무척추동물들의 체내에 존재하거나 분비되는 생체방어인자를 확인, 정제하여 구조적 혹은 기능적 특성을 분석한다. 본 과제에서는 먼저 각 동물군들에 존재하는 면역 인자들을 확인 및 분리하여 이들의 활성 및 구조적 특성과 면역학적 특성을 밝혀내는 데에 주된 목적을 두었다. 특히, 무척추동물의 체액성 면역인자의 존재를 확인하여 분리하는 일은 본 과제를 시작하는 데에 있어 가장 먼저 수행되어야 할 것으로 판단하였던 바, 우선 protease, recognition factor(fungus binding proteins: FBP), lysozyme과 항균 peptides 등에 초점을 맞추었다. 이를 위해 본 연구에서는 각 단백질들을 연구하기 위한 대상 무척추동물들을 설정하였는데, protease는 환형동물(갯지렁이: Neanthes diversicolor), FBP는 극피동물(보라성게: Anthocidaris crassipina), lysozyme은 절지동물, 항균 peptides는 원시척색동물(비단멍게: Halocynthia aurantium)과 연체동물(개조개: Saxidomus purpuratus)에서 각각 연구되었다. 본 과제의 연구에서는 위 다섯 가지 체액성 면역인자들 중 항균 peptides에 대해서 많은 가시적인 성과를 얻어냈다. 그러므로 본 보고서에서는 이 항균 peptide에 대한 내용을 기술하기로 한다. 항균 peptides는 이제까지 다양한 동물체들에서 발견되어 온 매우 잘 알려진 동물의 생체방어인자이다. 현재, 항균 peptides는 사람을 포함한 포유류에서부터 하등한 무척추동물들에 이르기까지 다양한 동물들에서 발견되어 왔으나 실제 무척추동물들로부터 분리된 항균 peptides의 숫자는 상대적으로 부족한 실정이다. 따라서 본 과제를 통해 여러 무척추동물들의 체내에서 새로운 항균 peptides가 분리된다면 동물면역인자들의 진화적 기원을 추적하거나 신규 peptide antibiotics를 고안하는 데에 있어 중요한 자료가 될 것이다. 이제까지 항균 peptides는 많은 동물군들에서 발견, 분리되어왔다. 그러나 전술한 바와 같이 무척추동물들로부터의 항균 peptides의 탐색은 아직 더 많은 연구가 있어야할 분야라 하겠다. 본 과제에서는 먼저 국내에 서식하고 있는 Halocynthia aurantium의 체액세포에 존재하는 항균 peptides를 확인하였으며, H. aurantium 로부터 새로운 구조의 항균 peptides를 정제하여 물리화학적 특성 및 항균활성의 특성을 규명했고 cDNA cloning 및 항진균 활성과 작용 mechanism을 밝혔다. 과다한 항생제의 남용으로 기존 항생제들의 작용기전을 피할 수 있는 돌연변이 병원 체들이 급속한 속도로 출현하고 있는 실정이지만 인류는 지난 30 여년 동안 기존 항생제들과 전혀 다른 작용기전을 지닌 진정한 의미의 새로운 항생제를 개발하질 못했다. 이런 때에 내성균에 강력한 항균활성을 지닌 halocidin은 새로운 항생물질 후보물질중 하나라 사료되어진다.

In this subject, we focus on activity, structural properties and immunological characteristics of immune proteins which exist in invertbrates. Particularly, because it was thought that identification and purification of humoral immune proteins from invertbrates should be achieved for the first time, we focused on purifying protease, recognition factor (fungus binding proteins : FBP), lysozyme and antibacterial peptides etc. Protease, FBP, lysozyme and antimicrobial peptides were studied from annelid (Neanthes diversicolor), echinoderm (Anthocidaris crassipina), an arthropod and chordate (Halocynthia aurantium) respectively. In this study, there were definite results of antimicrobial peptide among the above immune proteins. Therefore in this paper, we descrided study of antimicrobial peptide. Antibacterial peptides is well-known immune factor which is discovered in various vertbrates and invertbrates. Present, antibacterial peptides have discovered in various animals until invertebrates but antibacterial peptides' number purified from invertebrates is lacking relatively. In this subject, a novel antibacterial peptide was identified and purified from hemocytes of Halocynthia aurantium that is inhabiting in domestic, and studied for cDNA cloning, antibacterial test and fungicidal action mechanism of halocidin. In recent years overuse of antibiotics has resulted in the rapid evolution of multiple drug-resistant bacteria, which have caused a variety of incurable infectious diseases. In this time, halocidin that have strong antibacterial activity to resistant bacteria is considered one of new antibiotic candidate substance.
