Ⅱ. 연구개발의 목적 및 필요성 폐수 중의 오염 물질을 연료로 이용할 수 있는 미생물연료전지를 개발하기 위해 1960년대 이래 많은 연구가 진행되었다. 전형적인 미생물연료전지는 음극부에서 미생물의 작용에 의하여 기질이 산화되면서 발생하는 전자에 의하여 산화환원전위가 낮아질 수 있으나 일반미생물의 세포표면이 전기적으로 절연이기 때문에 전자전달이 세포 안에서 이루어지는 일반미생물은 전자를 전극으로 전달할 수 없다. 그러므로 전자전달 매개체가 일반미생물을 이용하는 미생물연료전지에서는 전자를 전극으로 전달하기 위하여 반드시 필요하다. 이러한 매개체들은 독성을 띠고 있고 매우 비싸다. 그러므로 미생물연료전지가 바이오센서나 전기생산 등의 다양한 목적으로 응용될 수 있지만 장시간 운전에 적합하지 않았기 때문에 산업화된 예는 없었다. 본 연구실에서 처음 그 가능성이 입증된 무매개체 미생물연료전지는 이런 단점을 극복한 것으로 현재 폐수처리용 장치로 사용하기 위하여 본 연구를 진행하고 있다. 이러한 미생물연료전지를 폐수 처리 공정으로 개발할 경우 1) 기존의 폐수 처리 공정에서 필요한 동력을 줄일 수 있을 뿐 아니라 2) 생산하는 전기를 공정에 필요한 단계에서 사용할 수 있으며, 3) 오염 물질이 갖는 에너지의 많은 부분을 전기로 전환시켜 폐수 처리에서 발생하는 오니의 양을 1/3 이하로 줄일 수 있다.
Ⅲ. 연구개발의 내용 및 범위 본 연구에서는 미생물연료전지 형태의 폐수처리 장치를 만들고자 한다. 이를 위해서는 먼저 미생물연료전지의 performance를 최적화하기 위하여 limiting step을 확인하였다. 양이온교환막의 가격이 매우 비싸고 운전 도중에 proton permeability가 제한 될 수 있기 때문에 미생물연료전지의 경제성을 가늠할 수 있는 척도로써 중요한 인자이다. 새로운 형태의 미생물연료전지는 이 양이온교환막을 제외하고 구성하였다. 양극부 반응에서 일어날 수 있는 제한 현상을 제거하기 위해서 양극부는 산소에 대한 친화도가 높은 전극을 사용하였으며 양이온 교환막을 통하여 투과되는 산소에 대한 영향도 관찰하였다. 한편 미생물연료전지에서 일어나는 전기화학적인 특성을 파악하기 위하여 미생물연료전지에 분포하고 있는 미생물들을 배양을 하였으며 한편으로 배양과는 무관하게 분자생물학적인 방법을 이용하여 미생물의 분포를 파악하였다.
Ⅳ. 연구개발결과
(1) 미생물연료전지의 대사속도 측정과 각 구성단계 파악 - 미생물연료전지를 폐수처리 장치로의 가능성을 탐색하기 위하여 성능을 파악하였다. NCBE-type cell의 미생물연료전지는 전분가공공정에서 발생하는 폐수를 연료로 하여 약 68 ppm/h의 처리속도와 17% 수준의 coulomb yield를 보였다. 미생물연료전지의 효율을 결정하는 인자를 다음과 같이 확인하였다. 1) 음극부에서 미생물에 의한 기질의 분해속도, 2) 음극부에서 전자의 전극으로 전달 속도, 3) 음극부와 양극부의 외부 저항, 4) 양이온 교환막을 통한 수소이온의 전달 속도, 5) 양극부에서 산소에 의한 전자와 수소이온의 소비 속도, 6) 양극부로부터 전달되는 산소에 의한 유기물 산화 그리고 7) 음극부에 공급되는 연료의 channeling 현상 등이다.
(2) 양극부 전극의 임계산소농도 측정과 개선 - 양극부에 사용하는 전극 (graphite felt)의 임계산소농도는 약 6.6 ppm이었고 graphite felt에 백금을 처리한 경우 2 ppm의 임계산소농도를 나타내었다. - 양극부 전극에 mediator로 conducting polymer, 산화환원효소 또는 ferricyanide를 처리하는 것은 일시적으로 효과를 보였지만 장시간 운전하기에는 적합하지 않았다. 백금처리는 효과적이었으나 경제성을 고려할 필요가 있다.
(3) 연속식 미생물 연료전지를 이용한 전력 생산 특성 - 연속적으로 폐수처리와 동시에 전력 생부피당 전력밀도 51 mW/L와 volumetric COD removal rate 2.5-8.9 kg/m3 d를 얻을 수 있었다.
(4) 공기호흡형 미생물연료전지 개발 - Catholyte로 공기포화된 물을 사용하지 않음으로써 동력비를 절감할 수 있는 공기호흡형 미생물연료전지를 개발하였다. 단위부피당 최대 전력밀도는 90 mW/L이었다. 이것은 공기포화된 물을 사용하는 미생물연료전지의 약 1.8배 되는 값이다. Volumetric COD removal rate는 최대 21.6 kg/m3 d를 얻을 수 있었다.
(5) 폐수처리용 미생물연료전지의 구성 및 운전 - 양이온 교환막을 제거한 미생물연료전지를 폐수처리용 미생물연료전지 (ML-MFC-1)로 구성하고 이를 운전하였다. - 음극부의 channeling현상을 최소화하고 내부 저항을 줄이면서 새로운 폐수처리용 미생물연료전지 (ML-MFC-2, MLMFC-3)를 설계하였다. - Separator를 사용하여 음극부와 양극부를 밀착시킨 결과 전류 발생량을 약 1.4배 향상시킬 수 있었고 양극부에서 백금 촉매량을 증가시켜 쿨롬수율 99% 이상을 얻을 수 있었다. - 막없는 미생물연료전지의 설계 및 운전 조건 최적화를 통해 폐수를 최대 0.86 kg/m3 d의 속도로 처리 할 수 있었고 최대 volumetric power density는 7.6 mW/L를 얻을 수 있었다. - 폐수처리용 미생물연료전지에서 발생하는 슬러지의 발생량이 활성슬러지법 대비 18% 수준으로 발생하였다.
(6) 미생물연료전지의 microbial diversity와 전기화학활성 미생물 분리 - 전분가공폐수, 아세트산과 인공폐수 등을 사용한 미생물연료전지의 microbial diversity를 분석하기 위한 16S rDNA clone library를 구축하였다. - 전기화학활성 미생물 분리용 plate를 제작하고 38종의 미생물을 분리하였으며 분리균 중에서 5종이 전기화학활성을 나타내었다. 분리균 중 2균주는 신규성을 확인하였다.
(7) 난분해성 폐수의 처리 - 인공폐수에 phenol을 첨가하고 미생물연료전지로 운전한 결과 phenol 분해에 의한 전류발생을 확인할 수 있었다. - 인공폐수에 nitrilotriacetiate (NTA)를 첨가하여 운전한 결과 oligotrophic-type 미생물연료전지에서 NTA가 분해됨을 확인하였다. - 석유화학공업에서 발생하는 폐수를 미생물연료전지로 처리할 수 있는 가능성을 확인하였다.
Ⅴ. 연구개발결과의 활용계획 연구 결과는 연구의 최종목표를 달성하기 위하여 제 2단계 연구의 중요한 참고자료로 활용될 계획이다.

Ⅱ. The aim of this study Since ‘60s, microbial fuel cells have been studied to use organic materials for electricity generation. In a typical microbial fuel cell, an anodic electrode potential is developed when the electrons from the oxidation of fuel by microorganisms are available to electrode. However, electrons can not be transferred from normal microbial electron transport systems to the electrode due to the non-conductive nature of the cell surface structures. Electrochemical mediators were employed to render electron transfer from the microbial cells to the electrode. These mediators are usually toxic and expensive. Therefore, the long-term operation of mediated MFCs cannot be achieved with limited commercial applications although MFCs can be used for various purposes including biosensors and electricity generation system. In this laboratory, we showed that electrochemically active bacterium could be used as the biocatalyst in the mediator-less microbial fuel cell. Studies have been made to develop this mediator-less microbial fuel cell as a wastewater treatment process throughout the study. After construction of microbial fuel cell as wastewater treatment process, 1) energy can be saved in the wastewater treatment process, 2) the system can be operated using electricity from microbial fuel cell, 3) reduced sludge generation by70% is expected because energy contained in the organic contaminants is converted to electricity.
Ⅲ. Research objectives Studies have been made to develop a microbial fuel cell as a wastewater treatment process. The limiting steps have been identified to optimize the performance of the microbial fuel cell. The use of cation exchange membrane is one of the important factor determining the economic feasibility of the microbial fuel cell because it is expensive and limits proton permeability. A novel microbial fuel cell was constructed without using membrane. In order to eliminate limitations exerted by poor cathode reaction, cathode was modified to increase the affinity for oxygen and the oxygen permeability through the cation specific membrane was determined. In addition to the electrochemical aspects of the microbial fuel cell, studies were made to identify the microbial population responsible cultivation and cultivation-independent methods.
Ⅳ. Results
(1) Determination of microbial activity and rate-limiting steps in an MFC -To test microbial fuel cell as a wastewater treatment process, NCBE type cells were operated using wastewater from starch processing factory. These microbial fuel cells showed about 68 mg/l/h COD consumption rate, and 17% coulomb yield. The rate-limiting steps in an MFC were identified as; 1) fuel consumption rate in the anode, 2) electron transfer rate from the microbial cells to the anode, 3) external load (resistance), 4) proton transfer rate through the membrane, 5) oxygen consumption rate in cathode, 6) oxygen diffusion to the anode compartment and 7) channeling phenomenon in the anode compartment.
(2) Determination of critical oxygen concentration and enhancement of cathodic reaction - The critical oxygen concentration ([O2]crit) of graphite felt and platinum-coatted graphite felt were 6.6 and 2 ppm, respectively. - The mediators such as conducting polymer, redox active proteins and ferricyanide were effective for the cathodic reaction enhancement, however, it was not suitable for long term operation. Although platinum treatment to graphite was most effective one, it is expensive material for wastewater treatment process.
(3) Continuous electricity generation using microbial fuel cell - Continuous microbial fuel cell was optimized with respect to electricity generation. The maximum volumetric power density of 51 mW/L and the volumetric COD removal rate of 2.5-8.9 kg/m3 d were obtained using air-saturated water as cathode system.
(4) Development of air-breathing microbial fuel - An air-breathing microbial fuel cell was developted to reduce the operation cost by eliminating the aeration to the cathode. The maximum volumetric power density was 90 mW/L, which was 1.8 times higher than that of microbial fuel cell using air-saturated water. The maximum volumetric COD removal rate of 21.6 kg/m3 d was obtained also.
(5) Construction and operation of wastewater treatment microbial fuel cell - Membrane-less microbial fuel cell (ML-MFC-1) was constructed as a prototype of wastewater treatment process. - Second prototype ML-MFCs (ML-MFC-2, MLMFC-3) were developed to minimize channeling phenomenon and internal resistance in the anode of ML-MFC-1. - An ML-MFC configuration that the cathode compartment is directly connected to anode compartment using a separator could improve the current generation about 1.4 times higher than that with previous one. The coulomb yield could be increased to be over 99% by increasing platinum loading on cathode. - Sludge generation from MFCs was determined to be around 18% relative to the activated sludge process.
(6) Microbial diversity in microbial fuel cells and isolation of electrochemically active microorganism - Chromosomal DNA was extracted from anodes of MFCs operated using starch processing wastewater, acetate, and artificial wastewater to construct 16S rDNA libraries for the analyses of the microbial diversities. - Various combinations of electron-donors and electron acceptors were tried to isolate microrganisms from an acetate enriched MFC. The culturable clones were less than 0.01% of the expected bacterial cell present in the MFC. Thirty eight strains were isolated and five of them showed electrochemical activity. Two of electrochemically active isolates were identified as novel strains.
(7) Degradation of xenobiotic under the MFC conditions - MFCs generated current with the concomitant removal of phenol or nitrilotriacetate added to the artificial wastewater. - The wastewater from petrochemical industry could be treated in an MFC.
Ⅴ. Applications The results of this project provide a guideline for MFC operation to treat wastewater, that is the final goal of this project in the second phase research.
