세균의 화학적 언어 (acylhomoserine lactone)를 이용한 통신체계를 '정족수 인식(quorum sensing)' 이라 하며, 특히 병원성 세균의 경우 감염증 유발의 중요한 요인이다. 따라서 세균간의 통신체계를 인위적으로 교란 혹은 차단하는 연구를 통해서 전적으로 새로운 개념의 항 감염제를 개발할 수 있다. 본 연구에서는 병발에 관여하는 세균의 화학적 언어를 분해하여 무력화시키는 신호물질 분해미생물 및 분해효소의 개발과 이를 이용한 무름병 억제 연구를 수행하였다. 결과로서 병발 신호물질인 AHL을 분해하는 다양한 미생물 및 분해효소를 분리하여 특성화 하였다. pMal-His-Barallell벡터를 이용해 B. thurigiensis AHL 분해효소(AiiA,28kDa)를 고발현시켜, 순수분리 정제하였다. 정제한 AiiA 효소는 탄소수가 긴 decanoylhomoserine lactone에 대해 활성이 가장 높았으며, zinc에 의해 활성과 안정성이 증가하는 것을 확인하였다. 정제된 AiiA 효소를 이용하여 단백질 결정을 얻었으며, 또한 단백질의 3차 구조 분석을 위해 SeMet 으로 치환된 AiiA도 정제하여 결정화하였다. 6B beamline을 이용하여 이러한 두 종류 단백질 결정의 x-ray diffraction 실험을 수행하여 각각 $2.0{\AA}$, $2.4{\AA}$ resolution의 crystal diffraction data를 얻었다. HKL2000 pakage 프로그램을 이용하여 lactonase model을 분석한 결과 metalloenzyme group인 metallo-${\beta}$-lactamase의 folding 과 유사한 $\alpha\beta/\beta\alpha$ sandwich구조로 확인하였고, 3차 구조 분석 결과 7개의 아미노산잔기들이 $Zn^{2+}$ 결합에 관여하고 있음을 확인하였다. 이들이 $Zn^{2+}$ 결합부위임을 실험적으로 증명하고, 단백질에서 $Zn^{2+}$의 기능과 역할을 알아보기 위해 site directed mutation에 의해 7개의 아미노산을 각각 Alanine으로 전화시켰다. 이 결과 7개의 single mutant들은 AHL 분해 활성이 저해되는 것을 확인하였다. 효소 저해제인 homoserine lactone 과 단백질 complex의 3차 구조 분석을 통해 효소의 작용 메커니즘을 규명하였다. Bacillus 표면발현 motif를 이용하여, Bacillus thuringiensis 균주를 model로 한 신호물질 분해효소의 고발현 및 표면발현 system 구축 하였으며, 구축한 AHL 분해 미생물 및 분해 유전자를 이용한 무름병 억제 연구를 수행하였다.

Quorum sensing is a regulatory mechanism employed by bacteria to coordinate the behavior of their community in response to their population density. Bacteria recognize the changes in their population density by sensing the concentration of signal molecules which were accumulated as bacterial cell proliferates, In many Gram-negative bacteria including a number of pathogens such as Pseudomonas aeruginosa and Erwinia carotovora, virulence factor production and biofilm formation are linked to the quorum sensing (QS) systems which use diffusible N-acyl-L-homoserine lactones (AHLs) as intercellular messenger molecules. As AHL-mediated quorum sensing has been shown to be involved in pathogenesis and biofilm formation, the interference of quorum sensing, referred as quorum quenching, have recently received a great deal of attention. Among the quorum quenching strategies, the potent effects of enzyme-based AHL degradation have been identified in a wide diversity of bacteria. The AHL-degrading enzymes are fall into two groups according to the cleavage site of AHL. AHLases, identified in the Bacillus cereus group, degrade AHLs by hydrolyzing the lactone ring of AHLs,and produce the corresponding acylhomoserine molecules. The present study screened a more diverse range of AHL-utilizing bacteria on the basis of their ability to utilize non-substituted heaxnoyl homoserine lactone (HHL) as the sole carbon source. As a result, two strains, belonging to the genus Rhodococcur, were found to significantly differ in their AHL-utilization ability and AHL-degrading activities against various AHLs.
AHL-degrading enzymes have recently attracted intense interest for the development of antiinfection therapies for plants and animals. A number of organisms also contain genes coding for lactonases which hydrolyse AHLs into inactive products, thereby blocking the QS systems. Consequently, these enzymes attract intense interest for the development of novel anti-infection therapies. However, despite significant progress in the investigation of AHL-degrading enzymes, no structure is yet available, Accordingly, this study reports on the expression and purification of the AHL-lactonase from Bacillus thuringiensis subsp. kurstgki HD263 (hereafter referred to as BTK-AiiA), as well as the successful crystallization of the enzyme. We here report a $2.0{\AA}$ resolution structure of the AHL-lactonase from -Bacillus thuringiensis. In spite of limited sequence similarity, the enzyme shows remarkable structural similarities to glyoxalase II and RNase Z proteins, members of the $metallo-\beta-lactamase$ superfamily. We present experimental evidence that the AHL-lactonase is a metalloenzyme containing two zinc ions involved in catalysis, and we propose a catalytic mechanism for bacterial metallo-AHL-lactonases. The enzyme is a 250-residue-long protein with a conserved sequence motif $^{104}HXHXDH^{109}\;H^{169}$ similar to the $Zn^{2+}$ binding motif of several metallohydrolases, including the glyoxalase II and arylsufatase enzyme families, -lactamases, and RNase Z. However, there is controversy as to whether or not this motif is involved in metal binding in AHL-lactonases. To gain insight into the structure-function relationships in this novel class of enzymes, we investigated the AHL-lactonase from Bacillus thuringiensis subsp. kurstaki HD263. We solved the crystal structures of this protein with a bound competitive inhibitor L-HSL at $1.7{\AA}$ resolution, respectively. The protein shows remarkable structural similarities to $metallo-\beta-lactamase$, and an even closer relationship to the human glyoxalase II, and to the bacterial phosphodiesterase (RNase Z). The electron density map of the active site of the enzyme reveals a canonical dinuclear $Zn^{2+}$ center, and the presence of the metal ions was independently verified by atomic emission spectrometry. On the basis of the crystal structures we propose a catalytic mechanism for BTK-AiiA along with supporting mutagenesis and biochemical data.
