붉은곰팡이의 유성생식능 관련 유전자군의 기능 분석 및 균주집단간 병리학적 변이 특성 연구를 통해 토착 붉은곰팡이 집단의 생물학적 특성을 규명한다.

붉은곰팡이의 유성생식능 관련 유전자군의 기능 분석 및 균주집단간 병리학적 변이 특성 연구를 통해 토착 붉은곰팡이 집단의 생물학적 특성을 규명한다.
Gibberella. zeae 의 유성생식 특이 유전자 집단을 확보하기 위해 자가화합성 야생형 균주와 두 종류의 자가불화합성 균주 (MAT1-2 유전자 결실 균주와 MAP kinase 유전자 GMK1 결실 균주)를 이용하여 이 들 사이의 cDNA subtraction, cDNA-AFLP, reverse Northern, cDNA microarray, proteomics 분석 등을 수행하였다. Northern blot hybridization을 이용하여 동정된 유전자들의 생장단계별 발현양상을 분석하였으며, targeted gene deletion 방법에 의해 유성생식 특이 유전자의 기능을 분석하였다. 한편, AFLP 와 phylogenetics 분석을 통해 기주식물의 차이에 따른 우리나라 G. zeae 집단간 유전적 다양성을, 혼합접종 및 월동실험을 통하여 토착종의 기주선호도를 분석하였다.
본 연구는 붉은곰팡이 유전체 해독 후 국내외에서 수행된 최초의 대단위 유전자 기능연구이다. 또한 집단유전학적 관점에서 우리나라 토착 붉은곰팡이 균주집단의 유전적 다양성과 기주선호도, 벼와의 공생 가능성 등을 밝힌 국내외 최초 연구이다.
○ 붉은곰팡이 유전체에 대한 DNA chip 과 proteomics 분석을 국내외 최초로 수행하였 다. 이를 통해 총 400 여 종의 유성생식 과정 특이 유전자를 확보하였으며. 이 중 유 성생식과정에서 up-regulation 되는 유전자 300 여 종, down-regulation 되는 유전자 26 종을 확인하였다. 또한 생장단계별로 31 종 유전자의 발현양상도 검정하였다. 본 연구에서는 mating type 유전자에 의해 조절되는 유성생식 관련 유전자 집단에 집중 함으로써 기존 유전체 연구와 달리 분석된 유전자들의 상호 관련성이 매우 높았다.
○ targeted gene deletion 에 의해 총 20 종의 유전자 기능을 검정하였으며, 이 중 13 종 은 유성생식과정에 필수적임이 최종 확인되었다. 이러한 유전자 기능 확인은 선례가 없는 다량의 유전자 기능분석 사례이다.
○ 우리나라의 lineage VI 와 III 토착균주들에서는 이들 총 6 종 유성생식특이 유전자의 발현이 현저히 떨어짐을 확인하였다.
○ 기주식물인 옥수수와 벼로부터 분리한 총 500 균주의 분석결과, 옥수수기원 균주집단 에서 4 개의 lineage (2, 3, 6, & 7) 가 확인되었으며, 그 중 lineage 7 가 우점종 이었다. 하지만 접종원을 공유하고 있는 옥수수포장 인근 논의 벼로부터 분리한 균은 주종이 lineage 6 이었다. 유전적 다양성 분석대상 균주 수 또한 국내외 전례가 없는 대단위이며 이를 통해 연구결과의 과학적 타당성이 입증되었다.
○ 특정 유전자의 염기서열을 이용한 추가적인 phylogenetics 분석결과, 자연상태에서 균주집단 간 유성교배의 가능성을 확인하였다.
○ 이러한 유전적 다양성에도 불구하고 토착종은 벼와 옥수수에 대한 병원성에는 큰 변이를 나타내지 않았다.
○ lineage 6 에 속하는 토착 붉은곰팡이 균주들은 옥수수유래 lineage 7 균주에 비해 벼에서 생존능력이 뛰어났다. 이는 벼가 lineage 6 집단의 유지에 미지의 역할을 하고 있음을 강력히 시사한다. 벼에 대한 lineage VI 균주의 선호도는 국내외에서 최초로 확인된 결과이다. 이를 통해 붉은곰팡이병의 집중적인 방제와 저항성 품종 육성을 위해 우리나라에서는 lineage 6 균주집단에 대한 연구가 필수적임이 확인되었다.
○ 본 과제의 결과는 작물의 생산성 및 국민 보건향상에 크게 기여할 수 있을 것이다.
○ 이러한 연구성과는 이미 4 편의 SCI 저널과 각종 국내외 학술대회 (21 회)에 발표되 었으며, 1-2 편의 SCI 급 논문이 추후 투고 될 예정이다.
○ 국제경쟁력을 갖춘 전문 연구인력을 양성하였을 뿐 아니라 지방대의 연구 경쟁력확보 와 연구특성화 강화 등에 기여하였다.

For development of efficient and economic means to control crop diseases caused by Gibberella zeae, biological characteristics of Korean G. zeae populations have been intensively analyzed in terms of molecular and populational studies of sexual reproduction and host preferences.
To obtain a collection of the genes specifically required for sexual development in G. zeae, we performed several molecular and genomics approaches (cDNA subtraction cDNA AFLP, reverse Northen, cDNA microarray, and 2D-gel analysis) using the self-fertile wild-type H-11 strain and two self-sterile strains generated by deletions of MAT1-2 and MAP kinase gene (GMK1), respectively. In addition, we employed both Northern hybridization and targeted gene deletion analyses for functional study of the selected sexual development specific genes. For populational study of genetic diversity and host preference among Korean isolates of G. zeae, we attempted AFLP and phylogenetic analysis using nucleotide sequences of several genes.
This study is the first report on a high through-put functional analysis of G. zeae since its whole genome has been sequenced. Also, this is the first attempt to explore the genetic diversity, evolution, and host preference of the Korean G. zeae populations.
○ As far as we know, this study is the first report on genomics of G. zeae based on both microarray and proteiomics approaches. We identified more than 400 genes that were differentially expressed during sexual reproduction in G. zeae. Of these, more than 300 genes were confirmed to be up-regulated, and 26 genes were down-regulated. We also determined the time course expression patterns of the selected 31 genes during the entire life cycle of G. zeae.
○ Targeted gene replacement using DJ-PCR products proved that 13 genes out of the selected 20 differentially expressed genes were essential for sexual development in G. zeae, whereas the rest were dispensible. This would be unprecedentedly a large-scale gene-function study compared with the cases in other filamentous fungi.
○ Northern blot analysis revealed that the expression level of the selected 6 genes were much reduced in the Korean lineage VI or III isolates of G. zeae compared with those in the lineage VII isolates from either Korea or North America.
○ We performed phylogenetic analyses using AFLP and nucleotide sequences of Tri101 and MAT1-2 to characterize genetic structure of Korean G. zeae populations. AFLP analysis using more than 500 G. zeae isolates derived from either corn or rice revealed that lineage VII was predominant among the 4 different lineages (II, III, VI, & VII) found within the corn-derived fungal population, whereas lineage VI was predominant among the population derived from rice paddy fields that were closely located to corn field.
○ We identified a unique cluster consisting of 16 isolates in a phylogenetic tree, which was appeared by natural hybridization between different lineages of Korean G. zeae.
○ Virulence assay using the strains representative to each lineage confirmed that there was no significant variation in virulence toward host plants (corn and rice).
○ Fitness test revealed that Korean G. zeae isolates belong to lineage VI appeared to be more competitive against lineage VII isolates on rice, indicating that rice would play an important role as a selection pressure to maintain the population of lineage VI isolates in Korea. This is the first report on possibile host preference among G. zeae populations. Thanks to this result, we can target lineage VI isolates as a main pathogenic population for development of rice or barley resistant to head blight caused by G. zeae in Korea.
○ The results of this study would also contribute to enhancement of cereal productivity and public health as well as to comprehensive understanding of population structure of Korean G. zeae isolates.
○ The results have been already published in 4 international journals listed in SCI, and reported (21 times) in both domestic and international meetings. Also, 1-2 more papers are being prepared.
○ This grant has also contributed to training several students as high-quality researchers and to improving the research activity in a local university in Korea.
