Peer review 평가위원은 노출, 생약재 분석, 유전독성, 실험동물 등 전문위원을 선정하였으며, 자문회의와 자체 그룹 회의 등은 시험에 대한 기초자료와 보고서 설명 및 평가 등에 대하여 토론하였다. 생약재 3종에 대하여 감수는 유전독성시험의 세균을 이용한 복귀돌연변이시험에서 대사활성계 적용 및 비적용시 모두 양성으로 판정하였으나 peer review 결과 음성대조군의 재현성 있는 결과와 양성대조군의 확실한 양성발현을 일부 균주에서 확인하기 어려워 실험결과에 대하여 양성 또는 음성으로 판정하기 어려웠다. 정확한 판단을 위해서는 재시험을 실시하는 것을 고려하여야 할 것으로 사료된다. 또한 고삼의 경우, 염색체이상 시험에서의 농도설정에 있어 실제 OECD 시험지침에서는 세포독성이 없다고 판정하고자 할 경우, 거의 독성이 없거나 전혀 없는 농도가 포함되어야 한다고 제시되어 있으며 단순히 50% 이상인 생존율을 두고서 세포독성이 없다고 판정하는 것은 그 신뢰성이 낮을 것으로 사료된다. 따라서 농도설정 시 최고농도가 5000 ${\mu}g$/$m{\ell}$를 포함하여 공비를 2가 아니라 더 크게 두어 625 ${\mu}g$/$m{\ell}$ 보다 낮은 농도에서도 실험결과를 얻을 수 있도록 농도를 설정하는 것이 바람직하였을 것으로 사료된다. 원화 추출물의 중금속 분석 자료를 검토한 결과 비소(3.9 ppm)와 망간(118.9 ppm)의 농도가 식약청 고시의 허용기준(비소 3 ppm, 총중금속 30 ppm) 이상으로 나타났으나 이것은 단회 측정 및 분석결과이고 또한 생약재의 경우 생산지의 토양 등 환경에 따라 구성 중금속 성분 및 함유량이 매우 다르게 나타나므로 이에 대한 평가방법의 정립이 필요하다고 사료된다. 그러므로 본 연구에서는 이에 대하여 기술보고서에는 자세히 언급하지 않았다. 생약재에 대한 peer review 결과는 우리나라 독성물질 국가관리사업(KNTP)에서 평가된 바 있는 절차와 전문가 실무그룹평가에 의하여 기술보고서(technical report)로 작성되었다. 이 연구에서 작성된 독성시험 기술보고서는 향후 식품의약안전청과 국립독성과학원이 우리나라 독성연구의 중심점으로서 국가적인 생약재의 안전성 자료를 구축하고 미국 NTP와의 MOU의 일환으로 자료의 공유뿐 아니라 국내 평가기반 구축에도 도움이 될 수 있을 것으로 판단된다.

This study was performed peer review for the toxicity test results which already have been done to evaluate safety of herbal medicine(Sophorae flavescentis Radix, Euphorbiae kansui Radix, Genkwa Flos) in Korea, at the same time, under based on US National Toxicology Program(NTP) and EU toxicological assessment method of chemicals, was completed manual for peer review of KNTP toxicological study and made out KNTP technical reports for three kinds of herbal medicines. Committee for peer review was made up of experts in different fields(exposure evaluation, herbal medicine analysis, genetic toxicity, animal toxicity) and conducted to characterize and evaluate safety and data reliability about potentially toxic and hazardous herbal medicines through existing test results At the technical reports of herbal medicines, the chromosomal aberration test for Sophorae flavescentis Radix was not metioned clearly on supporting rationale for determining cytotoxicity with 50% survival rate. According to OECD guidelines (No. 473), where cytotoxicity occurs, concentrations of tested material should include concentration with little or no toxicity, and this study did not seem to be consistent with guidelines. Therefore, these results raise the possibility that chromosomal aberration test for Sophorae flavescentis Radix was induced in the harsh condition inducing potential cytotoxicity. It is desirable to confirm the cytotoxicity in tested condition again or to extend extract to lower concentration exhibiting no toxicity. The bacterial reverse mutation assay in Euphorbiae kansui Radix was identified as definitive criteria to determine "an increase"in colony number was not indicated, although some of results were concluded to be positive and the number of revertant colony was within historically normal range. Hence, it is difficult to conclude that Euphorbiae kansui Radix extract has the possibility to augment reverse mutation. It might be advisable to make adequate criteria and to repeat test to ensure reproducibility. In case of Genkwa Flos, it showed high values in heavy metal content, especially Mn was 118.9 ppm. However, the results were hard to be confirmed because the heavy metal were affected a lot from soil condition and surrounding. So it should be considered to establish the evaluation method of heavy metal. These peer reviewed technical reports would help KFDA and NITR as the central organization of toxicology study in Korea build up international safety data of herbal medicine, share smoothly information with US NTP, and form the foundation of domestic evaluation method for herbal medicine in the future.
