Ⅰ. 제 목 식물유전체연구소재은행 Ⅱ. 연구개발의 목적 및 필요성 (1) 연구개발의 목적 - 국내 식물유전체연구를 위한 각종 소재를 mutant library, 전장 cDNA library, 형질전환용 세포 주 등을 중심으로 개발, 수집하고, 이에 대한 DB를 구축하며, 소재와 정보를 국내 연구자들에 게 공급할 수 있는 국가적 수준의 인프라를 구축함. Ⅰ. 제 목 식물유전체연구소재은행 Ⅱ. 연구개발의 목적 및 필요성 (1) 연구개발의 목적 - 국내 식물유전체연구를 위한 각종 소재를 mutant library, 전장 cDNA library, 형질전환용 세포 주 등을 중심으로 개발, 수집하고, 이에 대한 DB를 구축하며, 소재와 정보를 국내 연구자들에 게 공급할 수 있는 국가적 수준의 인프라를 구축함. (2) 연구개발의 필요성 - 식물 functional genomics는 현재 산발적으로 이루어지는 식물 유용유전자의 일반적인 클로닝 방식을 획기적으로 개선하여 유용유전자를 대량으로 확보하는 기술로서 식물의 유용유전자는 종묘산업의 영역을 극대화할 것임. 아울러 식물 functional genomics는 농화학(제초제, 작물보호 제), 제약(신약), 식품(nutraceuticals, 기능성 식품), 환경(phytoremediation) 등의 연구분야에서 인프라의 역할을 하게 되므로 21세기 생물산업의 발전을 위한 가장 중요한 기반기술의 하나임. - 식물유전체연구를 위한 각종소재의 개발과 관련정보의 DB화, 연구자들의 소재에 대한 needs 의 신속한 파악과 소재 분양의 체계화는 매우 중요한 인프라로서 초기단계에 이를 국가적 인프 라로 구축하여 대외 경쟁력을 강화하고 식물 생명공학 연구개발 분야의 국가적 지원의 효율을 증강시킬 수 있음. - 특히 식물 유전체 연구 분야에서는 기술선진국에서도 모델식물인 애기장대(Arabidopsis)를 제외 - 4 - 하면 관련 mutant library, 전장 (full-length) cDNA library, 형질전환을 위한 식물세포주 등에 대한 소재분양을 위한 인프라 구축이 초보단계에 있으므로 국내에서 선투자하여 대외경쟁력을 조기 확보할 필요가 있음. -따라서 유용유전자 선점을 위한 식물분야의functional genomics에 대한 투자가 증대될 것이며 본 과제는 이런 예측 가능한 상황에서 국내 관련 인프라를 강화함으로써 관련 분야의 대외경쟁 력을 획기적으로 증대시킬 수 있을 것으로 사료됨.

Objectives and Importance of the Research - Collection and development of plant genetic resources (mutant line, full-length clones, cDNA library, and plant cell line) for functional genomics study - Construction of national infra for plant genetic resources III. Scopes and Contents of the Research - Development of full-length cDNA library and Construction of DB - Development of mutant line for metabolomics reference - Development of plant cell lines for genetic transformation - Establishment of the platform for analysis of biochemical properties from plant mutant lines using metabolic profiling IV. Results of the Research In this project, major results on development of plant genomics services can be summarized as follows. 1. Development of full-length cDNA library and Construction of DB - 10 - - Full-length clones are necessary for functional genomics studies. In this project, over than 8,000 full-length clones were isolated from pepper, periwinkle, soybean, sweet potato, and mistletoe. Also DB about these clones were constructed. Development of mutant line for metabolomics reference - To establish the platform for metabolic profiling analysis from plant tissue, 200 metabolic mutant lines of Arabidopsis and Catharanthus roseus were developed using sense and antisense technologies. 3. Development of plant cell lines for genetic transformation - Culture conditions for plant regeneration from 10 plant crops including lily, carnation, rose, Chrysanthemum, japanese honeysuckle, Catharanthus roseus, rice, Acanthopanx, and coriander were established. 4. Establishment of the platform for analysis of biochemical properties from plant mutant lines using metabolic profiling - To establish the platform for for analysis of biochemical properties from plant mutant lines using metabolic profiling, whole cell extracts from 201 activation-tagged hairy root mutants of Catharanthus roseus were subjected to FTIR and 1H NMR for spectral fingerprinting. FTIR and 1H-NMR spectral data were analysed by pattern recognition using multivariate statistical analysis . At present , we are developing the technology for categorizing gene function using PCA from spectral data. 5. Large-scale production of GABA from cell suspension of Coriandrum sativum L. - GABA(?-aminobutyric acid) is a non-protein amino acid that is widely distributed - 11 - throughout the biological world. In animals, GABA functions as the inhibitory neurotransmitter in the central nervous system by acting through the GABA receptors. Its synthesis results primarily from an -decarboxylation catalyzed by glutamate decarboxylase (EC 4.1.1.15). - In this study, relative quantification of GABA from Coriandrum sativum L. was established by 1H-NMR spectral analysis. Also, culture condition for GABA productivity from Coriandrum sativum L. was examined.
