Children have different susceptibilities during different life stages owing to their dynamic growth and developmental processes as well as physiological, metabolic, and behavioural differences. Exposure pathways and exposure patterns may also be different in different stages of childhood. As children may be at increased risk at different developmental stages, with respect to both biological susceptibility and exposure, new risk assessment approaches may be necessary in order to adequately protect children. Children-protective risk assessment approaches must be based on a better understanding of the interactions of biologic susceptibility and exposure factors at each stage of development. In order to gain a better understanding, we describe physiological, metabolic, functional, toxicokinetics, and toxicodynamic characteristics of children across life stage that contribute to unique difference in their susceptibility to hazardous substances. The impacts of exposure to hazardous substances on the organ systems and differences in exposure pathways and behavioural characteristics are addressed as a function of life stage. We review the major scientific principles and method to be considered in assessing health risks from exposure to hazardous substances in children during critical stage of development. Protection of children is at the core of the sustainability of the human species. This guidelines of health risk assessment on exposure of children to hazardous substances will help researchers, health officials, and regulatory scientist on the risk assessment field to understand of children's biologic susceptibility and exposure characteristics, and to carry out children risk assessment scientifically and systematically.
