본 연구에서는 천연물 유래 alkenylbenzene계 화합물의 생리 활성 및 독성 기작을 분자 수준에서 종합적이고 체계적으로 규명하는 것을 목표로 하였다. 이를 위해서 alkenylbenzene 계 화합물에 대해서 약물대사 효소계에 대한 영향 및 면역 조절 기작에 대한 연구를 실시하여, 다양한 생리활성 기능을 가진 alkenylbenzene계 화합물의 효과와 작용기작 및 이들의 독성을 신속하면서도 민감성, 재현성, 경제성을 가진 새로운 생리활성 측정방법의 도출 및 개발과 독성 평가에 필요한 기초자료를 제공하고, 약물대사조절제 및
본 연구에서는 천연물 유래 alkenylbenzene계 화합물의 생리 활성 및 독성 기작을 분자 수준에서 종합적이고 체계적으로 규명하는 것을 목표로 하였다. 이를 위해서 alkenylbenzene 계 화합물에 대해서 약물대사 효소계에 대한 영향 및 면역 조절 기작에 대한 연구를 실시하여, 다양한 생리활성 기능을 가진 alkenylbenzene계 화합물의 효과와 작용기작 및 이들의 독성을 신속하면서도 민감성, 재현성, 경제성을 가진 새로운 생리활성 측정방법의 도출 및 개발과 독성 평가에 필요한 기초자료를 제공하고, 약물대사조절제 및 면역조절제 등의 기능을 가진 신물질의 개발과 안전성 평가를 위한 중요한 자료를 제공하는 것을 목표로 하였다. 약물대사 효소계에 미치는 alkenylbenzene 화합물의 영향을 분석한 결과, in vitro system에 대한 영향은 in vivo의 경우와는 다르게 농도에 의존적으로 P450 효소의 활성을 저해시켰다. Rat의 in vivo system에서는, 거의 모든 alkenylbenzene 유도체가 여러 P450의 활성을 2-10 배 증가시켰다. 활성증가는 단백질 발현의 증가에 이에 선행된 mRNA의 합성증가에 기인함을 확인하였다. 모든 alkenylbenzene 유도체가 모든 phase II 효소들의 활성을 2-4배 증가시켰다. Alkenylbenzene 화합물은 hepa-1c1c7 cell line의 P450 활성을 2-8 배 증가시켰으며, 효소의 발현을 증가시켜 활성을 증가시킴을 알 수 있다. hepa-1c1c7 cell line이 in vivo 상태의 rat과의 반응과 매우 유사하며, 이 결과로부터 hepa-1c1c7이 동물의 alkenylbenzene 화합물에 대한 모델로 매우 적합함을 알았다. In vitro 및 in vivo system에서 보인 차이점은 activemetabolite의 존재가능성을 제시한다. Proteome analysis를 이용하여 hapa-1c1c7 cell line에 alkenylbenzene 화합물을 처리하였을 때 특정 P450 효소들 이외에 신호전달, 세포사멸, 면역 관련 단백질들의 발현이 뚜렷하게 증가함을 알 수 있었다. 또한 apoptosis에 관여하는 bcl-w의 단백질이 P450 및 reductase와 상호작용을 하는 단백질임을 확인하였다. lysoPS, lysoPA, lysoPC가 유의성 있는 PLD 활성증가 효과를 나타내었다. 이러한 결과는 P450 약물대사 기능시스템, 신호전달 네트워크, 세포사멸체계 사이에 밀접한 상관관계가 있음을 나타낸다. Anethole, eugenol, isoeugenol 및 myristicin이 면역 기능을 조절할 것이라는 사실을 알수 있었다. Cytokine 유전자 발현을 기준으로 downstream effect인 면역세포 증식 반응에 대한 영향을 예측할 수 있으며, 이는 T-cell 증식이 억제되는 실험 결과와 일치하였으며, 이러한alkenylbenzene들은 T-cell 특이성을 보였다. 유전자 발현의 upstream mechanism 분석은 antehole, eugenol, isoeugenol 및 myristicin이 cytokine 유전자 발현의 전사 단계에서 작용한다는 사실을 알 수 있었다. 특히 NF-AT 전사조절인자가 주요 작용 대상이 될 것이라는 결론을 내릴 수 있었다. 또한 anethole이 AP-1의 활성을 억제하는 반면, isoeugenol은 NF-kB의 활성을 억제하였다. 이 결과는 같은 alkenylbenzene 골격구조를 갖는 두 물질간의 매우 작은 구조적인 차이가 서로 다른 세포내 활성 경로에 관여한다는 것을 의미하므로, 향후 구조-활성간의 관계 (structure-activity relationship)의 측면에서 유용하게 활용할 수 있을 것이다. Anethole은 NF-AT와 AP-1의 활성을 억제하였는데, 이는 NF-ATc 단백질 수준의 변화와ERK1/2의 활성 변화와 관련이 있었다. NF-AT 및 NF-kB의 활성을 억제하는 결과를 보인 isoeugenol의 경우에는 NF-kB의 구성성분인 p65와 c-Rel 단백질 수준이 감소했다. 특히 p65와 c-Rel 단백질이 핵 내에서 감소했다는 결과로부터 isoeugenol은 NF-kB의 활성화 과정인구성요소의 핵 내로의 이동에 영향을 미친다. 염증반응과 관련된 중요 효소인 iNOS에 대해서는 anethole, safreole, estragol, allylbenzene이 억제 효과를 보였으며 전사조절인자들인 NF-kB와 Oct를 통하여 작용하였다. 또한 면역세포 활용을 통한 평가 이외에도 간세포와의 공동배양을 통한 간기능-면역기능의 연계 연구를 수행하는 기반 기술 또한 구축하였다. 측정하고자 하는 화합물의 대사산물에 의한영향을 평가하기 위하여 면역세포를 간세포와 공동배양하는 방식을 활용하였다. 구축된 실험법을 활용한 결과, 면역세포에 대하여 화합물 단독 처리 시에는 전사조절인자인 Oct 활성이 변하지 않았으나 간세포와의 공동배양을 통하여 화합물이 대사될 수 있도록 한 경우에는 Oct 활성이 억제되었다. 화합물이 생체 내에 투여된 경우에 직접적으로 면역계에 작용하는 것 이외에도, 간 등에서 대사가 이루어 진 이후에도 면역계에 작용할 수 있음을 제시한다. oAlkenylbenzene계 화합물에 의한 약물대사 및 면역기능 제어를 평가 시스템 확립 oalkenylbenzene계 화합물의 생리활성 및 독성기작을 확인하는 biomarker 개발

In the present study, we investigated the physiological action mechanisms of naturally occurring alkenylbenzene compounds at molecular level. For this purpose, the effects of alkenylbenzene compounds on drug metabolizing systems and immune functions were evaluated. In addition to the elucidation of the physiological effects and their action mechanisms of alkenylbenzene compounds, we also have developed a rapid and precise evaluation system for physiological action of certain compounds, which will be used for safety evaluation and development of new compounds regulating drug metabolizing systems and immune functions. The in vivo and in vitro effects of alkenylbenzene compounds on the P450 enzymes has been examined. In the in vitro system all alkenylbenzenes inhibited the P450 activity. The in vivo effect of alkenylbenzenes on the P450s and phase II enzymes in drug metabolism system, has been examined by several systems including animals and cell culture. We found that the alkenylbenzenes increase all P450 activities of the rat liver and liver cell lines, such as Hepa-1c1c7 and HepG2, by measuring the change of marker enzyme activities, protein formation, and mRNA formation. Most of alkenylbenzene compounds increased the activities of P450s and phase II enzymes. Myristicin and safrole showed a remarkable increase of activities of all types of P450s accompanying increased the formation of protein and mRNA. These results suggest that the compounds enhance the metabolism in animals. We also found Hepa-1c1c7 cell line is a valid model to study the effects of alkenylbenzens on the drug metabolism. We also found that P450 systems are related to the signal transduction systems including PLD activity and bcl-w, an apoptosis-related protein. Effects of alkenylbenzenes on immune functions were investigated. We set up the evaluation system for alkenylbenzene-induced lymphoproliferation and developed the method to detect and quantify several cytokines and iNOS gene expression. Anethole, eugenol, and isoeugenol inhibited IL-4 and IL-2 secretion, while myristicin suppressed only IL-2 secretion in PMA/ionomycin-stimulated EL4.IL-2 T-cells. In unstimulated cells, IL-4 secretion was decreased by safrole, allylanisole, and allylbenzene, while eugenol and myristicin increased IL-4 secretion. Nitric oxide production in LPS-stimulated RAW 264.7 macrophage cells was inhibited by safrole, anethole, allylansole, allylbenzene, and isoeugenol. Safrole decreased iNOS mRNA expression and the binding activity as well as transactivation of NF-kB and AP-1 factors. Anethole, eugenol, isoeugenol, and myristicin inhibited the concanavalin A-induced T-cell proliferation in mouse splenocytes. PMA plus ionomycin (Io) induced IL-2 mRNA expression and protein secretion in EL4.IL-2 T-cells, which were inhibited by anethole, eugenol, isoeugenol, and myristicin as determined by quantitative/competitive RT-PCR and ELISA, respectively. Anethole decreased the NF-AT and AP-1 binding activity, but isoeugenol decreased the NF-AT and NF-kB binding activity. These results suggest that the isoeugenol-induced T-cell dysfunction is due to the inhibition of IL-2 expression and is mediated, at least in part, through an inhibition of NF-AT and NF-kB. Several alkenylbenzenes have immunosuppressive activity, and the presence and the position of hydroxyl- and methoxy-group on allylbenzene backbone as well as the position of double bond at backbone propyl group are directly related with the regulatory activity of alkenylbenzenes.
Several alkenylbenzens increased the metabolic activities including P450s and phase II enzymes. They have also immunosuppressive activities. An integrative approach was performed to elucidate the connection of drug metabolism, immune system, and signal transduction. This study set up the research system for a rapid and precise functional evaluation of certain compounds, which can be used for the development and safety evaluation of new compounds regulating physiological functions. This study would provide the basic data for development of biomarker for physiological functions and toxicities of alkenylbenzenes. The results can promote the understanding of structure-activity relationship of alkenylbenzenes as well as correlation of drug metabolism and immune response. This also will provide the basic concept for the connection of drug metabolism and immune system to the control mechanism by signal transduction. This study would provide a system to evaluate the effects of certain chemical on the drug metabolism, immune response, and signal transduction at the same time.
