Pretenses play essential roles in numerous biological processes and pathology of many diseases. for example, human immunodeficiency virus (HIV) and hepatitis C virus, encode own protease which is critical fro proper viron assembly and maturation. Therefore, these viral protease has been considered as a promising target for the rational drug development. Tn this study, we have developed a genetic assay for site-specific proteolysis (GASP). We proposed that GASP may be useful to detect and characterize proteolysis in yeast. The resulting information should be beneficial for the understanding the molecular mechanism of protease activity and further development of protease inhibitors. We showed that GASP worked nicely as we proposed and the substrate specificity of viral proteases such as HGV NS3 protease, TuMV na protease, Coxsackievirus 3C protease, and Mengovirus 3C pretense has been investigated using GASP. For degradomics approach, we examined the possibility of employing GASP for the screening of relevant substrates. The C.elegans cell death pretense, CED-3 was used as model system. Using C.elegans genomic library, we were able to obtain eight candidates from half million transformants. Among candidates, three clones were confirmed by in-vitro cleavage assay. Another library which is cDNA library, was used for degradomics approach. One candidate could Ire isolated from million transformants, this clone showed having a correlation with cell death signal. Another approach is protease engineering for removing disease-related proteins such as poly-glutamine protein. For designing poly-Q pretense (refer PQP), we used the rational randomization. Through screening using GASP, modified protease which can cleave poly-glutamine stretch, could be obtained. Beside GASP, we have performed inhibitor development for disease related pretenses which were BACE and ADAM12. For this approach, GASP has been applied to a S2 cell. The S2 cell-based assay was utilized and candidates for BACE and ADAM12 were discovered. We further developed another genetic method referred to as sGASP (secretory GASP). The purpose of sGASP is to detect and study proteolysis conferred by membrane anchored or secreted protease such as type II membrane serine pretense and beta site APP cleaving enzyme (BACE). Using sGASP, we were able to investigate pretense activity of TMPRSSs (TransMembrane PRoteaSe, Serine) which are type II membrane serine pretenses. Another approach is generation of nobel protease. Using sGASP, we successfully developed novel protease derived KEX-2 using rational randomization and its substrate specificity was altered. In addition to GASP and sGASP, we used a split-ubiquitin technique which is capable of detecting interaction between membrane protein. Using this method, we characterized the interaction between C99, presenilin, and nicastrin.
