In recent years growing number of cases of prion diseases have been reported, including BSE, vCJD, scrapie, and FFI. To get a better understanding of the fundamental mechanism of these diseases, a prion-like protein, Sup35, from yeast was chosen. The structure-function relationship of Sup35 will be dissected by solid-state and solution NMR techniques, and the inhibitor peptide acting on the oligomerization domain of Sup35 will be developed and fusion protein with this peptide will be constructed. The complete or partial sequence of Sup35 will be produced in E.coli and purified for structural and biochemical studies such as CD, NMR, and fluorescence spectroscopy. A very short sequence of Sup35, responsible for the aggregation, will be chemically synthesized and studied by solid state NMR as a powder form or in the lipid membrane and by solution NMR as a detergent-solubilized form. The inhibitor peptides will be screened and developed by using the genetic selection method. The fusion protein will be developed to use the protein degradation pathway. In addition to the structures of the soluble part of several prion proteins, the structural data of the domain responsible for aggregation will provide priceless information to develop vaccines and drugs for BSE and vCJD. The inhibitor peptide found in this research will play a role as a lead compound to such purposes.
