전기방사법을 이용하여 실크 피브로인 나노섬유로 구성된 차폐막을 제조함으로써, 기존의 동결건조에 의해 제조되는 제품에서 발생하였던 형태 변형의 물성 문제를 크게 개선하였고, 마이크로구조를 가지고 있는 종래 차폐막의 한계를 벗어나 나노구조로 전개시켜, 높은 기공도와 비표면적을 갖는 나노차폐막을 제조함으로써 생체재료로서의 활용도를 높임. 비분해성 차폐막의 사용으로 인한 2차 제거 수술의 부담을 없애면서, 산분해 산물과 부작용이 없는 생분해성 차폐막을 개발하였다는데 의미가 있음. 생체적합성과 세포배양시험 결과에서 실크 피브로인 나노 차페막

A major goal of periodontal regeneration therapy is the regeneration of lost supporting tissue, including the alveolar bone, periodontal ligament, and cementum around a previously diseased tooth root. Guided tissue regeneration (GTR) has accomplished this goal, and has become a standard procedure for periodontal regeneration therapy since it was initially suggested for such therapy. In addition, it has been applied to bone and peri-implant defects, and for bone augmentation procedures prior to implant placement. In such situations, it is sometimes termed guided bone regeneration (GBR). For the membrane to be effective, it must satisfy criteria such as biocompatibility, tissue integration, cell occlusion, mechanical stability, and clinical manageability. The expanded polytetrafluoroethylene (e-PTFE) membrane, which is nonresorbable, fulfilled these needs. However, it has been limited by the necessity of a second surgery for membrane removal and by its high membrane exposure rate, which potentially result in patient discomfort, increased cost, post-surgical infection, and possibly suboptimal bone regeneration. In order to overcome these problems, a variety of resorbable membranes were developed and used. Currently used resorbable membranes are made of collagen, polyglycolic acid, polylactic acid, or copolymers of lactic and glycolic acid. However, several inherent disadvantages have been observed with these membranes, including weak structural integrity, variable degradation rates, and host immune reactivity. Silk fibroin (SF) is known as a potential candidate material for biomedical applications because it has several attractive properties, including good biocompatibility, good oxygen and water vapor permeability, and biodegradability. In addition, silk fibroin provides a remarkable combination of strength and toughness to provide enough stability. Therefore, it may have the additional advantages of space maintenance for bone ingrowth while preventing membrane collapse. Herein, the authors prepared nanofiber GBR membrane using silk fibroin. This membrane was anticipated to have a potential in application for guided bone and periodontal regenerative therapy. we prepared SF nanofiber membrane and evaluated the biocompatibilities. In this study, the preparation of SF nanofibers, cell attachment, the differentiation of cells grown on non-woven matrix made of SF nanofiber, the histologic evaluation of biocompatibility of implanted membrane, and GBR efficacy will be detailed discussed. The electrospun SF was shown to possess good biocompatibility and to enhance bone regeneration, and no evidence of any inflammatory reaction was confirmed in the in vivo study. Taken together, these results strongly suggest that the SF nanofiber membranes should be useful as a tool for GBR.
