전자전달반응에 관여하는 Glucose oxidase (GOx), Horse Radish Peroxidase (HRP), Laccse (Lc), Bilirubin Oxidase (BOD), Carbon Monoxide Dehydrogenase (CODH) 등의 산화환원효소(oxidoreductase)를 다양한 나노입자 및 나노표면에 연결시킨 바이오나노구조를 형성하여 전기화학촉매반응에 적용하는 반응성 연구에 대한 기초를 마련하고자 한다. 금 전극 및 유리탄소전극 표면에 전기화학적 펄스를 이용한 전기도금 방법으로 수~수십 나노미터 크기의 금 나노입자를 도입한 후 bis(N-hydroxysuccinimidyl) 3,3'-dithiopropionate를 linker로 사용하여 GOx와 BOD를 연결하여 전기화학적 특성 및 촉매반응성을 관찰하였다. 당센서 및 산소환원의 촉매로 좋은 효과를 보여줌 을 확인할 수 있었다. 또한, AAO를 template로 이용하여 약 100 nm 크기의 nanopillar를 표면에 형성시킬 수 있었는데, 이 전극의 roughness factor를 6~80 정도로 조절할 수 있었고, 이에 따라 dopamine과 ascorbic acid를 선택적으로 측정할수 있음을 확인하였다. Polystyrene bead를 template로 이용하여 Ni, Pt,polypyrrole 등의 nanoporous 구조를 전극표면에 구성할 수 있었고, 다시polypyrrole의 nanoporous 구조를 template로 이용하여 Ni, ZnO 등의 나노점을 형성시킬 수 있었다. 또한, zinc phosphate를 전기화학적 산화에 의해 표면에 10~100nm 정도의 나노점을 만들 수 있었고, 이 구조를 이용하여 nanoporous 금 표면을 구성할 수 있었다. Zinc phosphate (hopeite)를 적당한 용액 조건에서 표면에 구성하면 Zn2+는 conducting하며 산소는 blocking하는 표면을 구성할 수 있어 생체 용액에서 작동하는 아연전지도 구성 가능하였다. 또한, 유리탄소전극 표면을 nitrobenzene diazonium 화합물은 전기화학적 환원에 의해 적절하게 nitrophenyl group을 표면에 도입하면 ferrocynide의 amplification에 의해 ferrocenemethanol을 nM 농도까지 CV로 측정할 수 있는 표면을 구성하였다. 이러한 연구는 나노구조의 형성과 이에 따른 전극 효소 반응 및 다양한 측정에 이용할 수 있는 기반을 마련하 였다고 여겨진다. 본 연구를 통하여 금 나노점을 전극표면에 도입하여 이에 GOx, BOD 등의 산화환원효소(oxidoreductase)를 연결하여 바이오나노 복합체를 형성하여 이를 이용한 촉매반응을 비교한 것은 관련 분야에서 시도된 첫 연구로서 전기화학 바이오나노 관련 연구의 기초를 마련하였다. 또한, 다양한 template의 이용 및 이를 이용한 nanoporous 전극표면의 구성을 일반화시킬 수 있어서 관련된 연구에 지대한 영향을 미칠 것으로 기대한다. 이러한 연구를 진행하는 과정에 생체 내에서 작동하는 우수한 아연 전지를 발견한 것도 큰 성과에 속한다. 그리고 diazonium 화합물을 적절하게 표면에 고정하면 전자전달 반응 및 촉매반응성을 조절할 수 있다는 것도 중요한 연구 성과라 할 수 있다. 이와 같은 연구를 전기화학/효소/나노 관련 영역에 다양한 경험을 갖는 연구 인력을 교육할 수 있게 되어 경계영역의 학문분야로 다양화되어지는 과학발전에 필요한 연구 인력을 양성할 수 있게 된 것이 가장 큰 성과라 여겨진다.

Oxidoreductases such as Glucose oxidase (GOx), Horse Radish Peroxidase (HRP), Laccse (Lc), Bilirubin Oxidase (BOD), Carbon Monoxide Dehydrogenase (CODH) will be linked to gold or cabon nanoparticles and nanoporous surfaces to invesigate the control of the catalytic and electrocatalytic activity of the enzymes. GOx, HRP, laccase, BOD, CODH are the enzymes which our group has been actively worked on. The enzymes was linked to gold or carbon nanoparticles and the reactivties were compared. The gold nanoparticles were synthesized in 100∼ 101 orders of nanometer size and connected to the enzyme using bis(N-hydroxysuccinimidyl) 3,3'-dithiopropionate as a linker. The terminal lysine part of the enzyme was used for the connection. The relative size of the enzyme and nanoparticle is one parameter to control the structure and the reactivity of the bionanomaterial. The conventional electrochemical methods was used for the characterization and the SECM technique to trap the nanomaterial between tip and substrate was useful to investigate the single or a few numbers of nanomaterials. The immobilization of the nanomaterials was tried and the electrochemical properties and the catalytic activities was compared. Carbon-based nanomaterials was used, too. In this case, the electroreduction of the functionalized diazonium salt was used to link the nanoparticle and the enzyme. The template synthesis method was applied to produce nanosurfaces to immobilize enzymes. The structure and the reactivity of the enzyme on the nanosurface compared to the normal surface was compared, too. The electroreduction of osmium redox polymer can immobilize the polymer and enzymes onto the nano-structured carbon surfaces and the catalytic activities and the biosensor applications will be studied. The integrated effort of the use 5 kinds of oxidoreductases for the linkage to gold or carbon nanoparticles and nanosurfaces is the first trial to generalize the preparation of the bionanomaterials based on oxidoreductases. The fundamental concept and technique will be developed by this investigation. The enhancement of the mass transport on the nanostructures will enhance the electrocaltalytic reaciton of the enzyme to develop better biosensor. The interdisciplinary-trained researchers in the area of electrochemistry/enzymology/nanoscience will be trained through this research program.
