RLK와 CBNAC 단백질이 KO된 식물체에서 SA를 처리 하였을때 PR-1 gene의 발현이 WT과 비교하여 증가하였다. 애기장대에서 TIR domain을 가지는 CaM 결합 protein(AtCBP54)의 분리하고 형질전환체의 구축을 통해서 병저항성을 부여함을 밝혔다. 병저항성 기작 연구를 위해서 AtCBP54 과발현 식물체를 구축하여 분석한 결과 애기장대의 주요한 phytoalexin인 camalexin을 대량으로 축적하는 것으로 확인하였다. CaM 결합 단백질 중의 하나인 DsPTP1를 분리하여 생체내의 기능을 확인하고자, 이 단백질과 결합하는 또 다른 단백질을 연구하여 기존에 스트레스 신호전달에 관여하는 MPK3, MPK4, MPK6를 분리하였다. In vitro상에서 DsPTP1은 결합 단백질인 MPK3, MPK4, MPK6를 항인산화 작용을 통해 비활성 시키는 것을 확인 할 수 있었으며 CaM은 이러한 DsPTP1의 효소 활성을 저해 하였다. 벼에서 CG-1 homology를 갖는 CaM 결합 전서조절 인자(OsCBP)를 분리하여 결합 DNA sequence를 포함하여 생화학적인 특성들을 조사하였으며, GFP를 통해서 localization을 본 결과 핵으로 위치하지만 CaM에 의해서 OsCBP의 핵으로의 이동이 저해되어 OsCBP에 의한 transcription activity가 저해됨을 확인하였다. 이들 유전자들의 in vitro 및 in vivo function 규명 등을 통하여 국제학술지 6편 논문을 발표하였다.

The dynamics of a Ca2+ signal are largely controlled by influx through channels and efflux through ATPases and antiporters. The Arabidopsis genome encodes 14 Ca2+ ATPases, 10 of which belong to a family of autoinhibited Ca2+ ATPases (ACA) that are predicted to be activated by Ca2+/calmodulin, and 4 of which belong to ER-type Ca2+-ATPases (ECA). The different stresses trigger distinct patterns of cytosolic $Ca^{2+}$ signals, which are transduced by $Ca^{2+}$-binding proteins such as calmodulin (CaM). The transient Ca2'signals trigger changes in enzyme activity, transport across membranes, cell structures and gene expression that allow plants to cope with stressful environments. Cellular $Ca^{2+}$ signals are transduced to the transcription machinery by different mechanisms including phosphorylation or dephosphorylation of TFs, which may affect entry of the TFs to the nucleus or their DNA binding properties. Alternatively, $Ca^{2+}$ may bind to sensor proteins that consequently bind to TFs and modulate their activities. In this study to know the importace of Ca2+ influx and efflux in plant growth and development we isolated 10 different Ca2+-ATPase mutant from Arabidopsis and screened their mutant phenotypes extensively. Vacuole Ca2+ATPase (aca11) showed ABA insensitive and Ca2+ insensitive phenotypes on stomata opening and closing. The vacuole Ca2+ATPase double mutant (aca4/aca11) showed the HR-like cell death phenotype. Interestingly the cell death of the mutant was come from the disruption of vacuole membrane in aca4/aca11 plant vacuole. We figured out that there is close relation between Ca2+ homeostasis and the disruption of vacuole membrane. Interestingly the gene encoding vacuole processing enzyme(VPE) was highly expressed in the knock out mutant. To investigate CaM mediated Ca2+ signaling pathway we isolated lots of CaM binding protein from Arabidopsis and rice expression libraries such as protein phosphatase(DsPTP1), TIR-like protein, tanscription factors. We isolated cDNA encoding a dual specificity protein phosphatase 1, which is capable of hydrolyzing both phosphoserine/threonine and phosphotyrosine residues of the substrates. Using a gel overlay assay, we identified two Ca2+-dependent CaM binding domains (CaMBDI in the N terminus and CaMBDII in the C terminus). Specific binding of CaM to two CaMBD was confirmed by site-directed mutagenesis, a gel mobility shift assay, and a competition assay using a Ca2+/CaM-dependent enzyme. At increasing concentrations of CaM, the biochemical activity of dual specificity protein phosphatase 1 on the p-nitrophenyl phosphate (pNPP) substrate was increased, whereas activity on the phosphotyrosine of myelin basic protein (MBP) was inhibited. Our results collectively indicate that calmodulin differentially regulates the activity of protein phosphatase, dependent on the substrate.
