복잡한 뇌의 기능을 결정하는 신경회로망은 환경적 요인인 학습 및 기억에 의해서 기능적, 구조적으로 재구성될 수 있는 유연성을 가지고 있다. 본 과제는 이러한 신경 회로망을 구성하는 시냅스의 효율성이 조절되는 양상을 스트레스 자극에 초점을 맞추 어 연구 하였다. 첫째, 군소(Aplysia)가 갖고 있는 단순한 신경계를 모델로 하여 민감화 통각 스트레스에 의해 나타나는 반사기억의 메커니즘인 시냅스 촉진 과정(LTF)에 관여하는 유전자들을 발굴하고, 기능을 분석하여 이들이 시냅스 가소성에 미치는 영향을 규명하였다.

Neural networks involved in the cognitive function of the brain are thought to be morphologically and functionally modified and restructured mainly by learning and memory processes. Our purpose of this research is to study how synaptic efficacy is modulated by stress by using Aplysia and the mammalian nervous system. At first, we examined the effects of a noxious sensitizing stress on synaptic modulation in Aplysia. These studies showed that the expression pattern of many of the genes are changed by stress in the nervous system, and that these genes could be involved in long-term synaptic plasticity. Secondly, in vitro studies using HiB5 cell lines showed that a stress hormone, glucocorticoid induced cell death. In vivo studies using a stress model of pregnant mouse revealed that not only prolonged stress on pregnant mice caused retardation in development and growth of embryos, but also it induced alteration in gene expression in the hippocampus which is a critical structure for spatial memory formation. Furthermore, those mice showed severe impairments in hippocampal long-term potentiation and even in spatial memory. Third, C-terminal fragment of amyloid precursor protein was previously known to be involved in Alzheimer's disease. We found that C-terminal fragment caused a cellular toxicity, which leads to neuronal cell death. Finally, studies on anti-stress function of neurotrophic factors showed that PDGF and BDNF were involved in neuronal growth. Moreover, behavioral studies showed that BDNF was partly involved in neuronal regeneration which can compensate neuronal cell death induced by stress. Taken together, these studies clearly showed that stress could induce gene expression and impaired long-term synaptic plasticity. Furthermore, impairments in synaptic plasticity by stress were strictly related to behavioral impairments. Our results also showed that these stress-induced impairments could be partially restored by anti-stress effects of neurotrophic factors. Our study will not only contribute to the better understanding of the mechanism underlying learning and memory, but also provide the important clues for the prevention of dementia caused by various stresses.
