Fusarium graminearum (teleomorph: Gibberella zeae) is a filamentous homothallic ascomycete that causes diseases such as scab, head blight, and stalk or ear rot in corn, barley, wheat, and rice. Severe epidemics of these diseases caused serious economic losses in the 1990s in several countries including Korea, Japan, and the United States. In addition, this fungus is a potential threat to humans and animals as it produces mycotoxins on diseased crops. In spite of the economic importance of F. graminearum, the molecular mechanisms of disease development caused by this fungus are poorly understood. Recently, a functional genomics approach has been launched to explore the pathogenic biology of G. zeae at the molecular level. As a first effort, the entire genome of G. zeae has been sequenced and has been publicly available since 2003. Using the genome database, functional analyses of the entire gene set involved during the pathogenesis of F. graminearum are being conducted. To accelerate this process, we have generated a collection of insertional mutants from a representative strain of F . graminearum using the restriction enzyme-mediated integration (REMI) procedure during the last three years.
The objectives of this project are to massively isolate pathogenesis-related genes from the genome of F . graminearum, the causal agent for Fusarium head blight of rice, using functional genomics and to select genes from the gene collection, which would be useful for construction of transgenic rice plants resistant to Fusarium head blight. To pursue the goals, we propose the following approaches. First, an additional collection of 20,000 REMI transformants will be constructed using a new method. After screening the REMI transformants for several traits involved in disease development such as pathogenicity, mycotoxin production, and sporulation, each mutant will be outcrossed with a M AT gene-deleted strain for genetic analysis. A mutant carrying a tagged mutation will be further analyzed for identification of a genomic DNA region flanking the vector insertional site at the genome. Subsequently, the entire region of a mutated gene will be cloned, sequenced and characterized. In addition to the REMI mutagenesis, we will employ a transformation-mediated targeted gene deletion strategy for functional analysis of possible pathogenesis-related genes. To do this, we will use both inverse PCR and double-joint PCR for construction of a transforming vector. With these strategies, we hope to assign functional roles of more than 200 genes during the disease development by F . graminearum and to select at least 5 genes derived from the gene collection for generation of transgenic crops resistant to the diseases caused by F. graminearum.
