세포치료제는 다양한 미생물성 감염원에 의한 오염이 가능하며, 개발의 모든 과정에서 미생물성 위해요인의 철저한 검증이 요구된다. 또한 세포기질 가이드에 세균, 곰팡이, 바이러스 및 마이코플라스마 등의 직접배양에 의한 무균시험이 안전성 평가대상으로 수재되어 있을지라도, 특이성, 감도, 정밀도를 지닌 진보된 기술이 미생물성 위해인자의 평가에 적용되는 것이 바람직하다. 그러나 최근의 분자생물학적 기술에 기반을 둔 NAT법 및 면역학적 방법들은 특정 미생물군을 대상으로 한정하기 때문에 미지의 세균에 대한 오염을 판정하는데 한계를 가질 수밖에
세포치료제는 다양한 미생물성 감염원에 의한 오염이 가능하며, 개발의 모든 과정에서 미생물성 위해요인의 철저한 검증이 요구된다. 또한 세포기질 가이드에 세균, 곰팡이, 바이러스 및 마이코플라스마 등의 직접배양에 의한 무균시험이 안전성 평가대상으로 수재되어 있을지라도, 특이성, 감도, 정밀도를 지닌 진보된 기술이 미생물성 위해인자의 평가에 적용되는 것이 바람직하다. 그러나 최근의 분자생물학적 기술에 기반을 둔 NAT법 및 면역학적 방법들은 특정 미생물군을 대상으로 한정하기 때문에 미지의 세균에 대한 오염을 판정하는데 한계를 가질 수밖에 없다. 따라서 세포치료제의 무균시험법의 표준균 주 7종 (Staphylococcus aureus, Pseudomonas aeruginosa, Clostridium sporogenes, Bacillus subtilis, Candida albicans, Aspergillus niger, Micrococcus luteus) 및 .P. acnes에 대한 자동화 배양검출법을 도입하여 무균시험의 신속성 및 정확성을 제고함으로 서 세포치료제의 미생물학적 안전성을 확보하고자 하였다. BacT/ALERT 3D 자동배양기 및 표준 호기성(SA)/혐기성(SN) 배지를 적용하여 시험한 결과 7종의 표준균주 중 5종의 세균 및 진핵 미생물인 C. albicans는 약 33시간 이내에, A. niger의 경우 70시간 이내에 약 1 CFU까지 검출이 가능하였다. 또한 이 때의 검출감도를 직접배양법과 비교한 결과, 본 시험방법이 검출감도 면에서 약 10-10만 배 더 sensitive하며 약 2-12시간 정도 검출 시간을 단축할 수 있다는 결과를 획득하였다 더욱이 최적배양온도에서는 물론 $37^{\circ}C$의 온도에서 배양이 모든 균종에 대하여 검출이 가능함을 확인하였다. 이 같은 사실은 배양 조건 중 산소의 존재 유무는 세균의 성장에 절대적으로 영향을 주나, 온도에 의한 영향은 그리 크지 않음을 시사한다. 또한 세포배양액에 spiking 하여 일정양의 미생물에 대한 검출을 시험하여 세포배양액이 미치는 영향을 검토한 결과, 고등동물 세포배양액에 항생물질이 포함되어 있지 않는 한 미생물 검출에 어떠한 영향도 미치지 않음이 확인되었다. 또한 공정서의 직접배양법과 비교분석한 결과 호기성균의 경우 거의 동일한 시험 결과를 보여 주었고, 혐기성균은 자동배양시스템에서 보다 우수하게 검출됨을 확인하였다. 즉, 세균류는 약 32시간 이내에 진균류는 약 70시간 이내에 검출 가능하므로 BacT/ALERT 3D 자동배양기와 표준호기성배지(SA bottle) 및 표준혐기성 배지(SN bottle)를 이용한 무균시험은 세포치료제의 대체무균시험으로 적합하다고 판단된다.

Human cells are important resources for medical treatments and can be used to treat many people after ex vivo propagation, expansion and selection. During the in vitro manipulation, living materials can be easy to contaminate with a variety of bacterial or fungal contaminants, so it should be tested for microbiological safety before administration. Even though various detection methods based on modern molecular technology have been developed, limitations such as narrow detection range and high cost remain to be solved. The use of automated microbial culture system based on culture method has been advocated as a means to detect unknown microorganisms from the biological specimen and avoid the limitations. The BacT/ALERT system (BioMeriux, Inc. France) is the fully automated, continuous-monitoring microbial culture system and has been widely used throughout the world. Here we provide evidence that the automated microbial culture system Is a good tool for testing the microbiological contamination in biologics. Direct culture of eight microorganisms has been accomplished to determine whether the BacT/ALERT standard aerobic (SA) and standard anaerobic (SN) media will support the growth of 6 bacteria (S. aureus, P. aeruginosa, C. sporogenes, P. acnes, M. luteus, B. subtilis) and 2 fungi (C. albicans, A. niger). Both five of aerobic microorganisms (P. aeruginosa, M. luteus, B. subtilis, C. albicans and A. niger) and one of facultative bacterium (S. aureus) were detected in this systems up to 1 CFU in 32 hours. In addition, anaerobic bacterium, C. sporogenes was detected up to 1 CFU in 16.56 hours and P. acnes was detected up to 10,000 CFU in 129.36 hours. In comparison with direct culture method defined in pharmacopoeia, this automated using BacT/ALERT 3D is more efficient about $10-10^5$ and save detection time up to 2-12 hours. Therefore, this automated microbial culture system enables to efficiently detect microorganisms contaminated in cell-based medicine. The Improved rapidness and sensitivity of the detection system enable to replace the classical culture method with the automated culture system for microbial safety test evaluating microbial contamination in cellular based products.
