발에 활용할 수 있을 것이다.방법 : 본 연구목표를 효과적으로 달성하기 위하여 우리는 두 개의 연구팀을 구성하여 상호 유기적이고 보완적인 관계 속에서 연구를 진행하였다. 제 1 세부과제는 NK 세포-종양세포 상호작용의 분자기전 연구로서 분리된 NK 세포를 이용하여 분자세포 생물학 및 분자세포 면역학적인 접근방법을 통해 수행 되었고, 제 2 세부과제는 NK 세포의 활성화 및 비활성화 신호전달 기전 연구로서 NK 비활성화 수용체를 대장균 및 T 세포에 발현하여 생화학, 분자생물학, 및 세포생물학적인 접근방법을 통해 수행되었다.결과 : 제 1세부과제에서는 말초혈액으로부터 분리한 NK 세포와 leukernia에 기원된 NK cell line을 이용하여 이들 NK 세포에 표현되어 있는 receptor들의 표현형을 분석하였다. 또한 각종 종양세포들에 표현되어 있는 MHC type 및 Fas 등을 분석하였다. NK 세포에 lsrka한 세포주와 둔감한 세포주사이에 이들 molecule들의 표현에 어떠한 차이가 있는가를 분석하고, NK 세포가 종양세포를 제거하는 기전을 규명함으로써 NK cell과 종양세포사이의 관계를 분자레벨에서 이해하고 이들의 발암기전을 이해하는 데이터를 구축하였다. 흥미롭게도 NK 세포는 백혈병 세포주들 뿐망 아니라 갑상선암이나 간암세포주들과 같은 조직에서 유래된 암세포들도 효과적으로 제거할 수 있는 것으로 나타났고, 이 때 perforin/granzyme을 통한 세포괴사 기전 뿐만 아니라 death receptor/ligand 상호작용을 통한 세포고사 기전도 중요한 역할을 하는 것으로 나타났다. 이러한 결과는 NK 세포를 이용한 항함요법 개발에 밑거름이 될 것이다. 제 2 세부과제에서는 NK cell의 cytotoxicity를 조절하는데 핵심적인 역할을 하는 KIR에 결합하는 signaling molecule들을 확인함으로써 KIR을 통해 어떠한 signal이 전달될 n 있는 지를 상당부분 밝혀내었고, 흥미롭게도 KIR가 T 세포에서 AICD를 억제하는 기능이 있다는 사실을 추가로 밝혀내었다. 또한 KIR의 면역학적인 기능 (cytoxicity 억제 기능과 AICD 억제기능)에 대하여 좀더 구체적인 signal transduction pathway를 규명하였고 이를 바탕으로 면역반응을 원하는 방향으로 조절할 수 있는 가능성을 제시하였다. 이러한 연구결과를 잘 활용하면 NK 세포 및 세포독성 T 세포가 관여하는 세포성 면역반응을 숙주에게 이로운 방향으로 조절할 수 있는 선택적인 면역제어기술을 고안할 수 있을 것이다.연구결과의 활용계획 : 본 연구를 통해 얻은 성과는 NK 세포를 이용한 항암요법 개발의 이론적 배경이 될 수 있으며, 특히 NK 세포의 분리, 배양, 및 활성화방법은 NK 세포를 이용한 항암요법 개발에 직접적으로 활용될 수 있을 것이다. 또한 본 연구를 통해 얻은 NK 세포 비활성화 수용체의 면역학적 기능 및 조절기전에 대한 정보는 NK 세포 및 세포독성 T 세포가 관여하는 세포성 면역반응을 숙주에게 이로운 방향으로 조절할 수 있는 선택적인 면역제어기술을 고안하는데 활용할 수 있을 것이다.

Purposes : The final aim of this study was to understand the molecular mechanism of cancer development and to approach cancer therapy by regulation of NK cell cytotoxic function. In this study, we tried to understand how NK cells distinguish normal host cells and tumor cells, how tumor cells escape immune surveilance function of NK cells, and how KIR functions in killer cells. The information obtained by this study could be used to understand the molecular mechanism of cancer development, and ultimately to regulate the NK cell function in a favorable way to the host.Methods : To effectively accomplish the research purposes, we operated two research teams in a systematic and complementary manner. In the first research team, cellular and molecular biological and immunological approaches were employed using purified NK cells and established tumor cells to reveal the molecular mechanism of NK cell~tumor cell interaction. In the second research teams, biochemical, molecular and cell biological approaches were employed by expressing the KIR in E. coli and Jurkat T cells to reveal the activating and inhibitory signal transduction pathways in killer cells.Results : NK cells were successfully purified from PBMC, and tumor cell lines were obtained from ATCC and routinely maintained in our lab. Using these cell lines, we performed NK cell Cytotoxicity assays. Leukemia cell lines (Molt4, K562, U937, Jurkat, and HL60), hepatoma cell lines (HepG2 and Hep3B), and thyroid cancer cell lines (NPA, ARO, FRO) appeared to be sensitive to NK cells, while Daudi and Raji cell lines appeared to be resistant to NK cells. We investigated in more detail what molecules make differences between NK sensitive and resistant cell lines focusing on the expression of KIRs on NK cells and MHC molecules on tumor cells. NK cells appeared to kill tumor cells by two different killing mechanisms: perforin/granzyme mediated cell death pathway and death receptor/ligand mediated apoptotic pathway. We also investigated the KIR mediated signal transduction pathways by charactering signaling molecules bound to KIR. By in vitro coprecipitation experiments and in vivo transfection analyses, we demonstrated that KIR constitutively associates with Shc in Jurkat T cells and 293T cells as well as in peripheral NK and T cells. In addition, expression of KIR significantly inhibited the AICD by inhibiting the FasL expression in Jurkat T cells upon activation. Interestingly, the KIR-mediated AICD inhibition appeared to be accomplished by recruitment of protein kinase C to the membrane-proximal region of KIR. These results suggest that the sequestration and/or inactivation of PKC by the membrane-proximal region of KIR renders the activated T cells less sensitive to AICDUsefulness : The information about the characteristics of the killing mechanism of human NK cells against carcinoma cell lines obtained from this study could be used to design a useful protocol for prevention or cure of cancers. In addition, the information about the activating and inhibitory signal transduction pathways could be used to design a specific immunomodulating strategy.
