이 연구는 화학자가영양세균의 한 그룹으로 일산화탄소(CO)를 유일한 에너지 및 탄소 원으로 이용하여 성장하는 호기성 CO 산화세균(carboxydobacteria)인 Mycobacterium sp. strain JC1을 대상으로, 이 세균의 CO 대사과정(CO 산화 및 CO2 동화) 중 CO 산화와 관련된 일련의 유전자들을 찾아 특성과 기능을 밝히는 한편, 동 유전자들의 발현 및 조절 양상을 규명하기 위한 것이다.

Carboxydobacteria는 그들이 소유하고 있는 CO dehydrogenase (CO-DH)를 이용하여 CO를 물과 반응시켜 산화한 다음(CO + $H_2O$ → $CO_2$ + $2H^+$ + $2e^-$), 반응산 물 중 이산화탄소(CO2)는 Calvin cycle을 이용하여 세포구성물로 전환하고, 전자는 산화적 인산화에 의한 에너지 생산을 위하여 CO-insensitive 전자전달계로 보낸다.
Carboxydobacteria에 대한 현재까지의 연구는 이들 세균의 생물학적 특징과 함께 CO 산화 기작, CO 산화를 위한 전자전달계 및 CO 산화과정의 주효소인 CO-DH의 물리·화학적 특성 및 $CO_2$ 동화과정의 주효소인 ribulose bisphosphate carboxylase/oxygenase (RubisCO)의 물리·화학적 특성 등 CO의 산화와 $CO_2$의 동화와 관련된 생리·생화학적 연구는 활발히 진행되었으나, $CO의 산화 및 동화와 관련된 분자유전학적 연구는 일부 세균에서 수행된 CO-DH 유전자의 특성과 한 세균에서 수행된 RubisCO 유전자의 특성에 대한 연구만 있을 뿐, 현재까지 CO 대사 (CO metabolism)와 관련된 유전자 전반의 특성과 기능 및 그들의 발현 체계에 대한 연구는 물론이고 CO에 의한 동 유전자들의 발현 조절에 대한 연구도 전무하였다.
따라서 carboxydobacteria의 CO 대사 전반에 대한 근본적인 이해의 바탕을 마련하기 위한 노력의 일환으로, 먼저 carboxydobacteria에서만 볼 수 있는 특이한 능력인 CO 산화과정에 대한 분자유전학적인 연구를 수행하기로 하고 연구책임자의 연구실에서 분리하여 지난 20년 동안 연구를 계속하고 있는 carboxydobacteria인 Mycobacterium sp. strain JC1을 대상으로 이 세균의 CO 산화관련 유전자들의 특성과 기능을 밝히고, 동 유전자들의 발현 및 조절 양상을 조사하기 위한 이 연구를 수행하였다.
이와 같은 연구 목표를 효율적으로 달성하기 위하여, Mycobacterium sp. strain JC1의 CO 산화와 관련된 유전자 중 현재 확보하고 있는 CO-DH 유전자와 가상적인 전사조절 유전자 이외의 유전자를 분리하여 그들의 특성과 기능을 조사하는 한편, 동 유전자들의 전사단위(operon)와 발현 및 조절 양상에 대한 연구도 수행하였다.
이 연구는 CO를 에너지 및 탄소 원으로 이용하여 성장하는 특이한 화학자가영양 세균인 carboxydobacteria의 CO 산화와 관련된 유전자 전반의 기능과 동 유전자들의 발현 및 조절에 대한 최초의 연구이다. CO-DH의 구조 유전자인 cutA, cutB, cutC와 CO-DH의 전사 조절자로 추정되는 cutR 이외에 CO-DH 유전자의 upstream과 downstream에서 27개의 ORF를 확인하였다. 확인된 27개의 ORF 중 16개의 ORF가 CO 산화에 관련된 유전자로 확인되었다. 돌연변이체를 통한 연구, 아미노산 서열 분석을 통한 연구, gel mobility shift assay를 통해 CO 산화에 관련된 16개의 유전자 중 10개의 유전자의 기능을 확인하였다.
CO-DH 유전자의 전사 조절에 대해 연구한 결과, CO에 의한 CO-DH 발현의 증가는 CutR, stationary phase에서 CO-DH 발현의 증가는 Rv3676-homolog 단백질에 의해 조절되는 것으로 확인되었다. 또한 Orf6는 CutR, Rv3676-homolog 단백질 같은 전사조절자보다 더 상위 단계에서 전사 조절에 관여한다고 확인하였다.

This study is intended to isolate and characterize genes involved in the oxidation of carbon monoxide (CO) and investigate mechanism of expression and regulation of those genes in Mycobacterium sp. strain JC1 to elucidate molecular genetic background of CO oxidation in carboxydobacteria capable to grow on CO as a sole source of carbon and energy.
CO is oxidized to CO2 by reacting with H2O in the presence of CO dehydrogenase (CO-DH) in carboxydobacteria (CO + $H_2O$ → $CO_2 + $2H^+$ +$2e^-$). Among the reaction products, CO2 is converted to cellular carbon through the Calvin cycle and electrons are transported to the electron transport system, resulting in the production of energy.
There have been extensive studies on the biological, physiological, and biochemical characteristics of CO metabolism in carboxydobacteria such as biological properties of these bacteria, mechanism of and electron transport system for CO oxidation in the bacteria, and physical and chemical properties of CO-DH and ribulose bisphosphate carboxylase/oxygenase, the key enzymes for CO oxidation and CO2 assimilation, respectively. However, little is known on the molecular mechanism of CO metabolism. There have been no studies on the molecular genetic background of CO oxidation and CO2 assimilation such as characteristics and functions of whole genes involved in the overall CO metabolism and mechanism of regulation and expression of those genes including molecules involved in the CO sensing and signal transduction, except several studies on the properties of genes for CO-DH and ribulose bisphosphate carboxylase/oxygenase.
Therefore, first of all we decided to carry out this 3-year study to investigate molecular genetic background of CO oxidation in an effort to establish fundamental understanding of CO metabolism in carboxydobacteria. To reach the goal efficiently, we isolated and characterized genes involved in the oxidation of CO in Mycobacterium sp. strain JC1, except those for CO-DH and a putative regulator we currently possess, identify operon units, investigate expression pattern and mechanism of regulation of those genes.
This is the first study on the functional analysis, expression pattern, and regulation of genes involved in the oxidation of CO. We newly identified the 27 ORFs, except the structural genes for CO-DH and a putative regulator that we had already identified in the upstream and downstream regions of CO-DH genes.
Among the 27 ORFs, 16 ORFs involved in the oxidation of CO in Mycobacterium sp. strain JC1. The function of 10 genes of 16 gene related to the CO oxidation were identified by mutagenesis, comparative analyses of their primary structures, or gel mobility shift assay.
In the study of transcription of CO-DH genes, we revealed that CutR regulates the transcription of CO-DH genes under CO-inducible conditions and that Rv3676-homolog protein regulates the transcription of CO-DH genes in stationary phase. Furthermore, we provided evidence that Orf6 is related to the regulation of CO-DH genes at the level upstream of the transcription factors such as CutR and Rv3676-homolog proteins.
