본 연구과제의 주요목표는 신경계의 synuclein 단백질과 여러 가지 비정형구조 단백질들을 모
델로 사용하여 비정형구조 단백질의 구조적 특성을 비롯한 생물리화학적 성질을 규명하여 이
들 단백질을 간단히 확인할 수 있는 assay방법을 개발하고 이들 단백질이 수행하고 있는 생물
학적 기능과 구조와의 관계를 규명하는 일이었다. 또한 비정형구조 단백질의 경우 흔히 관찰되
는 단백질 응집 및 침착으로 발전하는 선택적 단백질 상호작용의 기본 작동기작을 분자수준에
서 철저히 이해하고 규명하며, 이와 같은 비정상적

서 철저히 이해하고 규명하며, 이와 같은 비정상적 상호작용을 촉진 또는 억제 시켜줄 수 있는
물질 및 방법을 동정하고 개발해 왔다. 아울러 중추신경계에서 유래된 신경 세포를 모델로 신
경계에서 기능하는 대표적인 비정형구조 단백질인 synuclein과 그 중합체에 의한 신경세포의
분화, 성장 및 세포사멸에 미치는 효과와 그 신호전달기전을 연구함으로써 비정형구조 단백질
의 생리학적 기능 및 병리학적 역할에 대한 하나의 모델을 제시해 보고자 하였다.
본 연구계획을 충실히 수행하기 위하여 우리는 비정형구조 단백질의 단백질화학팀, 병리연구팀
과 기능 연구팀을 구성하여 각 연구팀간에 상호보완 및 유기적인 관계에서 비정형구조 단백질
에 대한 총체적인 연구를 수행하였다. 단백질화학팀에서는 비정형구조 단백질들을 대량발현시
켜 순수정제 해낸 후 이들 단백질의 구조적 특성과 유체역학적 성질 등의 생물리화학적 성질
을 규명하고 이를 이용하여 이들 비정형구조 단백질을 간단히 확인할 수 있는 assay 방법을
개발하였으며 이들 비정형구조 단백질의 단백질 상호작용 및 이때 수반되는 구조적 변화도 연
구함으로써 이들 단백질의 생물학적 기능과 구조와의 관계를 규명하였다. 또한 synuclein 단백
질이 chaperone 활성을 보이는 기작을 중점적으로 연구하였으며 synuclein 단백질의 구조변화
와 chaperone 기능과의 관계를 연구하였다. 병리 연구팀에서는 비정형구조 단백질의 경우 흔히
관찰되는 단백질 응집 및 침착으로 발전하는 선택적 단백질 상호작용의 기본 작동기작을 분자
수준에서 철저히 이해하고 규명하며, 이와 같은 비정상적 상호작용을 촉진 또는 억제 시켜줄
수 있는 물질 및 방법을 동정하고 개발해 왔다. 즉, 변형된 단백질의 열역학적 안정성을 유도
하여 자가상호작용을 최소화시키는 방법을 연구하고, 단백질 응집의 중심핵에 대한 존재여부를
확인하며 그 형성을 억제 또는 촉진시킬 수 있는 물질을 개발하고, 응집과정의 중간산물의 양
을 감소시키는 방법등을 강구하였다. 또한 병리 및 생리적 의의를 갖는 α-synuclein 상호작용
성 리간드를 동정하고 선정하여, 이들이 단백질 응집에 미치는 효과와 특성을 분석함으로써 단
백질응집물의 형성기작을 규명하고 그 응용 가능성을 타진해 나가고자 하였다. 비정형구조 단
백질의 기능 연구팀에서는 중추신경계에서 유래된 신경 세포를 모델로 신경계에서 기능하는
비정형구조 단백질에 의한 신경세포의 분화, 성장 및 세포사멸에 미치는 효과와 그 신호전달기
전을 연구함으로써 비정형구조 단백질의 생리학적 기능 및 병리학적 역할을 규명하였다. 또한
지금까지 밝혀낸 synuclein과 선택적으로 결합하는 유전자들과 synuclein 사이의 관계를 구체
적으로 규명하고 신경세포 발달에 미치는 분석모델 시스템을 개발하였다.
그동안 우리는 비정형구조 단백질의 생물리화학적 성질 연구, 단백질 자가상호작용 및 응집현
상의 기전연구, synuclein에 의한 신경세포의 성장, 분화 및 세포사멸의 효과규명과 신호전달
연구 분야에 계획했던 연구를 충실히 완수하였다. 본 연구를 통하여 많은 흥미로운 결과를 얻
었고, 그 결과들을 SCI 국제저널에 11편의 논문을 발표하였고 3편을 투고하였으며, 국내저널에
1편의 논문을 발표하였다. 국제학술대회에 12차례 포스터형태로 발표하였고, 국내학술대회에 20
차례 포스터를 발표하였다. 그리고 10여명의 석박사 연구인력을 배출하였고, 국내외 연구진과
학문적 네트워크를 형성하고 공동연구를 수행하였다. 그동안의 연구성과를 발전시킬수 있는 후
속 연구도 과학재단의 지원으로 계속 수행하게 되었다. 본 연구를 통해 얻은 결과들은 앞으로
비정형구조 단백질의 구조 기능 및 질병과의 관계 분야 연구에 유용하게 사용되어질 것이다

The main purpose of this study was to reveal the structural and functional properties of

The main purpose of this study was to reveal the structural and functional properties of
unstructured proteins systemically and in more detail. We aimed to investigate the
structural, hydrodynamical, and biochemical properties of unstructured proteins, and
eventually to set up a simple assay method that can be used to determine whether a
protein is unstructured or structured. We also aimed to investigate the mechanism of self
assembly and aggregation process of unstructured proteins, and planed to screen molecules
and conditions which promote or inhibit the amyloid formation. We were also interested in
the biological function of unstructured protein in relation to its unusual structure. Using α
-synuclein as a representative of unstructured proteins functioning in neuronal cells, we
aimed to investigate the effect of this unstructured protein on the development,
differentiation and cell death of neuronal cell lines. This functional study was expected to
make it possible to present an example of how the unstructured proteins function in health
and disease.
To accomplish our research purposes, we organized three research teams, protein chemistry
team, pathological study team, and functional study team of unstructured proteins. Our
teams have performed a systematic studies on unstructured proteins in a complementary
and organic manner. In the protein chemistry team, biochemical and biophysical properties
of unstructured proteins were investigated to set up a detection method for the
characterization of unstructured proteins. In addition, protein-protein interactions and
structural changes of unstructured proteins were also investigated to reveal the
structure-function relationship. Chaperone function of synuclein and the relation between
the chaperone activity and structural changes were also investigated. In the pathological
study team, molecular mechanisms of protein self assembly and protein aggregation were
investigated in detail. Furthermore, molecules and methods that can accelerate or inhibit the
protein self assembly and aggregation processes were searched throughly to develop a
potential therapeutic method for neurodegenerative diseases caused by protein aggregation.
In the functional study team, effects of a-synuclein on the neuronal cell growth,
differentiation and apoptosis were investigated using a hipocampal neuron cell line. Signal
transduction pathways of the above processes were also investigated to reveal the
physiological and pathological role of a-synuclein. In addition, proteins that can bind and
interact with a-synuclein were searched by yeast two hybridization technique, and
characterized by cellular and molecular biological studies.
During the previous 5 years we successfully completed all the research plans on the
biophysical properties of unstructured proteins, the mechanistic studies of protein self
assembly and protein aggregation, and the effects and mechanism of neuronal cell growth,
differentiation and apoptosis mediated by a-synuclein. In this study, we obtained many
interesting results. As a result, we published 11 papers and submitted 3 papers on the
outstanding international journals, such as JBC and Biochemistry, and we published 1 paper
on a Korean journal. We also presented the results 12 times in the international biological
conferences, and 20 times in the Korean biological conferences. We have grown many
excellent researchers, and set up research networks with many outstanding researchers and
performed many collaboratory works with them. Our results accomplished through this
project can be utilized to further our understanding of unstructured proteins in health and
disease.
